Изобретение относится к иммунобиотехнологии, а именно к получению гибридом, продуцирующих моноклональные антитела к дигоксину. Полученные гибридомы могут найти применение в иммуноаналитических методах количественного определения дигоксина в биологических жидкостях. Измерение концентрации дигоксина производится с целью лекарственного мониторинга при терапии этим препаратом. Целью изобретения является создание гибридомы, технологичной для наработки мкАТ к дигоксину в виде асцита и продуцирующих мкАТ, пригодные для создания иммунодиагностикумов для определения дигоксина в биологических жидкостях. Спленоциты мышей линии BALB/с, иммунизированных конъюгированным антигеном дигоксина с человечьим сывороточным альбумином, сливали с клетками миеломной линии Р3-Х63.Ag 8653. Клоны, продуцирующие моноклональные антитела к дигоксину, отбирали с помощью твердофазного иммуноферментного анализа. Для проведения скрининга в качестве антигена использовали конъюгат дигоксина с бычьим сывороточным альбумином. Дополнительно качество антител оценивали по ингибированию дигоксина и его аналогами реакции антител с сорбированным антигеном. Отобранные клоны подвергали 2-3 кратному реклонированию для получения стабильных клонов и затем использовали для наработки мкАТ либо в культуре, либо в виде асцита. Очищенные мкАТ использовали для разработки иммуноферментной тест-системы для определения дигоксина.The invention relates to immunobiotechnology, and in particular to the production of hybridomas producing monoclonal antibodies to digoxin. Splenocytes of BALB / c mice immunized with conjugated digoxin antigen with human serum albumin were fused to P3-X63.Ag 8653 myeloma cell line. Clones producing monoclonal antibodies to digoxin were selected using enzyme-linked immunosorbent assay. For screening, a digoxin conjugate with bovine serum albumin was used as antigen. Additionally, the quality of antibodies was evaluated by the inhibition of digoxin and its analogs in the reaction of antibodies with a sorbed antigen. Selected clones were subjected to 2–3 times cloning to obtain stable clones and then used to generate mAb either in culture or as ascites. Purified mAbs were used to develop an enzyme-linked immunosorbent assay for digoxin.
