Description
сокой физиологической активностью и могут найти применение в фармацевтической промышленности .The invention relates to the production of new asen-yin derivatives, which possess high physiological activity and can be used in the pharmaceutical industry.
The azepine derivatives of the general formula are obtained.
где RI - алкил;where RI is alkyl;
R2 is hydrogen, lower-branched Ci-C4-alkyl, isopronyl, or allyl. The most interesting pharmacological properties are those in which R2 is a lower alkyl and also E are salts with inorganic or organic acids. The proposed method for the preparation of these compounds is based on the reaction of an alkyl alcohol on the nitrogen atom and consists in that the compounds of the general formula

Эта реакци протекает преимуцествепно в органическом растворителе, в когором образующа с соль трудно раствор етс , что позвол ет отделить ее фильтрацией, акнми растворител ми вл ютс , например, метанол, ацетон, метилэтилкетон.where R2 is as defined above, is alkylated with alkanol reactive esters of the general formula RI-OP, where RI is as defined above, in the presence of organic solvents and basic condensing agents. Pelled products are isolated as a base or converted into salts, for example, by "acid." This reaction proceeds predominantly in an organic solvent, in the coorus the resulting salt is difficult to dissolve, which allows it to be separated by filtration, and the solvents are, for example, methanol, acetone, methyl ethyl ketone.
пл. 194-196°С.poured with stirring to 40 ml of d formamide. Then, 0.8 g of sodium hydride was added to the resulting suspension. At a temperature of 25-35 ° C. The mixture is stirred for 1 hour lrn: temperature 80 ° C, cooled to 20 ° C and pacTiBop 5.1 g of propyl iodide in 15 ml of absolute benzene is poured into the mixture at this temperature for 30 minutes. The reaction mixture is then stirred for 18 hours at a temperature of 50-55 ° C, cooled to 20 ° C, and water or frost ether is added. The organic phase is separated and extracted with 2 L hydrochloric acid. From the hydrochloric acid extract, the free base is precipitated with the help of ammoniated ammonium nitrate and treated with diethyl ether. The emulsion solution is washed with water, dried over potassium carbonate and evaporated to dryness in a rotary evaporator. The residue is recrystallized from alcohol; 2-ethyl-8-propyl-1,2,3,8-tetrahydro-dibenzo-b is obtained, f-pyrrol-3,4-c is azepine, t. Pl. 105-107 ° C. To a solution of 2e of the obtained base in 20 ml of acetone was added 0.87 g of oxalic acid in 5 ml of absolute ethanol, the oxalate crystals were isolated, m.p. 192-194 ° C (from absolute ethanol). Similarly, 2,8-dimethyl-1,2,3,8-tetrahydro-dibenzo-6, / -pyrrolo-3,4-d -azepine, m.p. 120-121 ° C (ethanol), its oxalate, t. Pl. 230-232 ° C; 2-ethyl-8-methyl-1,2,3,8-tetrahydrodibe-iso-b, / -pyrrolo-3,4-α-azepium, its oxalate, m.p. 208-210 ° С (from metapol); 2-allyl-8-methyl-1, 2,3,8-tetrahydrod ibenzo - b, f lirrol-3, 4-c-azepine, t. Pl. 137-139 ° C (from absolute ethanol), methanesulfonate, so pl. 194-196 ° C.
