本發明係說明含有穩定的水性亞錫離子之組成物及其製備及使用的方法。
亞錫離子(二價)易被大氣中的氧氣,特別於此等組成物中所發現之主要水介質之中的氧氣所氧化。該氧化作用使錫離子(四價)於水介質中大量地水解而形成導致沉澱物及混濁性之不溶性氫氧化物或氧化物。這是,例如於口腔護理調配物之任何定量亞錫離子分析時所不想要的,其必須排除大氣中的氧氣來進行,以避免可溶性、可量化之亞錫離子損失如沉澱,而不易量化之錫離子。
本發明之某些具體實例係提供穩定的含有水性亞錫離子之組成物,其含有包括呈形式氧化態+II之溶解錫,一種或多種溶解之硝酸鹽及不與呈形式氧化態+II之溶解錫螯合的溶解之羧酸之液體相,該液體水相具有pH值2至4。

"液體水相"一詞於本發明之目的時較佳係指該液相於室溫為液體。"水性"應瞭解為該液相典型地含有70至99重量百分比的水,如藉由卡爾-費氏滴定法(Karl-Fischer titration)所測定者,據此,該水含量之上限亦可藉著於該液相中所發現之其他組份之量來測定。
於某些具體實例中,該液體水相於室溫具有動態黏度不超過1500 mPa.s。於其他具體實例中,該液體水相於室溫具有動態黏度為500至1500 mPa.s。於某些具體實例中,該分子氧為可溶解於該液體水相中之大氣氧。
然而令人驚奇地,現今已觀察到溶解之硝酸鹽,特別當與溶解之羧酸(其係不與呈形式氧化態+II之溶解錫螯合者)合併使用且於pH為2至4時,不會被其他pH-依賴(例如過碘酸鹽、過錳酸鹽、過硫酸鹽、鐵(III)氯化物、過羧酸鹽如過苯甲酸及過醋酸,過一硫酸鹽、過二硫酸鹽)或pH-獨立之氧化劑可觀地改變其之可氧化性,其中,"pH-獨立"宜瞭解為pH-獨立於1至7,更佳於2至6之pH範圍(例如分子鹵素如分子氯、溴或碘,特別分子碘;三碘化物如三碘化鉀;六氰基鐵酸鹽
如六氰基鐵酸鉀,或銀鹽如硝酸銀或過氯酸銀)。
此係指呈形式氧化態+II之溶解錫的定量測定,使用對於硝酸鹽之水相中之錫離子的定量氧化步驟沒有不利的影響,因而,亦可無需操作特別之預防措施來對抗大氣中的氧氣如脫氣及/或惰性氣體。用於定量測定呈形式氧化態+II之溶解錫的方法,包括終點檢測法,其於硝酸鹽存在下,於該液體水相中可類似液體水相中不含硝酸鹽之相關測定。該終點檢測法,當氧化劑於其之氧化及還原狀態具有不同的顏色時,可藉顏色來進行。因此,實例為氧化性測定,係使用分子碘或三碘化物作為氧化劑;本文中,存有過量氧化劑可藉碘與澱粉之錯合物之藍色來顯示,然還原性碘化物陰離子為無色。終點檢測法之其他實例為電化學檢測法,當存有不再還原之過量氧化劑時,可藉電極如鉑電極上之電位升高而看到。該呈形式氧化態+II之溶解錫的定量測定可為直接測定法,亦即,其中該呈形式氧化態+II之溶解錫係用氧化劑直接滴定至終點,或其可為間接者,亦即,首先加入過量氧化劑使呈形式氧化態+II之溶解錫全部氧化成錫離子,且然後將該過量氧化劑用另一種還原劑,如硫代硫酸鹽反滴定至終點。本文中所用之"呈形式氧化態+II之溶解錫"一詞意欲涵括溶解於該液體水相中呈形式氧化態+II之所有的離子性錫類。此等錫類之實例為水合之亞錫離子、亞錫離子之可溶離子性或非離子性錯合物及亞錫離子之離子性氫氧錯合物。該呈
形式氧化態+II之溶解錫的來源不重要且可藉所要分析之樣品來給定。僅係藉舉例來說明,氯化亞錫、氟化亞錫、氫氧化亞錫,及硫酸亞錫係作為該來源之實例而給定。
於某些具體實例中,係使用溶解於水相中之硝酸鹽。"硝酸鹽"一詞係涵括所有含有一個或多個NO3 -基團(配位至陽離子或作為相對陰離子(類))的水溶性無機類。硝酸鹽之實例為存在於水相中之金屬陽離子nitrato(硝酸配基)錯合物,特別該呈形式氧化態+II之溶解錫的亞錫離子形成之部份,溶劑化之硝酸鹽相對陰離子及未解離之硝酸。該溶劑化之硝酸鹽陰離子係假設為被水分子所溶劑化且具有基本上非錯合陽離子,如鈉、鉀之硝酸鹽陰離子或胺氟化物陽離子之陽離子(參見如下)作為相對離子。因為所有此等硝酸鹽一般係彼此熱力平衡,其不可能個別地測定此等硝酸鹽之各個莫耳數。然而,所有此等硝酸鹽之共同特點是,於鹼化作用時其等係藉去質子作用及/或藉金屬陽離子的沈澱作用由任何硝酸配基錯合物之不溶性氫氧化物或氧化物而轉化為溶劑化之硝酸鹽陰離子。該溶解之硝酸鹽可藉著將硝酸鹽,宜為鹼金屬硝酸鹽(例如鋰、鈉或鉀硝酸鹽),鹼土金屬硝酸鹽(例如鎂或鈣硝酸鹽),或銨或四烷基銨鹽(例如銨、四甲基銨、四乙基銨或四丁基銨硝酸鹽)的鹽加至水相而獲得。更佳係添加硝酸鹽為其鹼金屬鹽,最佳者為硝酸鈉或硝酸鉀。
於某些具體實例中,不與呈形式氧化態+II之溶解錫螯合的羧酸係任意地共-使用。此應瞭解對呈形式氧化態+II之溶解錫,該酸之共軛鹼的錯合形成常數Kcb
至多十倍於對呈形式氧化態+II之溶解錫之醋酸鹽之錯合形成常數Ka
此係與log(K cb /K a )1同義。
於上式中,Kcb及Ka[M]為未錯合之呈形式氧化態+II之溶解錫的濃度;[L]為非錯合共軛鹼的濃度;[Ac]為非錯合醋酸鹽的濃度;[MAc 2 ]為經醋酸鹽錯合之呈形式氧化態+II之溶解錫的濃度,且[ML n ]為被所說明之共軛鹼所錯合之呈形式氧化態+II之溶解錫的濃度(以n為共軛鹼之莫耳數)。
此等K cb 及K a 之測定可於室溫於水溶液中,於pH高於所研究之酸/共軛鹼對之pKa值二個單位時電化學地完成(例如當測定Ka時,以6.75用於醋酸/醋酸鹽配對);其係藉a)下式之值
對ln([M])之值繪製,於給定、恆定之共軛鹼總濃度時得到,[L 0 ],但於呈形式氧化態+II之溶解錫之可變總濃
度時,[M 0 ],對應之[M]係用亞錫離子-選擇性電極電化學地測定,如說明於Int.J.Electrochem.Sci.5,215-231(2010)頁中者,b)於能斯特(線性)區域中以固定斜率1線性地內推該繪製值對,且c)得到ln(K cb )或ln(K a ),作為所得到之內推線與該標繪圖之y-軸的交叉點。
此等測定可於足夠低之濃度(於[L0]及[M0]兩種情形時典型地不超過0.2 M)下完成,該濃度自身(非活性)可用於該公式中。
因為依如上之定義該共軛鹼被要求為不與呈形式氧化態+II之溶解錫螯合,所以甚至該羧酸本身為不與呈形式氧化態+II之溶解錫螯合者。
於某些具體實例中,該非螯合羧酸宜為一元羧酸,其未被任何羥基所取代及/或係選自於包括下列之基團:C1-C4直鏈或分支的鏈烷酸,任意經烷基取代之C3-C5直鏈烯酸及C6-C14芳基羧酸。於未被任何羥基所取代之一元羧酸之較佳的實例為C1-C4直鏈或分支的鏈烷酸,其係選自由下列組成之基團:甲酸、醋酸、丙酸及2-甲基丙酸;,任意經烷基取代之C3-C5直鏈烯酸,其係選自由下列組成之基團:丙烯酸,甲基丙烯酸,甘菊花酸及白芷酸;及C6-C14芳基一元羧酸,其係選自由下列組成之基團:苯甲酸及1-或2-萘羧酸。未
被任何羥基所取代之一元羧酸之更佳的實例為C1-C4直鏈或分支的鏈烷酸,且最佳者為醋酸。
該羧酸,依該液體水相之pH而定,可與特定數量之其之共軛鹼達成平衡。然而,為了本發明之目的,該共軛鹼不需要以此等數量呈現,以與游離酸一起形成實際的緩衝系統。
本文中所用之"酸之共軛鹼"係指可由所述之羧酸藉由移除羧基質子而獲得之化學種類;或,於完全質子化之多羧酸的情況中,係指可藉由移除第一個(最酸的)羧基質子而獲得之化學種類,或,於多羧酸其等之羧基已部份去質子之情況中,係指可藉由移除另一個羧基質子而獲得之化學種類。
為了本發明之目的,所需之羧酸應溶解於水相中。此意味著該羧酸宜具於室溫水中之溶解度至少約0.1克/100毫升水。
於某些具體實例中,與共使用之非螯合羧酸其整體液體水相的pH應於2至4之範圍。於某些具體實例中,該液體水相之pH係於2.5至3.5之範圍且甚至更佳於2.7至3.3之範圍。於其他具體實例不含共使用非螯合之羧酸者,該液體水相之pH宜於4至6之範圍,更佳於4.5至5.5之範圍且甚至更佳於4.7至5.3之範圍。
呈形式氧化態+II之溶解錫的總數量並非係絕對的且可藉所想要之該液體水相的目的及/或溶解於該水相中之上述亞錫離子之鹽的溶解度而單獨地測定。為了本
發明之目的,硝酸鹽之總量為以硝酸鹽陰離子可測得之氮的總數量者,{N},其較佳係經選擇使得硝酸鹽過量超過呈形式氧化態+II之溶解錫。硝酸鹽對呈形式氧化態+II之溶解錫之莫耳比典型的範圍係0.5:1至20:1較佳為5:1至15:1。
本發明組合之溶解之硝酸鹽與非螯合羧酸,於含有呈形式氧化態+II之溶解錫之穩定液體水相對抗於酸性介質中被大氣中的氧所氧化時,係為有效的方法。
溶解之硝酸鹽單獨,或與非螯合之羧酸合併,可使用於下列者中,例如:
a)分析應用,其中,該呈形式氧化態+II之溶解錫的含量係藉氧化作用定量地測定,而此等氧化分析之一般說明業已給定於前。該含有穩定的水性亞錫離子之組成物,其包括可為口腔護理調配物之液相的液體水相。本文中,已觀察到本發明之組合物於避免呈形式氧化態+II之溶解錫氧化的功效不會被口腔護理調配物液相中所發現之其他組份所減低。該液體水相亦可為鍍錫廢金屬去錫處理之母液。於此等去錫處理中,習用係將含有水性硝酸鈉或硝酸鉀於鹼性介質中,與大氣中的氧氣作為輔氧化劑沸騰。該氧化性溶解的錫應定量地予以沉澱如錫氧化物或氫氧化物,且基本上於該母液中沒有餘留之呈形式氧化態+II之溶解錫。此等情況為當呈形式氧化態+II之溶解錫的殘留微量需要定量地測定;此等
微量於藉由氧化劑氧及大氣中的氧之分析中非常容易氧化地降解且因此特別需要穩定作用者。本文中,硝酸鹽一般係已存在,且可添加無機酸或非螯合羧酸以改變該pH至有用的範圍,而得到適用於該分析之液體水相的試樣。
b)分析其中呈形式氧化態+II之溶解錫的功效係於各個態樣,如鑒於抗細菌活性上進行分析。本文中,於分析時,該呈形式氧化態+II之溶解錫再次容易地被氧化轉化為不溶性者且從而成為不活性錫化合物。該氧化作用使該呈形式氧化態+II之溶解錫對抗所分析之細菌得到人為之低效益;使用本發明組合物係提供具有呈形式氧化態+II之溶解錫含量之穩定的分析溶液,其於分析期間係穩定的。
c)再者,硝酸鹽、非螯合之羧酸及呈形式氧化態+II之溶解錫可於另一個較佳具體實例中預形成為包含液體水相的組成物,其具有pH 2至4,可用作貯存之儲備溶液或中間溶液,其可於當且根據需要作為組成份時加至口腔護理調配物之製備中。因為此等儲備溶液或中間體溶液係再次穩定的對抗由分子氧的氧化作用,其可貯存且於口腔護理調配物之生產設施時延長於槽中或容器中之處理時間,而無需特殊防止氧化作用之預防措施。可藉著使用如此之儲備溶液或中間溶液而製備之口腔護理調配物可為任何型式含亞錫離子之習用包括含水液相的
口腔護理調配物,如漱口劑、牙膏、接觸溶液或人工唾液。組成此等口腔護理調配物所需之其他組成份或賦形劑為習用者且可不受限制地使用及類似其等於相關口腔護理調配物使用。因為該儲備溶液或中間溶液具有pH自2至4之微酸性,其可直接賦予生理上可接受之pH,亦即如由4至6,更佳由4至5至製成的口腔護理調配物。
本發明之適用性的其他領域可由下文所提供之更詳盡的說明會變得顯而易見。應瞭解當指明本發明之較佳具體實例時,該詳細的說明及特定之實例意欲用於闡明之目的且並非意欲限制本發明之範圍。
該測定係藉電勢碘滴定法使用自動滴定器(Metrohm公司之Titrando 809,瑞士),鉑電極(Metrohm公司之6.1204.310電極型,瑞士)及參考電極而完成。該方法係闡明多至約400ppm呈形式氧化態+II之溶解錫之目標含量。
將大約50毫升硝酸鹽/醋酸溶液(大約2000 ppm硝酸鹽及pH大約3且添加醋酸)轉移至150毫升燒杯中。
使用容量吸移管精確地添加50.0毫升樣品。
於攪拌下,將5.00毫升標準0.1 N KI3溶液精確地加至該產生之測量溶液中。這是對呈形式氧化態+II之溶解錫之過量三碘化物。
未被呈形式氧化態+II之溶解錫所還原之餘留三碘化物/碘,於添加標準KI3溶液後,用標準0.1M硫代硫酸鈉反滴定至終點。當接近終點看得到三碘化物之褐色明顯減弱時,可添加一些馬鈴薯澱粉,只要存有任何三碘化物由於其之包含碘之錯合物型式,其將變成且維持藍色。
呈形式氧化態+II之溶解錫以ppm計的含量係以
計算,其中,V為於終點時所添加之標準0.1 M硫代硫酸鈉溶液之體積(以毫升計)。
呈形式氧化態+II之溶解錫的原理、裝設儀器及最大目標濃度係如於實例1者。
將精確已知數量之牙膏樣品稱重於50毫升燒杯中。該數量不應如此之大而使得呈形式氧化態+II之溶解錫之莫耳數超過5毫莫耳。將20.0毫升硝酸鹽/醋酸溶液(參見實例1)於磁棒攪拌下加入,直到該樣品完全溶解且該產生之懸浮液為均勻。將該產生的懸浮液倒入拋棄式50毫升離心試管中。將該燒杯用另外10.0毫升硝酸鹽/醋酸溶液沖洗且將此加至試管。將精確之10.0毫升二乙醚加至離心試管中,將該管用溶劑潮汐帽

呈形式氧化態+II之溶解錫以ppm計之含量係以
計算,其中,V為於終點時所添加之標準0.1 M硫代硫酸鈉溶液之體積(以毫升計)且m為如上牙膏樣品之稱重數量,以克計。
首先將該水相過濾以移除任何固體,然後將1.0000克±0.1毫克之濾出物於容器中精確地稱重且添加水使總重量為20.0000克±0.1毫克。
該測定係藉離子層析法於該溶液之20微升樣品上完成:儀器:Dionex IC 25離子層析儀,其具有自動取樣器AS 50、沖提液產生器EG 40及EluGen筒KOH。
管柱:Dionex Ion Pac AS 14,內徑(i.d.)4毫米,長度250毫米,含預-管柱Ion Pac AG14A,內徑4毫米,長度50毫米。
抑制系統:Dionex陰離子自體產生抑制器(Self Regenerating Suppressor),(ASRS-ULTRA II內徑4毫米)。
沖提液:40 mM氫氧化鉀溶液。該鹼性沖提液與樣品接觸後,其中所含之所有的硝酸鹽皆轉化為硝酸鹽陰離子。
流速:每分鐘0.9毫升。
於20微升樣品中之硝酸鹽陰離子{NO3 -}以微莫耳計之莫耳數,係於該樣品之離子層析法中,由硝酸鹽峰之面積,使用硝酸鹽陰離子之莫耳數(以微莫耳計)對峰面積之校正曲線來評估。該校正曲線係藉著於相同離子層析條件下測量20微升等分量含有已知,但於有用範圍內之可變莫耳數量之硝酸鉀溶液而製得。
如此所得到之{NO3 -}的數值,以微莫耳計,等於如上1.0000克濾出物中之可測量作為硝酸鹽離子之氮的莫耳數,{N}(以毫莫耳計)。然後,假設硝酸鹽之全部類型皆溶解於該水相中而各含有一NO3 -部分(經配位或作為陰離子),則該{N}直接等於硝酸鹽本身溶解於如上1.0000克水相中之莫耳數。
將五等份之試驗溶液,每500毫升溶液各含有267毫克SnF2及1.2克硝酸鈉(呈形式氧化態+II之溶解錫的名目含量400ppm,0.03 M硝酸鹽)每者進行試驗。將該試驗溶液任其與大氣中的氧接觸且攪拌達2天。呈形式氧化態+II之溶解錫的實際含量係使用類似於一種實例1的方法間隔地測量。
該五種試驗溶液之其他性質及所觀察之呈形式氧化態+II之溶解錫(ppm)的實際含量係如下表:
試驗溶液1顯示當於中性pH使用非螯合去質子化pH調節劑(醋酸鈉)及硝酸鹽時,該呈形式氧化態+II之溶解錫快速地氧化為錫離子;生成氧化錫/氫氧化錫沉澱物。試驗溶液2顯示當於酸性pH使用非螯合質子化之pH調節劑(醋酸)及硝酸鹽時,該呈形式氧化態+II之溶解錫於製備該溶液中起始損失後,緩緩地開始再提升略微趨向該名目值。試驗溶液3顯示當於中性pH使用螯合去質子化之pH調節劑(葡糖酸鈉)及硝酸鹽時,進一步繼續進行超過30小時後該呈形式氧化態+II之溶解錫具有輕微的起始損失。試驗溶液4顯示當於酸性pH使用螯合質子化pH調節劑(葡糖酸)及硝酸鹽時,非常緩慢地繼續進行超過30小時後該呈形式氧化態+II之溶解錫具有起始損失。試驗溶液5顯示當於微酸性pH使
用硝酸鹽而不含另外之pH調節劑時,該呈形式氧化態+II之溶解錫於正常含量下保持穩定的對抗大氣中的氧超過30小時。
本文中所說明之實施例及其他具體實例係為範例且非意欲用來限制說明本發明之組成物及方法的全部範圍。特定具體實例、材料、組成物及方法可於本發明之範圍內進行具有實質上類似結果之相同改變、修正及變化。
Claims (20)
- 一種穩定的含有水性亞錫離子之組成物,其含有包括下列之液體水相:呈形式氧化態+II之溶解錫,溶解之硝酸鹽;及溶解之羧酸,其不與溶解呈形式氧化態+II螯合之錫,其中,該液體水相具有2至4之pH值。
- 如申請專利範圍第1項之組成物,其中該羧酸為未被任何羥基所取代之一元羧酸。
- 如申請專利範圍第1或2項之組成物,其中該一元羧酸係選自由下列所組成之基團:C1-C4直鏈或分支的鏈烷酸、任意經烷基取代之C3-C5直鏈烯酸及C6-C14芳基羧酸。
- 如前述申請專利範圍中任一項之組成物,其中該羧酸為C1-C4直鏈或分支的鏈烷酸。
- 如前述申請專利範圍中任一項之組成物,其中該羧酸為醋酸。
- 一種用於改良溶解於液體水相中呈形式氧化態+II之錫對抗被分子氧氧化的穩定性之方法,其包括溶解於液相中之硝酸鹽及不與呈形式氧化態+II之錫螯合的羧酸,且調整該液體水相之pH至2到4。
- 如申請專利範圍第6項之方法,其中該羧酸為未被任何羥基所取代之一元羧酸。
- 如申請專利範圍第6或7項之方法,其中該羧酸係選自下列所組成之基團:C1-C4直鏈或分支的鏈烷酸、任意經烷基取代之C3-C5直鏈烯酸、C6-C14芳基羧酸及胺基酸。
- 如申請專利範圍第6至8項中任一項之方法,其中該羧酸為C1-C4直鏈或分支的鏈烷酸。
- 如申請專利範圍第6至9項中任一項之方法,其中該羧酸為醋酸。
- 一種用於定量測定溶解於液體水相中呈形式氧化態+II之錫的方法,其中該呈形式氧化態+II之錫係藉由對錫離子定量氧化而測定,其中該液體水相包含溶解之硝酸鹽。
- 如申請專利範圍第11項之方法,其中該液體水相還包含溶解之羧酸,其不與溶解呈形式氧化態+II之錫螯合且具有pH值於2至4之範圍。
- 如申請專利範圍第11或12項之方法,其中該羧酸為未被任何羥基所取代之一元羧酸。
- 如申請專利範圍第11至13項中任一項之方法,其中,該羧酸係選自由下列所組成之基團:C1-C4直鏈或分支的鏈烷酸,任意經烷基取代之C3-C5直鏈烯酸及C6-C14芳基羧酸。

- 如申請專利範圍第11至16項中任一項之方法,其中對錫離子之氧化作用係用pH-獨立之氧化劑來完成。
- 如申請專利範圍第18項之方法,其中該pH-獨立之氧化劑為碘或三碘化物。
- 一種用於製備口腔護理調配物之方法,其包括將如申請專利範圍第1至5項中任一項之組成物與習用口腔護理調配物組成份或賦形劑混合。
