6 웰 플레이트 (well-plate)에 5×104 세포 / 웰로 분주하고 24시간 배양한 다음 시료를 농도별로 처리하고 72시간 동안 배양하였다. 배양한 세포를 1× PBS로 세척한 후 Protease inhibitor cocktail을 1%로 첨가한 RIPA 버퍼 (NaCl : 150mM Tritonx-100 : 1% sodium deoxycholate : 1% SDS : 0.1% Tris-HCI, pH7.5 : 50mM EDTA, pH 8.0 : 2mM)로 세포를 라이시스(lysis) 하였다. 원심분리하여 상층액을 분리하였다. 상층액의 단백질 농도는 BCA 단백질 어세이 키트(protein assay kit)로 정량하였다. 그런 다음 96-웰 플레이트에 20 ㎕의 정량한 단백질과 2 ㎎/㎖의 L-DOPA 80㎕을 넣고 37℃에서 2시간 동안 반응시켰다. 그 후 spectrophotometric microplate reader를 사용하여 490 nm에서 흡광도를 측정하였다.In a 6-well plate (well-plate), 5×10 4 cells/well were aliquoted and cultured for 24 hours, then samples were treated by concentration and cultured for 72 hours. Then, 20 μl of the quantified protein and 80 μl of 2 mg/ml L-DOPA were added to a 96-well plate and reacted at 37° C. for 2 hours. Then, the absorbance was measured at 490 nm using a spectrophotometric microplate reader.
<실험예 1> 크리소에리올(chrysoeriol)의 세포독성 평가<Experimental Example 1> Cytotoxicity evaluation of chrysoeriol
B16F10 멜라노마(melanoma) 세포에서 크리소에리올(chrysoeriol)이 세포 생존에 미치는 영향을 평가하기 위해 MTT 실험을 진행하였다. B16F10 melanoma 세포에 크리소에리올(chrysoeriol)을 0.625 내지 5 ㎍/㎖로 72 시간동안 처리하였다. 세포 생존율은 아무것도 처리하지 않은 세포에 대한 백분율을 표시하였다.In order to evaluate the effect of chrysoeriol on cell survival in B16F10 melanoma cells, an MTT experiment was performed. 분석결과 5 ㎍/㎖ 농도로 처리한 경우, 세포의 생존율이 87% 로 약간의 세포독성을 나타내는 것을 확인하였다. 반면, 2.5 ㎍/㎖ 이하의 농도로 처리시 생존율일 90% 이상을 유지하는 것을 확인하였다(도 1). 양성대조군은 α-MSH (100 nM)을 사용하였다. 분석결과, 크리소에리올(chrysoeriol) 처리 농도가 0.3125 ㎍/㎖에서 2.5 ㎍/㎖으로 증가함에 따라 멜라닌 함량이 증가하는 경향을 나타내는 것을 확인하였다. 구체적으로 처리 농도 중 2.5 ㎍/㎖ 농도로 처리한 세포에서 가장 멜라닌 함량이 높으며, 양성군 보다 높게 나타냄을 확인하였다(도 2). 전술한 실험결과에서 크리소에리올(chrysoeriol)이 멜라닌 함량을 증가시킴을 확인하였는바, 크리소에리올(chrysoeriol)이 B16F10 세포에서 멜라닌 합성의 중요한 효소인 티로시나아제(tyrosinase)의 활성에 미치는 영향을 분석하였다.
