Abstract
Description
The invention relates to the field of agriculture, in particular to plant growing, and can be used in obtaining golden root plants ( Rhodiola rosea L.) with increased productivity without the use of chemical plant protection agents or plant stimulation.
There is a known method for growing Rhodiola rosea L. (Rhodiola rosea, golden root) [1]. A well-known method is to use shading and watering vegetatively propagated Rhodiola rosea plants when grown in culture. For shading, special shields are used, which are installed on plots horizontally or at an angle of 45°C. The shields must be kept on the plots throughout the summer, removed only during watering. At the end of July - August, when the plants get stronger, the shields can be gradually removed: first during inclement weather, then in the evening and in the morning, and finally removed from the plots completely.
The disadvantages of this known method are the presence of a number of labor-intensive and energy-consuming plant cultivation techniques associated with shading. In addition, the method does not provide for an increase in any indicators of raw material productivity, but is aimed only at the successful survival of individuals of the species in the unnatural conditions of the plain.
There is a known method of growing Rhodiola rosea described in patent RU 2597244 C2 [2]. It is based on sowing and growing strips of iris rosea to protect Rhodiola rosea from unfavorable conditions with a projective coverage of 40-60%, with the addition of a moisture-retaining partially swollen hydrogel. According to the authors, the method helps to increase the mass of roots by 39.4% from 0.88 g to 2.22 g in individuals of the first year of cultivation, propagated by vegetative means.
Rhodiola rosea.The disadvantage of this method is the use of a foreign agent (hydrogel), which increases the cost of agricultural technology; its use is not always advisable; in areas with sufficient soil moisture, it is necessary to look for other ways to increase the underground mass of Rhodiola rosea .
[3, 4, 5, 6].It is known that algae are a valuable organic fertilizer and can be used as biostimulants for plant growth. In recent years, seaweed extracts have been increasingly used in agriculture to increase crop yields. The effect is achieved by stimulating the processes of plant growth and development, as well as by improving the quality of the final product. Experiments show that the effect depends on the concentration of the extract. It was noted that plants treated with A. nodosum extract always surpassed the control grown in the usual way in all yield parameters. In addition, the addition of A. nodosum extract at a rate of 1.5 kg/ha significantly increased the content of anthocyanins, phenols and dry matter in grapes [3, 4, 5, 6].
A known procedure for obtaining biomass of Rhodiola rosea in vitro according to patent MD894 [7], the method according to the invention involves cultivating callus from the leaves of sterile shoots of Rhodiola rosea on Murashige-Skoog agar medium. nutrient medium with the addition of 1.5 mg/l 6-benzylaminopurine, 0.5 mg/l α-naphthylacetic acid and 0.1% sterile aqueous solution of 5% solids obtained from the biomass of the green alga Spirogira sp. extraction with ethanol.
Thus, the use of algae extract in the cultivation of Rhodiola rosea in vitro is known. However, no information has been found on the use of algae extracts to increase the main indicator of raw material productivity (weight of the underground part) of Rhodiola rosea .
The technical challenge is to develop a method for increasing the main indicator of raw material productivity (the mass of the underground part - rhizomes and roots) of Rhodiola rosea using environmentally friendly stimulants that do not have the disadvantages of chemical means of protecting or stimulating plants, characterized by less labor intensity and greater efficiency.
The technical result consists in a reliable increase in the raw material productivity of the underground part (rhizomes with roots) of Rhodiola rosea by at least 80% intensification of metabolic processes, without the use of chemical means of protection or stimulation of plants.
0,5%.The technical result is achieved through foliar treatment of vegetative plants with a biostimulant during the period of active growth at intervals of 1 time every 2 weeks; an aqueous extract of Ascophyllum nodosum is used as a biostimulator at a concentration of 0.5%.
Rhodiola rosea. Determination of optimal and technological regimes for foliar treatment of vegetative plants Rhodiola rosea with a biostimulator.
During one growing season, foliar treatment of Rhodiola rosea L. was carried out with aqueous solutions of biostimulants - aqueous extract of Ascophyllum nodosum at a concentration of 0.5% and 0.3%, in different application schemes 2 times in 1 week and 1 time in 2 weeks to determine optimal and technological processing modes. The VITAMAR seaweed extract fertilizer (produced by Neotech bio, Russia) was used as an aqueous extract of Ascophyllum nodosum . This preparation of Ascophyllum nodosum extract was chosen due to its availability and low cost.
The control experiment was carried out at the same time and in the same application scheme as with the Ascophyllum nodosum extract; distilled water was used as a control solution for foliar feeding.
1.Individuals of Rhodiola rosea of the pregenerative and generative periods of development were subjected to treatment. The effect of biostimulants on two-year-old Rhodiola rosea individuals was assessed using indicators of the mass of roots and rhizomes, as well as specific surface density of the leaf (SLD). The experimental design is presented in Table 1.
The specific surface density of a leaf is an integral indicator that reflects the accumulation of dry matter per unit leaf area. This indicator expresses the ratio of the dry mass of a leaf to its area and indirectly characterizes the thickness of the leaf. The mass and area of dry leaves were measured, and from the data obtained, the specific surface density of the leaf was calculated as the ratio of dry mass (mg) to area ( cm2 ). Leaf area was measured using AxioVision software (Carl Zeiss). The indicator is used to demonstrate the overall productivity of the plant, understood in this work as an indicator of the ability to create, preserve and transform organic matter.
The calculation was carried out according to the formula.
УППЛ = M/S,UPPL = M/S,
где М - воздушно сухая масса листовой пластинки, мг;where M is the air-dry mass of the leaf blade, mg;
2 [8,9].S is the area of the leaf blade, cm 2 [8,9].
[10]. Raw material productivity was assessed as the mass of the wet underground part (rhizomes with roots) dug out at the end of the growing season, after the above-ground parts died off. Excavation was carried out without choice [10].
All numerical data were initially tested for normality of distribution using the Shapiro-Wilk test. Due to the anomalous distribution of variables, the nonparametric statistics method U-test was used for further statistical processing. Methods of nonparametric statistics were used even if part of the data used in statistical processing had a normal distribution, and the other part had an abnormal distribution.

Examples of the method are presented below.
1.Example 1.
The development of a method for increasing the raw material productivity of the underground part of Rhodiola rosea L. was carried out in open ground conditions on the territory of the educational and experimental site of the Siberian Botanical Garden, located in the south-eastern part of the city of Tomsk on an area of more than 100 hectares during the growing season of 2023. As model plants, we used our own reproductions of Rhodiola rosea L. plants, which have been under the conditions of an introduction experiment for more than twenty years. Rhodiola rosea plants were grown by seedlings in 2021; by the time the experiment began, they represented a multi-age agropopulation of individuals of the pregenerative and generative periods of development. Cultivation took place on meadow-chernozem soils, which are characterized by a high humus content (5.0%) and are well enriched with nutrients phosphorus and potassium (P 2 O 5 235 mg/kg K 2 O 180 mg/kg). The total sample size of the analyzed individuals participating in the experiment was more than 40 specimens for each experimental variant. Foliar application of an aqueous extract of Ascophyllum nodosum 0.3% was carried out at intervals of 2 times every 1 week and 1 time every 2 weeks. The treatments continued throughout the growing season from May to August and were stopped only after the active outflow of plastic substances into the rhizome and roots began.
The data obtained indicate that there is a tendency to increase the LPL indicator when using a treatment scheme of 1 time in 2 weeks. UPPL indicators increased by 3.6%. However, it should be noted that no significant differences were found. When analyzing the effect of a concentration of 0.3% under different application schemes, a tendency was established for an increase in the mass of roots and rhizomes with an application scheme of 1 time in 2 weeks. These data are correlated with each other, however, in this case, it is possible to talk only about a tendency towards an increase in indicators of general and raw material productivity.
2.Example 2.
2).In the case of the technical possibility of carrying out treatments with a higher concentration of the test solution, all other conditions being equal (example 1), the effect of the concentration of 0.5% aqueous extract of Ascophyllum nodosum was assessed in the same application schemes (Fig. 2).
In the case of the FLW indicators at a concentration of 0.5% aqueous extract of Ascophyllum nodosum , no significant differences were found; only a tendency to increase was found with the application schedule of 2 times per week. However, when treated once every 2 weeks, a significantly different increase in the weight of rhizomes and roots was noted by 82.7% with a p value of <0.05 (Fig. 3). The parameter varies in the control from 3.39 g to 22.0, when treated with an aqueous extract of Ascophyllum nodosum at a concentration of 0.5% and the application schedule of 1 time in 2 weeks, the parameter varied from 9.62 g to 29.84 g.
Thus, the developed method eliminates the use of labor-intensive methods for preserving the viability of individuals, and also contributes to a significant increase of up to 82.7% in raw material productivity (weight of rhizomes with roots) of Rhodiola rosea . The developed method allows treatment with an aqueous extract of Ascophyllum nodosum at intervals of 1 time every 2 weeks to obtain positive morphophysiological effects. The reliability of the difference in the increase in the mass of the underground part of Rhodiola rosea when treated with a 0.5% concentration of an aqueous extract of Ascophyllum nodosum with an application schedule of 1 time in 2 weeks was proven with a value of p<0.05, which indicates the objectivity of the developed method.
The proposed method can be used in the development of resource-saving agricultural technologies for the production of medicinal plant raw materials, with the possibility of developing agricultural technology according to the principles of organic production.
