現今,一般認為益生菌(亦稱為直接飼餵微生物(direct-fed microbial))是合適的飼料添加劑。它們主要作為抗生素的替代品。除了將它們用作飼料成分之外,已經進一步揭露可將益生菌替代地應用於飲用水。在這種情況下,該技術的狀態中已經揭露益生菌的不同應用形式。
因此,例如WO 2008/107746揭露含有益生菌微生物和大量多醣(40至80wt%的微晶纖維素和8至25wt%的羧甲基纖維素鈉)作為崩解助劑的錠劑。
根據本發明,令人驚訝地發現,飼料組成物(特別是壓實的飼料組成物)確實溶解迅速而沒有留下不想要的殘留物,並且如果它們包含大量的甜菜鹼鹽,則進一步實現了飼料添加劑(特別是益生菌)的均勻分佈。
進一步發現,藉由使用本發明的組成物可避免在儲水器中形成不想要的生物膜。
此外,本發明之壓實的組成物使得飼料添加劑可被舒適地處理並容易地施用於動物。再者,它們展現非常良好的儲存穩定性。
因此,本發明的第一標的是包括至少一種甜菜鹼鹽和至少一種飼料成分的組成物。
甜菜鹼也稱為三甲基甘胺酸或2-三甲銨基乙酸酯(2-trimethylammonioacetate)。原則上,根據本發明可以使用各種甜菜鹼鹽。根據本發明較佳使用的甜菜鹼的鹽是甜菜鹼與強酸(特別是強無機酸)的鹽,其中特別較佳是甜菜鹼鹽酸鹽。其他甜菜鹼鹽的實例特別是甜菜鹼溴化物和甜菜鹼水楊酸鹽。
較佳地,甜菜鹼鹽(特別是甜菜鹼鹽酸鹽)在組成物中的含量為20至90wt%,特別是30至80wt%,更佳是40至75wt%,最重要的含量是45至65wt%。與組成物是僅包括一種甜菜鹼鹽或不同甜菜鹼鹽的混合物這一事實無關,如前所述的較佳範圍亦與組成物中所含甜菜鹼鹽的總量有關。
根據本發明的組成物可例如是粉末、顆粒或凝膠,但較佳是壓實的組成物,特別是壓實的粉末或壓實的顆粒。
因此,本發明的較佳標的特別是一種壓實的組成物,其包含至少一種甜菜鹼鹽,特別是甜菜鹼鹽酸鹽,其含量為20至90wt%,較佳為30至80wt%,更佳是40至75wt%,最重要的含量是45至65wt%,其中與組成物僅包括一種甜菜鹼鹽或不同甜菜鹼鹽的混合物這一事實無關,如前所述的較佳範圍亦與組成物中所含甜菜鹼鹽的總量有關。
根據本發明的「壓實的(飼料)組成物」通常是指包含至少一種甜菜鹼鹽和至少一種飼料添加劑的組成物,其中藉由施加外部壓力進行壓實。根據本發明之壓實的飼料組成物較佳是錠劑。
因此本發明的另一標的亦為製備根據本發明之壓實的組成物的方法,其包括以下步驟:提供包括組成物的所有成分(特別是至少一種甜菜鹼鹽和至少一種飼料添加劑)的顆粒狀組成物,並且隨後較佳在添加黏合劑之後壓製該顆粒狀組成物以形成壓實的組成物(特別是錠劑)。
用於製備該壓實的組成物的顆粒狀組成物具有較佳為小於3wt%(特別是0.5至3wt%)的水含量。其進一步較佳地包括平均粒徑為2 µm至6 mm(特別是5 µm至5 mm)的顆粒。較佳藉由首先以粉末形式提供組成物的不同組分,接著例如藉由流體化床製粒將這些組分製粒來獲得顆粒狀組成物。
較佳藉由施加5至100 kN(較佳是10至 50 kN,特別是20至40 kN)的壓力來進行組成物的壓實。較佳是藉由使用錠劑壓製來進行壓實。例如可使用旋轉壓力機(rotary press)或偏心壓力機(eccentric press)作為用於進行壓實的壓力機(press)。
壓實的飼料組成物(特別是錠劑)較佳展現80至150 N(特別是90至130 N)的硬度。例如,可藉由使用MultiTest 50 Tablet Hardness Tester(瑞士Sotax AG的Dr. Schleuninger® Pharmatron)容易地確定組成物的硬度,而不論其形狀如何。
該組成物可具有任何種類的空間形狀,特別是可具有球形、立方體、圓錐形、圓柱體、棱柱形、金字塔形、四面體或錠劑的形狀。較佳地,其具有帶有兩個等距平面的錠劑的形狀,特別是具有圓形或角形形狀的平面,其中具有圓形的平面的片劑是非常佳的。
壓實的飼料組成物(特別是錠劑)較佳具有10至50 mm(特別是20至40 mm,更佳是25至35 mm)的直徑。該錠劑進一步較佳具有25至140mm(更佳是60至100mm)的厚度。
壓實的飼料組成物(特別是錠劑)較佳具有3至25 g(特別是6至20 g,更佳是6至15 g)的重量。其進一步較佳具有3至15 ml(更佳是4.5至9 ml)的體積。
甜菜鹼鹽在該組成物中作為質子供體的作用,並且當溶解於水時,在碳酸鹽和/或碳酸氫鹽存在下,其能起泡沫作用。因此,為了能起泡沫作用,根據本發明之組成物較佳亦包括至少一種碳酸鹽和/或碳酸氫鹽。
如果存在,碳酸鹽在根據本發明之組成物中的含量較佳為10至35wt%,特別是15至30wt%,更佳為20至30wt%。原則上,根據本發明可使用各種碳酸鹽。根據本發明之較佳的碳酸鹽是碳酸與強鹼(特別是與強無機鹼)的鹽。根據本發明所使用的非常佳的碳酸鹽是碳酸鈉。 如前所述,較佳範圍與組成物中所含碳酸鹽的總量有關,而與組成物是否僅包含一種、兩種或幾種不同的碳酸鹽無關。
除了碳酸鹽以外或代替碳酸鹽,較佳在組成物中亦存在至少一種碳酸氫鹽。如果存在,碳酸氫鹽在組成物中的存在量較佳為1至10wt%,更佳為2至8wt%,特別是3至6wt%。原則上,根據本發明可使用所有種類的碳酸氫鹽。根據本發明之較佳的碳酸氫鹽是質子化碳酸與強鹼,特別是與強無機鹼的鹽。根據本發明所使用的非常佳之碳酸氫鹽是碳酸氫鈉。如前所述,較佳範圍是關於組成物中包含的碳酸氫鹽的總量,而與組成物是否僅包含一種、兩種或幾種不同的碳酸氫鹽無關。
除了甜菜鹼鹽外,組成物可進一步包含更少量的其他質子供體,特別是選自多官能酸,較佳是選自多官能果酸,尤其是選自檸檬酸、己二酸、琥珀酸、戊二酸、蘋果酸、酒石酸、葡萄糖酸和那些酸的鹽,以及所述化合物之混合物。如果存在,另外的質子供體較佳以小於20wt%的總量存在,更佳是以0.1至15wt%的量存在,特別是以5至15wt%的量存在。
在本發明的較佳實施例中,組成物包括檸檬酸或其鹽,其量為2至20wt%,特別是5至15wt%。

潤滑劑特別用於促進壓實的組成物從壓力機的基質分離。潤滑劑可包含在組成物本身中,或者恰好在壓實步驟之前放在組成物上和/或基質上。
如果使用乳化劑,則較佳為少量使用它們,特別是總量為0.1至3wt%。較佳的乳化劑之例子是卵磷脂。乳化劑使組成物中所含組分的混合物穩定。

更佳的水生動物是養殖魚,其隨後被加工成魚粉(fish meal)或魚油。就此而言,魚較佳是鯡魚(herring)、緣青鱈(pollack)、油鯡(menhaden),鯷魚(anchovie)、毛鱗魚(capelin)或鱈魚。
在另一較佳實施例中,動物是飼養供食用或作為食品生產者的農場動物,例如家禽、豬和反芻動物。
家禽可選自生產性或家禽,也可選自花式家禽(fancy poultry)或野禽。在本文中,較佳的生產性家禽是雞、火雞、鴨和鵝。在本文中,生產性家畜較佳是為生產幼畜而優化的家禽或為帶肉而優化的家禽。較佳的花式家禽或野禽是孔雀、野雞、鷓鴣(partridge)、石雞(chukkar)、珠雞(guinea fowl)、鵪鶉、紅松雞(capercaillie)、松雞(grouse)、鴿子和天鵝,特別較佳是鵪鶉。進一步較佳的家禽是平胸鳥(ratite),特別是鴕鳥和鴯鶓(emus),以及鸚鵡。
根據本發明的反芻動物較佳是選自牛、山羊和綿羊。在一實施例中,可將本發明的組成物餵給反芻動物以增強它們的健康,特別是減少這些動物腹瀉的發生率。前反芻動物(preruminant)是反芻動物(ruminant),包括小牛、年齡範圍從出生到大約十二週。
根據本發明可使用的益菌生(prebiotics)較佳為寡糖,特別是選自於半乳糖寡糖、甲矽烷基寡糖(silayloligosaccharide)、乳果糖、乳糖蔗糖、果糖寡糖、帕拉金糖(palatinose)或異麥芽糖寡醣、糖基蔗糖、麥芽寡糖、異麥芽寡糖、環糊精、龍膽寡糖(gentiooligosaccharide)、大豆寡糖、低聚木糖(xylooligosaccharide)、葡聚醣、果膠、半乳醣醛酸聚醣(polygalacturonan)、鼠李糖半乳醣醛酸聚醣(rhamnogalacturonan)、甘露聚醣(mannan)、半纖維素、阿拉伯半乳聚醣(arabinogalactan)、阿拉伯聚醣(arabinan)、阿拉伯木聚醣(arabinoxylan)、抗性澱粉、甲基二糖(mehbiose)、幾丁聚醣、瓊脂糖(agarose)、菊粉(inulin)、塔格糖(tagatose)、聚葡萄糖和藻酸鹽。
可用於根據本發明之飼料組成物且可添加在飼料消化中的酵素較佳是選自於植酸酶(phytase)(EC 3.1 .3.8或3.1.3.26)、木聚醣酶(xylanase)(EC 3.2.1.8)、半乳聚醣酶(galactanase)(EC 3.2.1 .89)、半乳糖苷酶(galactosidase)(特別是α-半乳糖苷酶(EC 3.2.1.22))、蛋白酶(protease)(EC 3.4)、磷脂酶(phospholipase)(特別磷脂酶A1 (EC 3.1 .1.32)、A2 (EC 3.1.1.4)、C (EC 3.1.4.3)和D (EC 3.1.4.4))、溶血磷脂酶(lysophospholipase)(EC 3.1 .1.5)、澱粉酶(amylase(特別是α-澱粉酶(EC 3.2.1.1 ));溶菌酶(lysozyme)(EC 3.2.1 .17)、聚葡萄糖酶(glucanase)(特別是β-聚葡萄糖酶 (EC 3.2.1.4或EC 3.2.1.6))、葡糖澱粉酶(glucoamylase)、纖維素酶、果膠酶(pectinase)、或任何其混合物。
市售可得的植酸酶之實例包含Bio-Feed™植酸酶(Novozyme)、Ronozyme® P和HiPhos™ (DSM營養產品)、Natuphos™ (BASF)、Finase®和Quantum® Blue(AB Enzyme)、Phyzyme® XP (Verenium/DuPont)以及Axtra® PHY (DuPont)。其他較佳的植酸酶包含例如WO 98/28408、WO 00/43503和WO 03/066847中所描述的那些。
市售可得的木聚醣酶之實例包含Ronozyme® WX和G2 (DSM營養產品)、Econase® XT和Barley (AB Vista)、Xylathin® (Verenium)以及Axtra® XB (木聚醣酶/β-聚葡萄糖酶,DuPont)。市售可得的蛋白酶之實例包含Ronozyme® ProAct (DSM營養產品)。

可使用的免疫調節劑是例如抗體、細胞素、噴霧乾燥的血漿、白介素或干擾素,或其組合。
根據本發明可使用的礦物質是例如硼、鈷、氯化物、鉻、銅、氟化物、碘、鐵、錳、鉬、硒、鋅、鈣、鎂、鉀或鈉,或其組合。
根據本發明可使用的胺基酸是例如離胺酸、丙胺酸、蘇胺酸、甲硫胺酸、纈胺酸或色胺酸、或其組合。

有機酸與其鹽和酯。此有機酸的實例特別是乙酸、α-酮戊二酸、砷酸、苯甲酸、檸檬酸、阿魏酸(ferulic acid)、甲酸、富馬酸、沒食子酸、葡萄糖酸、腐殖酸、酮異己酸(ketoisocaproic acid)、乳酸、蘋果酸、丙酸、樹脂酸(resin acid)。
如果一些成分被列在上述不同種類的組中,或者由於它們的特性而分配給這些不同種類的組,則組成物中成分的較佳上限是由具有最高上限之組的極限確定,且組成物中成分的較佳下限是由具有對低下限之組的極限確定。再者,在此情況下,僅在具有最高上限的組的較佳總量中考慮成分的量。
工作實例
實例1:測定含有益生菌的錠劑之溶解時間
如表1所示,藉由將各成分按表1中揭露的量混合來製備含有不同量的檸檬酸、甜菜鹼鹽酸鹽、碳酸氫鈉、碳酸鈉、二氧化矽和益生菌(平均粒徑(d50)約為0.9 µm的B. amyloliqufaciens)之粉末。檸檬酸加甜菜鹼鹽酸鹽的總量相當於每個製備樣品中質子供體的總量。使用Eirich混合機EL1將如此獲得的粉末以20m/sec的速度充分混合240秒,最後以15m/sec的速度製粒45秒。
隨後藉由分別施加30 kN的壓力將2.3 g粒狀組成物壓製成圓形錠劑。
隨後將由此獲得的錠劑溶解在5L水中,並確定溶解時間。
從表中可以看出,僅含有甜菜鹼鹽酸鹽作為質子供體的錠劑之溶解時間是最佳的。檸檬酸的量越高,溶解時間越短。再者,組成物中檸檬酸含量的增加亦確實導致錠劑從壓制機基質中釋放的問題增加。
再者,觀察到益生菌均勻地分佈在儲水器中。

實例2:根據本發明之錠劑的定性性質
為了確定與市售的具有糖基載體的益生菌粉相比之本發明的錠劑之定性性質(亦即,特別是使用者友好性(user-friendliness)),藉由將根據本發明之錠劑和市售可得的益生菌粉末溶解在水箱中所含之1000 L水中,調節益生菌濃度為5×10^11 CFU/1000 L水來測試差異。所觀察到的特性總結於下表中。

實例3:生物膜形成的測定
為了測定市售可得之含糖益生菌所製備的錠劑和根據本發明之錠劑的生物膜形成,在錠劑製備中將益生菌替換為市售可得之具糖基載體的益生菌粉末。製備沒有益生菌的錠劑作為負控制組。
根據本發明之錠劑含有以下成分:11wt%益生菌(解澱粉芽孢桿菌(B. amyloliquefaciens) CECT 5940)、51wt%甜菜鹼HCl、10wt%檸檬酸、23wt%碳酸鈉、4wt%碳酸氫鈉、1.5%發煙矽石(fumed silica) (Aerosil® 200F)。製備具有相同成分的錠劑,其中將益生菌以市售的含糖益生菌粉代替作為比較例。
為了模擬在養雞場中使用的飲用水管線的條件,將不同的配方溶於水(處理水,20℃)中,以將水中的益生菌濃度調節為約5×10^ 8 CFU/L。然後將含水的益生菌泵送經由一個由3個Schott 1L燒瓶組成的系統,該等燒瓶分別藉由直徑為4 mm且長度為300 mm的不鏽鋼管和PVC而連接。將Schott燒瓶中的水持續攪拌。藉由使用3個Watson Marlow 120U泵來建立水泵(pumping of the water)。每天更換含有益生菌的溶液。每天抽水16小時,模擬家禽飼養的條件,家禽每天休息8小時。流速從第一週的9.8 mL/min增加到第六週的46.2 mL/min。
6週後,藉由使用結晶紫將在PVC和不鏽鋼管中形成的生物膜染色來確定生物膜形成的量。藉由George A. O’Toole (2011), Microtiter Dish Biofilm Formation Assay. JoVE. 47. http://www.jove.com/details.php?id=2437, doi: 10.3791/2437中所述內容進行生物膜的染色。形成的生物膜越多,染色越深。藉由測量在550 nm波長(OD550
)的光密度來確定染色強度。結果如下表所示。

實驗數據顯示藉由使用本發明的錠劑,可將生物膜的形成最小化。生物膜的形成與負對照組幾乎相同,亦即就像使用完全不含益生菌的錠劑一樣。相對地,用市售可得的含糖益生菌代替錠劑中的益生菌會導致生物膜形成之顯著的不想要有的增加。
實例4:飼養試驗(Feeding Trial)
農場由6所房屋組成,每所房屋約有35.000隻鳥。試驗中使用的家禽為Ross 308,混交,平均起始重量為40 g。家禽飼餵標準飲食40天。向三個房屋(處理組(treatment group))提供飲用水中所含的益生菌解澱粉芽孢桿菌(B. amyloliquefaciens) CECT 59,而其他三個房屋(對照組)則沒有提供益生菌。
治療組的益生菌供應如下:
將具有約4.5 E11 CFU/錠劑的8 g錠劑溶解在20 L水中以製備儲備溶液。錠劑含有0.8 g檸檬酸、4.0 g甜菜鹼鹽酸鹽、0.3 g碳酸氫鈉、1.8 g碳酸鈉、0.9 g解澱粉芽孢桿菌(B. amyloliquefaciens) CECT 59 (Ecobiol®)、0.1 g發煙矽石(fumed silica) (Aerosil® 200F)。
在第0-2、10-12、21-23和27-29天的每一天,經由飲水線以1:50的比例向家禽提供儲備溶液12小時。
飼養試驗的結果總結於下表中。

在三個房屋中,將含益生菌的錠劑應用於飲用水導致家禽的平均體重增加,每隻鳥平均增加34克。與未處理的家禽相比,該應用進一步使處理過的家禽的飛節燒傷和足底皮炎減少超過5%。再者,工廠廢品的數量顯著減少。
Claims (15)
- 一種組成物,其包括至少一種甜菜鹼鹽和至少一種飼料添加劑。
- 如請求項1的組成物,其中該甜菜鹼鹽的含量為20至90 wt%,特別是30至80 wt%,更佳為40至75 wt%,尤其是45至65 wt%,以及該甜菜鹼鹽較佳為甜菜鹼鹽酸鹽(betaine hydrochloride)。
- 如請求項1或2的組成物,其中該組成物包含至少一種碳酸鹽,其含量較佳為10至35 wt%,特別是15至30 wt%,更佳為20至30 wt%,以及該至少一種碳酸鹽較佳為碳酸鈉。
- 如請求項1至3中任一項的組成物,其中該組成物包含至少一種碳酸氫鹽,其含量較佳為1至10 wt%,更佳為2至8 wt%,特別是3至6 wt%,以及該至少一種碳酸氫鹽較佳為碳酸氫鈉。
- 如請求項1至4中任一項的組成物,其中該組成物包含至少一種多官能酸,較佳是檸檬酸,其中該多官能酸(特別是檸檬酸)較佳係以0.1至15 wt%,特別是5至15 wt%的量存在。
- 如請求項1至5中任一項的組成物,其中該組成物包含至少一種選自以下的其他化合物:其他崩解劑、流動調節劑、抗結塊劑、潤滑劑、乳化劑、抗氧化劑、增積劑(bulking agent)、膠化劑、著色劑和調味劑。
- 如請求項1至6中任一項的組成物,其中該飼料添加劑是選自益生菌、礦物質、維生素、酶、益菌生(prebiotic)、胺基酸、有機酸、疫苗、免疫調節劑及其混合物,且其含量較佳是至多25 wt%,特別是1至20 wt%,更佳為2至18 wt%,尤其是5至15 wt%。
- 如前述請求項中任一項的組成物,其中該組成物包含至少一種益生菌微生物,其含量較佳為1×1010 至1×1013 CFU/g,較佳為1×1011 至1×1012 CFU/g。
- 如請求項8的組成物,其中該益生菌微生物是選自益生菌細菌,特別是芽孢桿菌屬(Bacillus),較佳是選自枯草芽孢桿菌(B. subtilis)、地衣芽孢桿菌(B. licheniformis)、解澱粉芽孢桿菌(B. amylolyquiefaciens)、糞腸球菌(E. faecium)及其混合物。
- 如前述請求項中任一項的組成物,其中該組成物是壓實的組成物並且較佳是具有錠劑(tablet)的形狀。
- 一種製備如請求項10的組成物之方法,其包括以下步驟: a) 提供具有如請求項1至9中任一項所揭露之成分的粉末組成物; b) 將該粉末造粒以製備顆粒狀組成物,其中該顆粒較佳具有2 µm至6 mm、特別是5 µm至5 mm的平均直徑; c)壓制該顆粒狀組成物以形成該壓實的組成物,特別是錠劑。
- 如請求項11的方法,其中壓制該顆粒狀組成物是藉由施加5至100 kN、較佳是10至50 kN、特別是20至40 kN的壓力來進行。
- 一種向動物提供飼料添加劑的方法,其包括以下步驟: a) 將如請求項1至10中任一項的組成物溶解在水或水溶液中,較佳是在飲用水或飼養用水中; b) 向動物提供該溶液。
- 如請求項13的方法,其中該動物是選自農場動物、寵物、外來動物、動物園動物,水生動物以及用於運動、娛樂或工作的動物。
- 如請求項13或14的方法,其中向該動物提供該飼料添加劑促進該動物的健康和/或改善該動物的一般身體狀況和/或改善該動物的飼料轉換率和/或降低該動物的死亡率和/或提高該動物的存活率和/或提高該動物的體重增加和/或提高該動物的生產力和/或提高該動物的疾病抵抗性和/或增加該動物的免疫反應和/或在該動物中建立或維持健康的腸道菌群和/或防止病原細菌的生長和/或促進有益細菌的生長和/或減少病原體經由該動物的糞便排出。
