Abstract

Prodn. of haemoproteins (esp. cytochrome C) and proteins from yeasts comprises (a) subjecting the yeast to cellular lysis in a liq. medium under pressure; (b) purifying the lysate by precipitating nucleic acids (e.g. with MnCl2) and salting out cell residues (e.g. with NH4 sulphate); (c) adjusting the purified lysate to pH 8-8.5 and passing it over an ion-exchange resin to adsorb the cytochrome; (d) adjusting the effluent to pH 3.3-4 and precipitating proteins with (NH4)2SO4; (e) washing the resin with an aq. alkaline soln. with a pH of 8-8.25; and (f) eluting the cytochrome with an aq. soln. of (NH4)2SO4. The process gives high yields (e.g. 250 g/tonne) of high-purity cytochrome C and high yields (e.g. 340 g/tonne) of high-purity proteins suitable for use in human and animal nutrition.
