Abstract

A process of separating and purifying tryptophan (Trp) from zymotic fluid by ion-exchange displacement chromatography, which comprises: (a) adjusting a zymotic fluid, which comprises 5.55~12 g/l of tryptophan and 30~40 g/l of ammonium ion, to pH of 3~4 and temperature of 20 deg.C~40 deg.C followed by pre-treatment of sedimentation and centrifugation so that the zymotic fluid comprises 5.55~11.69 g/l of tryptophan and 10~40 g/l of ammonium ion, adjusting the zymotic fluid to pH of 3~4 and temperature of 25 deg.C~35 deg.C; (b) injecting the zymotic fluid into a H-type ion-exchange resin column which contains particles at size of 5 mm ~ 600 mm and is at pH of 2~4; (c) using purified water to perform back washing in order to flush out un-adsorbed zymotic fluid retained in the column; and (d) using ammonia solution with concentration of 0.92~1.13 g/l to undergo displacement chromatography at space velocity of 5~10 ll-1hr-1.
