Description
本發明是有關於一種醣類衍生物於生醫檢測領域之應用,且特別是有關於一種螢光醣類衍生物之用途。
近年來,由於健康照護及醫療上的需求,生醫感測技術的發展逐漸受到重視。在許多生醫造影技術(biomedical imaging technique)中,常需要利用非侵入式的光學感測方法來檢測及追蹤細胞或分子,以利於疾病之診斷。
醣類分子為細胞能量的主要來源之一,且其於癌症及微生物感染等疾病進程(disease progression)中扮演相當重要的角色。此外,亦有許多與醣類相關的細胞標記(cell marker)已被用來檢測各種疾病。在先前研究中已發現,大部分癌細胞會比正常細胞攝取更多的醣類分子。因此,對細胞攝取醣類的情況進行觀察有助於檢測癌細胞的增生(proliferation)、分化(differentiation)、轉移(metastasis)和血管增生現象(angiogenesis)。
目前用於檢測癌症及腦部、心臟部位疾病等的主要診斷工具包括正子斷層掃描術(positron emission tomography,PET)。於正子斷層掃描術中,例如可藉由使用帶有正子同位素(positron isotope)標記的醣類分子(如氟-18去氧葡萄糖(fluorine-18-fluorodeoxyglucose;FDG))作為放射性造影劑(radiocontrast agent),來對組織進行
非侵入性的檢測。
然而,由於放射性造影劑中的正子同位素必須經由迴旋加速器(cyclotron)製造,且因其半衰期短(如氟-18的半衰期約為兩個小時),因此正子斷層掃描設備必須設置在迴旋加速器附近,以便於快速取得造影劑。另外,正子斷層掃描中會釋放出相當大的輻射量,其輻射劑量(radiation dose)甚至可達一般X光攝影的數十倍至數百倍,因此對於細胞仍會造成傷害,尤其對於年輕女性的生殖器官之傷害更為驚人。並且,由於正子斷層掃描設備及迴旋加速器設施的造價均十分昂貴,這也連帶造成檢測費用高昂。
此外,需要開發具有簡便、低成本且生物相容性良好而較無毒性等特性的特異性螢光檢測系統,以應用於臨床上之疾病檢測及藥物篩選等領域。
本發明提供一種螢光醣類衍生物於細胞檢測之用途,可降低檢測成本、無須搭配昂貴儀器或設備,且亦無暴露於高能量輻射線的風險。
本發明提出一種螢光醣類衍生物之用途,用於細胞之檢測,上述螢光醣類衍生物為由反應材料經亞胺生成反應(imine formation reaction)所得,且上述反應材料為選自以下的組成物(A)或組成物(B):組成物(A)包括還原醣類化合物以及含胺基化合物
(amino group-containing compound),且上述含胺基化合物具有至少一個一級胺基(primary amino group);組成物(B)包括胺醣類化合物(amino sugar compound)以及含羰基化合物(carbonyl group-containing compound),且上述胺醣類化合物具有至少一個一級胺基。
在本發明之一實施例中,上述之還原醣類化合物為選自由葡萄糖(glucose)、麥芽糖(maltose)、果糖(fructose)、乳糖(lactose)、半乳糖(galactose)、甘露糖(mannose)、纖維雙糖(cellobiose)、木糖(xylose)、阿拉伯糖(arabinose)、核糖(ribose)、去氧核糖(deoxy ribose)、糊精(dextrin)、D-葡萄糖胺(D-glucosamine)以及乙醯葡萄糖胺(N-acetyl-glucosamine)所組成的群組。
在本發明之一實施例中,上述之含胺基化合物為選自胺基酸(amino acid)或烷基胺(alkyl amine)。
在本發明之一實施例中,上述之含胺基化合物為選自由丁胺(butylamine)、辛胺(octylamine)、1-十二烷基胺(1-dodecylamine)、1-十六烷基胺(1-hexadecylamine)、1,6-己二胺(1,6-hexadiamine)、甘胺酸(Glycine)、丙胺酸(Alanine)、纈胺酸(Valine)、白胺酸(Leucine)、異白胺酸(Isoleucine)、苯丙胺酸(Phenylalanine)、色胺酸(Tryptophan)、酪胺酸(Tyrosine)、天門冬胺酸(Aspartic acid)、組胺酸(Histidine)、天冬醯胺(Asparagine)、谷胺酸(Glutamic acid)、賴胺酸(Lysine)、谷氨醯胺(Glutamine)、甲硫胺酸(Methionine)、精胺酸(Arginine)、

在本發明之一實施例中,上述之胺醣類化合物為選自由D-葡萄糖胺(D-glucosamine)、半乳糖胺(galactosamine)以及幾丁胺糖(chitosan)所組成的群組。
在本發明之一實施例中,上述之含羰基化合物為選自由丙酮、異戊醛、D-葡萄糖胺(D-glucosamine)以及乙醯葡萄糖胺(N-acetyl-glucosamine)所組成的群組。

基於上述,藉由本發明所提供的螢光醣類衍生物於細胞檢測之用途,可實現快速簡便、低成本、生物相容性良好且無毒性的螢光檢測機制,而非常適用於疾病檢測、微生物檢測、環境毒物簡易檢測及藥物篩選等。
為讓本發明之上述特徵和優點能更明顯易懂,下文特舉實施例,並配合所附圖式作詳細說明如下。
本發明提供一種螢光醣類衍生物之用途,上述螢光醣類衍生物為由一反應材料經亞胺生成反應所得,且上述反應材料為選自組成物(A)或組成物(B)。上述組成物(A)包括還原醣類化合物以及含胺基化合物,且上述含胺基化合物具有至少一個一級胺基;上述組成物(B)包括胺醣類化合物以及含羰基化合物,且上述胺醣類化合物具有至少一個一級胺基。
其中,在經上述亞胺生成反應後,於反應材料中可生成至少一亞胺基(C=N)。具體而言,上述亞胺基例如是醛亞胺基(aldimino group)及/或酮亞胺基(ketimino group)。
以下,將對本發明中所使用的螢光醣類衍生物之組成進行說明,而後再以各種實例詳述本發明之原理與其應用。應注意,於本說明書中,若未特別說明一基團(化合物)為經取代或未經取代,則應解釋為此基團(化合物)包含無取代基之基團(化合物)以及具有取代基之基團(化合物)兩種情況。舉例而言,用語「胺基」包含未經取代之胺基以及經取代之胺基。
組成物(A)包括還原醣類化合物以及具有至少一個一級胺基的含胺基化合物。
還原醣類化合物為含有醛基或酮基,且具有還原能力
者,其包含單醣、雙醣、寡醣及多醣。具體而言,還原醣類化合物例如是選自由葡萄糖、麥芽糖、果糖、乳糖、半乳糖、甘露糖、纖維雙糖、木糖、阿拉伯糖、核糖、去氧核糖、糊精(dextrin)、D-葡萄糖胺(D-glucosamine)以及乙醯葡萄糖胺(N-acetyl-glucosamine)所組成的群組,但並不限於此些化合物。
含胺基化合物為具有至少一個一級胺基的化合物,其可選自胺基酸或烷基胺。具體而言,含胺基化合物例如是選自由丁胺、辛胺、1-十二烷基胺、1-十六烷基胺、1,6-己二胺、甘胺酸、丙胺酸、纈胺酸、白胺酸、異白胺酸、苯丙胺酸、色胺酸、酪胺酸、天門冬胺酸、組胺酸、天冬醯胺、谷胺酸、賴胺酸、谷氨醯胺、甲硫胺酸、精胺酸、絲胺酸、蘇胺酸、半胱胺酸以及脯胺酸所組成的群組。然而,實際上並不限於此些化合物。
於組成物(A)中,上述還原醣類化合物以及含胺基化合物之含量並無特別限定,只要能夠依照需求產生上述亞胺基即可。還原醣類化合物與含胺基化合物的莫耳比例如是設定在1:20~20:1的範圍內,但不限於此。
組成物(A)亦可選擇性地包括適當的溶劑,使還原醣類化合物以及含胺基化合物能夠溶解於溶劑中,以進行亞胺生成反應。溶劑之種類可由所屬技術領域中具有通常知識者依照實際需求以及化合物種類加以選擇,而並無特別限定。此外,亦可依照需求調整亞胺生成反應的環境條件(如溫度、壓力等參數),以利於形成本發明中所應用
之螢光醣類衍生物。
具體而言,考慮到反應速率以及反應產物之穩定性,例如可將進行亞胺生成反應時的pH值控制在微鹼環境下,並將溫度控制在25℃~組成物(A)的迴流溫度,藉此可容易地獲得所需的螢光醣類衍生物。
胺醣類化合物例如是選自由D-葡萄糖胺、半乳糖胺以及幾丁胺糖所組成的群組,但並不限於此。含羰基化合物例如是選自由丙酮、異戊醛、D-葡萄糖胺以及乙醯葡萄糖胺所組成的群組,但並不限於此。
同樣地,於組成物(B)中,上述胺醣類化合物以及含羰基化合物之含量並無特別限定,只要能夠依照需求產生上述亞胺基即可。胺醣類化合物與含羰基化合物的莫耳比例如是設定在1:20~20:1的範圍內,但不限於此。
組成物(B)亦可選擇性地包括適當的溶劑,使胺醣類化合物以及含羰基化合物能夠溶解於溶劑中,以進行亞胺生成反應。溶劑之種類可由所屬技術領域中具有通常知識者依照實際需求以及化合物種類加以選擇,而並無特別限定。此外,亦可依照需求調整亞胺生成反應的環境條件(如溫度、壓力等參數),以利於形成本發明中所應用之螢光醣類衍生物。
具體而言,考慮到反應速率以及反應產物之穩定性,例如可將進行亞胺生成反應時的pH值控制在微鹼環境
下,並將溫度控制在25℃~組成物(B)的迴流溫度,藉此可容易地獲得所需的螢光醣類衍生物。
值得注意的是,於組成物(B)中的胺醣類化合物與含羰基化合物亦可為相同的化合物。意即,可使用一含羰基的胺醣類化合物,並藉由使其進行自身亞胺生成反應而生成所需的螢光醣類衍生物。此種含羰基的胺醣類化合物例如是D-葡萄糖胺,但並不限於此。此外,藉由使用此種含羰基的胺醣類化合物,可進一步降低材料成本,亦有利於實現簡便而快速的細胞檢測方法。
另外,由於上述本發明中所使用的螢光醣類衍生物為應用於細胞檢測,故上述還原醣類化合物、含胺基化合物、胺醣類化合物以及含羰基化合物較佳為不具有生物毒性、且生物相容性良好者。此外,具有可選擇性地通過細胞膜並進入細胞內的構形(configuration)亦較佳。其中,上述構形可為由上述組成物(A)或組成物(B)經亞胺生成反應後的化合物再經過環化、立體選擇性(stereoselectivity)的變換等形態轉換所得的構形。
透過將上述反應材料所得的螢光醣類衍生物使用於細胞檢測,可實現快速簡便、低成本、生物相容性良好且無毒性的螢光檢測技術。
以下,將參照圖1A以及圖1B概略地說明本發明之螢光醣類衍生物之反應機制。其中,圖1A為以組成物(A)作為反應材料之實例,圖1B則為以組成物(B)作為反應材料之實例。
請先參照圖1A,此實例中所使用的還原醣類化合物為D-葡萄糖,而含胺基化合物則以化學式RNH2
表示。其中,R表示經取代或未經取代的任意有機基團。
D-葡萄糖為醛醣(aldose)的一種,其分子結構中具有醛基和醇基,且D-葡萄糖於平衡狀態下可具有環狀以及直鏈狀形態之異構物。如圖1A所示,當D-葡萄糖與含胺基化合物在適當環境下混合時,胺基中親核性的氮原子會攻擊D-葡萄糖上醛基的碳,含胺基化合物與D-葡萄糖進行縮合反應,脫去水分子而形成含亞胺基的席夫鹼(Schiff base)。值得注意的是,上述葡萄糖衍生物可同時以直鏈狀的席夫鹼及經環化的D-葡基胺(D-glucosylamine)(即,N-取代的葡萄糖胺(N-substituted glycosylamine))存在,兩者間可相互轉換達到一平衡關係。
其中,上述席夫鹼包含具有螢光特性的C=N鍵結,故而在適當波長的激發下可放出螢光;上述經環化的D-葡基胺則由於其類似於葡萄糖之構形,而可透過細胞上的葡萄糖運轉蛋白(glucose transporter,GLUT)進入細胞內。經環化的D-葡基胺進入細胞後,可部分轉換為直鏈狀的席夫鹼之型態而發出可偵測之螢光。
因此,利用上述葡萄糖衍生物具有螢光以及可進入細胞內的特性,即可透過檢測細胞中的螢光強度,來檢測不同的細胞對於葡萄糖的攝取量之差異。此外,尤其所使用的材料均為與生物體相容性極高的分子,因此即使用於人體中亦不會對任何組織造成傷害。
接下來,請參照圖1B,此實例中使用了可進行自身亞胺生成反應的葡萄糖胺作為胺醣類化合物,因此,化學式R1

如圖1B所示,具有一級胺基的葡萄糖胺分子亦可與含羰基之化合物進行類似於圖1A所示機制之反應,從而形成具有螢光特性的含亞胺基之席夫鹼。由於在圖1B中,由葡萄糖胺衍生而來的席夫鹼之構形為原本的葡萄糖胺分子經一部份修飾後的結構,仍具備與原本的葡萄糖胺分子類似之構形,故其亦可透過細胞上可運送葡萄糖胺的蛋白進入細胞中。藉此,同樣可透過檢測細胞中的螢光強度,來檢測不同的細胞對於糖類的攝取量之差異。圖1A與圖1B是以葡萄糖及葡萄糖胺作為例子來進行說明,所屬技術領域中具通常知識者應了解本發明中其他螢光醣類衍生物之應用原理也是相類似的,其差異僅在於所辨識的分子或通過細胞時所利用的傳輸機制不同。
具體而言,本發明所提及的螢光醣類衍生物適用於癌症之檢測、微生物之檢測以及篩選與調控細胞攝取醣類能力相關之藥物等方面。以下,將先對於此些應用進行說明,但實際上的應用並不限於此,所屬技術領域中具通常知識者應可由本揭露內容進一步推知其他可能的實施態樣。
本發明所提及的螢光醣類衍生物可應用於癌細胞之檢測。一般而言,癌細胞的代謝活動旺盛,且於癌細胞表面會有異常大量的葡萄糖運送體,使得癌細胞相較於正常細胞會更快速地吸收醣類分子。又,特定癌細胞可能對特定醣類分子具特異性(如肝癌細胞對半乳糖之特異性),故利用此特性以及於修飾後醣類衍生物具螢光的特性,即可區別癌細胞與正常細胞。
在下述表1中示出數種醣類運轉蛋白之受質特異性(substrate specificity)以及各種組織表現(tissue expression)情形。
適用於此種檢測方法的癌症類型例如是乳癌、大腸
癌、直腸癌、食道癌、頭頸部癌、肺癌、淋巴癌、黑色素癌、甲狀腺癌、子宮頸癌、卵巣癌、攝護腺癌、口腔癌、血癌、肝癌、神經膠質瘤,但並不限於此。其中,較佳的是細胞對於特定醣類分子的攝取量明顯大於正常細胞數倍以上的癌症類型,例如乳癌、大腸癌、直腸癌、食道癌、頭頸部癌、肺癌、淋巴癌、黑色素癌、甲狀腺癌、子宮頸癌、卵巣癌、攝護腺癌、口腔癌、血癌、肝癌、神經膠質瘤。所屬技術領域中具有通常知識者應可依據臨床上的需求判斷其適用程度,因此無需加以限定。
本發明所提及的螢光醣類衍生物亦可應用於微生物之檢測。具體而言,由於醣類為大部分微生物的重要能量來源,因此亦可將本發明所提及的螢光醣類衍生物應用於食品、環境毒物簡易檢測等檢測技術中,檢測食品或環境中是否有微生物的存在,以及檢測環境中是否含有抑制微生物生長的毒物或致癌物之存在。舉例而言,可適用於檢測大腸桿菌、幽門螺旋桿菌、綠膿桿菌、鏈球菌、沙門氏菌、金黃色葡萄球菌、酵母菌、黴菌等微生物,或用於檢測人為因素產生的有機物質及重金屬之環境毒物,包括工業用溶劑、化學反應原料及產物、工業廢棄物、工廠廢水排放、農藥及鉛、砷重金屬等具生物毒性之物質與致癌物,但並不限於此。
此外,所屬技術領域中具有通常知識者應理解,可依
照所需檢測的微生物類型來選擇所使用的螢光醣類衍生物以及其他實驗條件,從而實現更具特異性的檢測技術。
本發明所提及的螢光醣類衍生物可進一步應用於篩選與調控細胞攝取醣類能力相關之藥物。此處所指的「調控細胞攝取醣類能力」包括增加或減少細胞對醣類攝取量,但不限於此,亦可為其他類型的調控。
在應用於此類藥物的篩選時,可藉由將各種藥物或不同濃度的藥物與細胞混合後,對細胞攝取醣類的情況進行偵測以及比較,而選出調控效果最佳的藥物種類或濃度。舉例而言,可將本發明所提及的螢光醣類衍生物應用於糖尿病、低血糖症、半乳糖血症、肝醣儲積症等醣類代謝異常疾病及醣類分子吸收阻斷劑應用於癌症治療的藥物篩選,但實際上不限於此。
此外,所屬技術領域中具有通常知識者可依照實際需求調整及選擇藥物種類、添加方式以及其他環境條件,以得到最適化的篩選結果。
下文中,將提出實驗例以對本發明進行更詳細的說明。應注意的是,以下各實驗例僅是用來進一步說明本發明中所使用的螢光醣類衍生物之結構特性、或是其在經特定處理或實驗後的結果,而並非用以限定本發明之範圍。
本實驗例中,將以D-葡萄糖及甘胺酸製備D-葡萄糖基甘胺酸(D-glucosylglycine)作為模式反應,來驗證亞胺基產物之存在及其螢光性,並用以驗證細胞之吸收(uptake)。此實驗例中,預期所進行化學反應如下所示,以D-葡萄糖及甘胺酸產生具有亞胺基的D-葡萄糖基甘胺酸(席夫鹼),而經環化的D-葡萄糖基甘胺酸可進入細胞,且可轉換回直鏈的席夫鹼形態並發出螢光。以下,將對於本發明的螢光醣類衍生物之製備與結構鑑定等進一步詳細說明。
於本實驗例中所使用之材料包括D-葡萄糖、甘胺酸、甲醇以及丙酮。螢光醣類衍生物的製備步驟如下(參考修改自P.S.Song,C.O.Chichester,J.Food Sci.31(1966)914.):
1.於圓底瓶中加入75ml甲醇、10g葡萄糖和1.4g甘胺酸。迴流攪拌反應5小時。
2.於反應液中加入25 ml甲醇和25 ml丙酮,加熱並且趁熱過濾,收集濾液後,於濾液中加入300ml丙酮,然後靜置於4℃冰箱中約36小時。
3.以甲醇:丙酮=1:4(v/v)混合液重複沖洗過濾後所得粗產物。以適量的甲醇加熱溶解粗產物,趁熱過濾後,於濾液加入丙酮,進行再結晶。在經過抽氣過濾後,再以混合液清洗,並進行真空乾燥後,得到最後產物。
接下來,對上述最後產物進行分子量鑑定。於此實驗例中,使用LC/MS同步測定產物純度及分子量。所使用的儀器與條件如下:

圖2A為所得的LC圖譜(注射體積:2 μl;錐口電壓(cone voltage):15V)。在圖2A的LC圖譜中,僅存在一明顯峰位(peak position),由此可確認所得的產物純度甚高。圖2B則為上述最後產物經分析所得的MS圖譜(錐口電壓15V;m/z範圍:200~250)。由圖2B的MS圖譜中,可確認該產物之分子量為237(M+1:238),與理論模式比對亦符合應得產物的分子特徵。
以FT-IR光譜來鑑定最後產物之官能基。圖2C為甘胺酸之FT-IR圖譜,而圖2D為上述最後產物經分析所得的FT-IR圖譜。參照圖2C以及圖2D,可知甘胺酸之FT-IR圖譜的δas
(N-H)(1614 cm-1
)、δs
(N-H)(1522 cm-1
)、ρ(N-H)(1112 cm-1
)、v
(C-N)(1034 cm-1
)在反應後消失,而最後產物表現出來的1627 cm-1
則符合v
(C=N)之特徵,顯見最後產物確實含有席夫鹼之型態。
使用HITACHI U3300紫外光-可見光(UV-Vis)光譜儀進一步進行產物鑑定,結果示於圖2E中。請參照圖2E,可知最後產物於290 nm及350 nm附近具有明顯峰位,前者與>C=N之π→π*轉移(transition)之峰位相符,後者則與>C=N之n→π*轉移之峰位相符。顯示最後產物確實具有C=N之鍵結。
綜合上述LC/MS、FT-IR以及UV圖譜之結果,可知所合成之產物確實為葡萄糖基甘胺酸,且其具有席夫鹼之型態。
此外,亦以HITACHI F4500螢光光譜儀分析上述產物的激發光譜及放射光譜,結果示於圖2F中。由圖2F可知,本實驗例中的產物於水溶液中的最佳激發波長為350 nm,而最佳之放射波長則為440 nm。此外,其激發波長又與UV吸收峰相符合,顯示螢光確實與席夫鹼型態之產物有關。
為了確認最後產物是否能為細胞所吸收,利用所合成之葡萄糖基甘胺酸來餵食纖維母細胞3T3細胞。
‧實驗材料
纖維母細胞3T3、RPMI 1640培養液、胎牛血清、丙酮酸(pyruvic acid)、麩胺酸(glutamic acid)、抗生素、黴菌抑制劑、PBS、胰蛋白酶(trypsin)、錐藍(trypan blue)、24孔培養盤、多聚甲醛(paraformaldehyde)以及螢光保護封片劑。
‧實驗步驟
1.將纖維母細胞3T3以DMEM培養液進行培養(上述DMEM培養液含10%胎牛血清、5 mM丙酮酸、5 mM麩胺酸、抗生素及黴菌抑制劑)。細胞培養於含5%CO2
之培養箱。
2.待細胞分裂生長到佔滿培養瓶底部面積約80%時,將培養液吸出,並以1X PBS清洗,加入0.05%胰蛋白酶反應一分鐘後,使細胞分離並加入5 ml培養液,再以8000 g離心10分鐘。
3.離心後,移除上清液,並加入5 ml培養液,將細胞與
培養液均勻混合後,取出30 μl的細胞液加上270 μl的錐藍混合,取出100 μl置於細胞計數器上,並以紅血球計數法計算細胞數。
4.細胞計數完成後,將細胞數調整至1×104
cells/ml,並取1 ml細胞種入置放有玻片之24孔培養盤中,最後將細胞置於37℃、5% CO2
的細胞培養箱,培養1天後加入0.5 mg/ml之螢光葡萄糖基甘胺酸100 μl,過夜反應後,以1X PBS清洗二次(10分鐘/次)。
5.添加4%多聚甲醛至培養盤中,並置放於室溫下二十分鐘以固定細胞,之後移除多聚甲醛,並以1X PBS清洗三次(10分鐘/次)。
6.將含有細胞之玻片加入50 μl螢光保護封片劑進行封片,即可以螢光顯微鏡觀察結果並進行照相。
由實驗結果(請參照圖2G)可知,經餵食後細胞表現出螢光,證明此螢光醣類衍生物能為細胞所吸收。
於此實驗例中,將以老鼠的星狀細胞瘤與非癌細胞之纖維母細胞(用以模擬正常細胞,實質上其繁殖速率較正常細胞為快)做為材料,利用兩者對於螢光醣類衍生物吸收速率差異所導致的螢光強度表現差異來辨識癌細胞的位置。
‧實驗材料
A.自行製備的各種螢光醣類衍生物:
(1)D-葡萄糖(6x10-3
M)與正丁胺(0.03M)於90℃下反應48小時所得之螢光葡萄糖衍生物
(2)D-葡萄糖胺(1.2x10-2
M)於90℃下自身反應48小時所得之螢光葡萄糖胺衍生物
(3)D-葡萄糖胺(6x10-3
M)與正丁胺(0.03M)反應48小時所得之螢光葡萄糖胺衍生物
(4)D-葡萄糖(6x10-3
M)與離胺酸(0.03M)於90℃下反應48小時所得之螢光葡萄糖衍生物
(5)麥芽糖(3x10-3
M)與離胺酸(0.015M)於90℃下反應48小時所得之螢光麥芽糖衍生物
B.細胞株與細胞培養材料
星狀細胞瘤細胞(C6 cell)、非癌細胞之纖維母細胞(3T3 cell)、RPMI 1640培養液、胎牛血清、丙酮酸、麩胺酸、抗生素、黴菌抑制劑、PBS、胰蛋白酶、錐藍、24孔培養盤、多聚甲醛以及螢光保護封片劑。
‧實驗步驟
1.星狀細胞瘤細胞(如C6)及非癌細胞之纖維母細胞3T3分別以RPMI 1640及DMEM培養液進行培養(上述培養液含10%胎牛血清,5mM丙酮酸、5mM麩胺酸、抗生素及黴菌抑制劑)。細胞培養於含5%CO2
之培養箱。
2.待細胞分裂生長到佔滿培養瓶底部面積約80%時,將培養液吸出並以1X PBS清洗,加入0.05%胰蛋白酶反應一分鐘後,使細胞分離並加入5 ml培養液,再以8000 g離心10分鐘。
3.離心後,移除上清液,並加入5ml培養液,將細胞與培養液均勻混合;其中,對癌細胞之星狀細胞瘤細胞進行活體紅色螢光追蹤劑DiDo染料(long-chain dialkylcarbocyanines,DiDo dye)之結合反應,反應30分鐘並以PBS清洗。
4.取出C6與3T3的細胞液各30 μl,加上270 μl的錐藍混合,取出100 μl置於細胞計數器上,並以紅血球計數法計算細胞數。
5.細胞計數完成後,將細胞數各自調整至1×104
cells/ml,並各取0.5 ml細胞種入置放有玻片之24孔培養盤中,將含活體紅色螢光追蹤劑之C6細胞及3T3細胞共同培養於37℃、5% CO2
的細胞培養箱,培養1天後,加入各組不同種類之螢光醣類衍生物。
6.細胞經與螢光醣類衍生物反應16小時後,以1X PBS清洗二次(10分鐘/次),接著加入4%多聚甲醛置於室溫下二十分鐘以固定細胞,之後移除多聚甲醛再以1X PBS清洗三次(10分鐘/次)。
7.將含有細胞之玻片加入50 μl螢光保護封片劑進行封片,即可以螢光顯微鏡觀察結果並進行照相。
圖3~圖7分別為上述(1)~(5)的各螢光醣類衍生物於螢光顯微鏡下所觀察到的影像。如圖3~圖7所示,上述螢光醣類衍生物(1)~(5)可為癌細胞吸收並發出螢光(綠色),此外,癌細胞亦可吸收DiDo染料而發出螢光(紅色)。將上述兩者影像進行疊合(MERGE)後,發現未顯現紅色
螢光的細胞亦不會顯現綠色螢光,而若細胞發出綠色螢光,即表示其為癌細胞。
由上述實驗結果,確認藉由偵測細胞吸收螢光醣類衍生物的情況,可辨識癌細胞的位置。
由於微生物具有需要碳源之特性,故其亦會吸收本發明中的螢光葡萄糖衍生物,而表現出螢光。於本實驗例中,將驗證可利用上述特性來進行微生物之檢測。
‧實驗材料
菌種(酵母菌)、載玻片、蓋玻片、接種環(針)、酒精燈、YPD酵母菌培養液。
‧實驗步驟
1.將接種環垂直於酒精燈燒紅,以進行滅菌,待其稍冷後沾取酵母菌液。將沾取之菌液各別置入含液體培養基之試管中,蓋上瓶蓋並置入震盪器培養器中培養12小時。
2.於酵母菌液中,加入D-葡萄糖胺(1.2x10-2
M)於90℃下自身反應48小時所得之螢光葡萄糖胺衍生物,並置入震盪器培養器中培養2小時。
3.準備以酒精浸過且以火焰滅菌過之載玻片,以滴管滴入少量無菌水於玻片上,並將接種環(針)以火焰滅菌後,沾取少量菌落(酵母菌),將沾菌的接種環塗抺於有水載玻片上,並將無菌水與菌落混合均勻,在室溫下將玻片置於無菌操作台內陰乾。
4.將陰乾後之玻片(塗抺面向上)快速通過火焰數次以進行固定,加入封片膠後,蓋上蓋玻片,並置於螢光顯微鏡下觀察,結果示於圖8中。
如圖8所示,於共軛焦螢光顯微鏡下可觀察到發藍色螢光的菌體,確認藉由此方法,可檢測微生物的存在。
圖9為依照本發明之一實施例所繪示的將螢光醣類衍生物應用於藥物篩選之反應機制示意圖。如圖9所示,在細胞經「能調控細胞攝取醣類能力之藥物」(含醣類分子吸收阻斷劑)處理後(左圖),其對醣類之吸收能力將受到抑制,因而表現出較未處理之原細胞(右圖)為低之螢光。依此原理,可藉由比較經處理與未處理細胞外顯螢光之強弱,簡易地判斷藥物是否確實具有調控細胞攝取醣類能力之作用。
一般而言,微生物於在含有生物毒性物質之環境中,其生長會受到威脅(甚或死亡),而對於碳源之吸收將下降,故對螢光醣類衍生物之吸收能力自然亦下降。依此原理,可利用比較在待檢測樣品中與正常環境下微生物餵食螢光醣類衍生物後外顯螢光之強弱,簡易地判斷待檢測樣品否含有生物毒性。
圖10為利用螢光醣類衍生物快速而簡易地檢測環境中是否含有危害生物之有毒物質的操作原理示意圖。如圖10所示,試管A及試管B中均含有培養液以及一定量的大腸桿菌,但試管B中添加有環境檢測物,試管A則作為對照組。
將試管A及試管B置放於37℃之培養箱中培養1小時,之後進行抽氣過濾,分別將試管中含有微生物的液體倒於含有濾紙(濾徑為0.2μm以下)的過濾器,於過濾後,微生物將留存於濾紙上。接下來,可利用固定式或可移動式紫外燈來照射含有微生物的濾紙,並比較兩者螢光的強弱,以判定環境檢測物中是否含有具生物毒性的物質。
若環境檢測物不含有具生物毒性的物質,則由試管A及試管B所得的濾紙發出的螢光強度應相當;若含有具生物毒性的物質,則包含環境檢測物的試管B所得的濾紙之螢光強度較弱。藉此方式,即可簡單而快速的進行環境毒物之檢測。
綜上所述,藉由本發明所提供的螢光醣類衍生物於細胞檢測之用途,可實現快速簡便、低成本、生物相容性良好且無毒性的螢光檢測技術。此外,由於其生物相容性良好,應用於生醫檢測時對細胞不具毒性,從而不會對細胞造成損害,故非常適用於癌細胞之檢測、微生物檢測以及藥物篩選等領域。
雖然本發明已以實施例揭露如上,然其並非用以限定本發明,任何所屬技術領域中具有通常知識者,在不脫離
本發明之精神和範圍內,當可作些許之更動與潤飾,故本發明之保護範圍當視後附之申請專利範圍所界定者為準。

Claims (11)
- 一種螢光醣類衍生物之用途,用於細胞之檢測,包括:將至少一樣品與該螢光醣類衍生物一起培養;對該至少一樣品放射一激發光;以及偵測從該至少一樣品放出的螢光,其中該螢光醣類衍生物為由一不具螢光特性的反應材料經一亞胺生成反應所得,且該反應材料為選自一組成物(A)或一組成物(B),該組成物(A)包括一還原醣類化合物以及一含胺基化合物,且該含胺基化合物具有至少一個一級胺基;該組成物(B)包括一胺醣類化合物以及一含羰基化合物,且該胺醣類化合物具有至少一個一級胺基。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其中該還原醣類化合物為選自由葡萄糖、麥芽糖、果糖、乳糖、半乳糖、甘露糖、纖維雙糖、木糖、阿拉伯糖、核糖、去氧核糖、糊精、D-葡萄糖胺以及乙醯葡萄糖胺所組成的群組。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其中該含胺基化合物為選自胺基酸或烷基胺。
- 如申請專利範圍第3項所述之螢光醣類衍生物之用途,其中該含胺基化合物為選自由丁胺、辛胺、1-十二烷基胺、1-十六烷基胺、1,6-己二胺、甘胺酸、丙胺酸、纈胺酸、白胺酸、異白胺酸、苯丙胺酸、色胺酸、酪胺酸、 天門冬胺酸、組胺酸、天冬醯胺、谷胺酸、賴胺酸、谷氨醯胺、甲硫胺酸、精胺酸、絲胺酸、蘇胺酸、半胱胺酸以及脯胺酸所組成的群組。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其中該胺醣類化合物為選自由D-葡萄糖胺、半乳糖胺以及幾丁胺糖所組成的群組。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其中該含羰基化合物為選自由丙酮、異戊醛、D-葡萄糖胺以及乙醯葡萄糖胺所組成的群組。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其中該胺醣類化合物與該含羰基化合物為相同的化合物。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其用於癌症之檢測,其中藉由偵測從與該螢光醣類衍生物一起培養後之該至少一樣品放出的該螢光來辨識癌細胞的位置。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其用於微生物之檢測,其中藉由偵測從與該螢光醣類衍生物一起培養後之該至少一樣品放出的該螢光來確認該微生物的存在。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其用於篩選與調控細胞攝取醣類能力相關之藥物,其中該至少一樣品包括經該與調控細胞攝取醣類能力相關之藥物處理後的細胞以及未經該藥物處理之細胞,且藉由 比較與該螢光醣類衍生物一起培養後之該處理後的細胞與該未處理之細胞的螢光強度,來判斷該藥物是否具有調控細胞攝取醣類的能力。
- 如申請專利範圍第1項所述之螢光醣類衍生物之用途,其用於環境毒物之檢測,其中該至少一樣品包括各自含有微生物的兩樣品,且該兩樣品中之一者更含有一環境檢測樣品,且藉由比較與該螢光醣類衍生物一起培養後之該兩樣品的螢光強度,來判斷待該環境檢測樣品之中是否含有生物毒性物質。
