Abstract
Способ повторений для получения гликозилтрансфераз, используемых для катализируемого гликозилтрансферазой синтеза сахаридного состава последовательным соединением предварительно выбранных сахаридных единиц с иммобилизованным акцепторным остатком, который является представителем, выбираемым из группы, состоящей из протеионов, гликопротеинов, липидов, гликолипидов и углеводов. Способ содержит в первой итерации (повторении) стадии (i) контактирования акцепторного остатка, иммобилизованного к твердой или полутвердой подложке, со смесью, содержащей по крайней мере одну гликозилтрансферазу, в условиях, эффективных для осуществления соединения указанной гликозилтрансферазы с иммобилизованным акцепторным остатком, с получением в результате комплекса иммобилизованный акцепторный остаток - гликозилтрансфераза, затем (ii) контактирования комплекса иммобилизованный акцепторный остаток - гликозилтрансфераза с предварительно выбранным сахаридом в условиях и в присутствии сореагентов, достаточных для осуществления соединения предварительно выбранной сахаридной единицы с иммобилизованным акцепторным остатком, катализированного гликозилтрансферазой, с высвобождением гликозилтрансферазы из комплекса и получением иммобилизованного продукта. За этим следует единичная итерация или множество последовательных итераций стадий (i) и (ii), так, что иммобилизованный продукт, полученный на стадии (ii) данной итерации, используется в качестве иммобилизованного акцепторного остатка на стадии (i) следующей итерации, до тех пор, пока не получится заданный сахаридный состав, и все нужные гликозилтрансферазы также поэт
Claims (1)
- Способ повторений для получения гликозилтрансфераз, используемых для катализируемого гликозилтрансферазой синтеза сахаридного состава последовательным соединением предварительно выбранных сахаридных единиц с иммобилизованным акцепторным остатком, который является представителем, выбираемым из группы, состоящей из протеионов, гликопротеинов, липидов, гликолипидов и углеводов. Способ содержит в первой итерации (повторении) стадии (i) контактирования акцепторного остатка, иммобилизованного к твердой или полутвердой подложке, со смесью, содержащей по крайней мере одну гликозилтрансферазу, в условиях, эффективных для осуществления соединения указанной гликозилтрансферазы с иммобилизованным акцепторным остатком, с получением в результате комплекса иммобилизованный акцепторный остаток - гликозилтрансфераза, затем (ii) контактирования комплекса иммобилизованный акцепторный остаток - гликозилтрансфераза с предварительно выбранным сахаридом в условиях и в присутствии сореагентов, достаточных для осуществления соединения предварительно выбранной сахаридной единицы с иммобилизованным акцепторным остатком, катализированного гликозилтрансферазой, с высвобождением гликозилтрансферазы из комплекса и получением иммобилизованного продукта. За этим следует единичная итерация или множество последовательных итераций стадий (i) и (ii), так, что иммобилизованный продукт, полученный на стадии (ii) данной итерации, используется в качестве иммобилизованного акцепторного остатка на стадии (i) следующей итерации, до тех пор, пока не получится заданный сахаридный состав, и все нужные гликозилтрансферазы также поэтому получатся.
