將正常人類頭髮毛乳頭細胞(東洋紡績股份有限公司製)使用毛乳頭細胞增殖培養基(東洋紡績股份有限公司製)培養後,經由胰蛋白酶處理將細胞回收。回收的細胞使用含有10體積%FBS的Dulbecco’s MEM(Dulbecco’s modified Eagle培養基(1),日水製藥股份有限公司製)稀釋為1.0×104個細胞/mL之濃度後,於塗布膠原纖維的96孔平盤中每孔接種200μL,並培養3日。培養後,移除培養基,於各孔中添加溶解於無血清DMEM(Dulbecco’s modified Eagle培養基(1),日水製藥股份有限公司製)的被驗試料200μL,再培養4日。毛乳頭細胞增殖作用係使用MTT分析來測量。培養終了後,移除培養基,於各孔中添加以終濃度0.4mg/mL之溶解於無血清之DMEM的溴化3-(4,5-二甲基-噻唑-2-基)-2,5-二苯基四唑鎓(MTT,同仁化學研究所股份有限公司製)100μL。培養2小時後,以2-丙醇100μL萃取於細胞內生成的藍色甲月替(formazan)。萃取後,測量於波長570nm的吸光度(OD570)。同時測量作為濁度之波長650nm中的吸光度(OD650),取兩者的差作為藍色甲月替生成量。Normal human hair dermal papilla cells (manufactured by Toyobo Co., Ltd.) were cultured using dermal papilla cell proliferation medium (manufactured by Toyobo Co., Ltd.), and the cells were recovered by trypsin treatment.
