Изобретение относится к иммунобиотехнологии, а именно к получению гибридом, продуцирующих моноклональные антитела к фенобарбиталу. Полученные гибридомы могут найти применение в иммуноаналитических методах количественного определения фенобарбитала в биологических жидкостях. Измерение концентрации фенобарбитала производится с целью лекарственного мониторинга при терапии этим препаратом. Целью изобретения является создание гибридомы, технологичной для наработки мкАТ к фенобарбиталу в виде асцита и продуцирующих мкАТ, пригодные для создания иммунодиагностикумов для определения ФБ в биологических жидкостях. Спленоциты мышей линии BALB/с, иммунизированных конъюгированным антигеном фенобарбитала с гемоцианином, сливали с клетками миеломной линии Р3-Х63.Ag 8653. Клоны, продуцирующие моноклональные антитела к фенобарбиталу, отбирали с помощью твердофазного иммуноферментного анализа. Для проведения скрининга в качестве антигена использовали конъюгат ФБ с человеческим сывороточным альбумином. Дополнительно качество антител оценивали по ингибированию ФБ и его аналогами реакции антител с сорбированным антигеном. Отобранные клоны подвергали 2-3 кратному реклонированию для получения стабильных клонов и затем использовали для наработки мкАТ либо в культуре, либо в виде асцита. Очищенные мкАТ использовали для разработки иммуноферментной тест-системы для определения фенобарбитала.The invention relates to immunobiotechnology, in particular to the production of hybridomas producing monoclonal antibodies to phenobarbital. The aim of the invention is the creation of a hybridoma, technologically advanced for producing mAbs to phenobarbital in the form of ascites and producing mAbs, suitable for creating immunodiagnostics for determining FB in biological fluids. Splenocytes of BALB / c mice immunized with hemocyanin conjugated phenobarbital antigen were fused to P3-X63.Ag 8653 myeloma cell line. Clones producing monoclonal antibodies to phenobarbital were selected by enzyme-linked immunosorbent assay. For screening, an FB conjugate with human serum albumin was used as antigen. Additionally, the quality of antibodies was assessed by inhibition of FB and its analogues, the reaction of antibodies with a sorbed antigen. Selected clones were subjected to 2–3 times cloning to obtain stable clones and then used to generate mAb either in culture or as ascites. Purified mAbs were used to develop an enzyme-linked immunosorbent assay for phenobarbital.
