Исследованы состав и структура микробного сообщества почвы при предпосадочной инокуляции клубней картофеля сорта Тулеевский штаммом B. thuringiensis ssp. morrisoni c титром 2,7⋅106 КОЕ/мл, который оказал существенный эффект на бактерии-аммонификаторы. Указанный бактериальный штамм угнетал развитие грибов родов Fusarium и Penicillium. The composition and structure of the soil microbial community were studied during pre-planting inoculation of potato tubers of the Tuleyevsky variety with the strain B. thuringiensis ssp. morrisoni with a titer of 2.7⋅10 6 CFU/ml, which had a significant effect on ammonifier bacteria. This bacterial strain inhibited the development of fungi of the genera Fusarium and Penicillium. A tendency was revealed to reduce the number of spring wheat seedlings affected by root rot when seeds were treated with the strain B. thuringiensis ssp. morrisoni . The inhibitory activity of the B. thuringiensis strain on the 10th day of the experiment against Fusarium oxysporum varied from 68 to 83%, Alternaria alternata from 71 to 89%. The analysis showed that the studied strain stimulated the growth of spring rapeseed and spring wheat plants and exhibited antifungal activity.

Штамм бактерий Bacillus thuringiensis ssp. morrisoni КА9 выделен из природных образцов (погибших личинок колорадского жука, собранных на картофельном поле в Новосибирской области, в 2014г.) с последующей многоступенчатой селекцией: к суспензии штамма Bacillus thuringiensis ssp. morrisoni КА9 в питательном бульоне Луриа-Бертрани добавляли 0,25М ацетата натрия и инкубировали 4ч при 30°С, постоянно перемешивая. Затем образцы прогревали при 80°С в течение 15 минут и точечно высевали на агаризованные среды с различным содержанием аминного азота и глюкозы. Выросшие колонии исследовали световой микроскопией для определения наличия и синхронности высыпания параспоральных кристаллов на всех средах. Отобранные варианты культивировали в жидких питательных средах для определения морфологической однородности и стабильности. Полученный штамм Bacillus thuringiensis ssp. morrisoni КА9 обладал морфологической однородностью и образовывал крупные близкие к прямоугольной форме параспоральные включения разных размеров после культивирования в жидкой питательной среде через 48 часов. Штамм давал положительную реакцию жгутикового антигена с типовой сывороткой H8ab. Штамм был депонирован во Всероссийскую коллекцию промышленных микроорганизмов под номером ВКПМ В-14326, что подтверждается справкой о депонировании от 27 сентября 2022 г. (Справка прилагается). Selected variants were cultivated in liquid nutrient media to determine morphological uniformity and stability. The resulting strain of Bacillus thuringiensis ssp. morrisoni KA9 had morphological uniformity and formed large parasporal inclusions of different sizes, close to rectangular in shape, after cultivation in a liquid nutrient medium after 48 hours. The strain gave a positive flagellar antigen reaction with the H8ab type serum. The strain was deposited in the All-Russian Collection of Industrial Microorganisms under the number VKPM V-14326, which is confirmed by a certificate of deposit dated September 27, 2022 (Certificate attached).

Через 96 ч роста на агаризованной среде «А» образуют овальные споры размером 0,5 ×× 1,0 мкм и параспоральные включения близкие к прямоугольной форме разных размеров (рис.1). На среде «А» штамм образует серые крупные плотные, плоские колонии с мелкозернистой поверхностью, край колоний слегка волнистый. На среде «Б» - колонии крупные плоские, чуть морщинистые с плотным беловатым центром, край - расплывчато-войлочный, серый, поверхность мелкозернистая. На среде Лурия-Бертрани формируются крупные, плоские, плотные, белые колонии с волнистым краем и мелкозернистой поверхностью. В жидких питательных средах рост умеренный, образует равномерную муть. 3. Физиолого-биохимические признаки. Органические источники азота, используемые данным микроорганизмом: пептон, мясные, рыбные и растительные экстракты. Соевая, кукурузная и рыбокостная муки, автолизаты дрожжей и казеина, гидролизаты соевой и кукурузной муки, кормовые дрожжи. Штамм усваивает аммонийные и нитратные формы азота, глюкозу и глицерин в виде углеводного питания. Штамм КА9 продуцирует ацетилметилкарбинол, проявляет амилазную, протеазную активности, не обладает уреазной и лецитиназной активностями. Не сбраживает салицин, но сбраживает маннозу, сахарозу с образованием кислоты. Штамм КА9 устойчив к бензилпенициллину (10ЕА) и оксациллину (1 мкг), не проявляет лизоцимную активность. Штамм не продуцирует бета-экзотоксин. Дает положительную реакцию жгутикового антигена с типовой сывороткой Bacillus thuringiensis spp. morissoni H-8ab. 3. После стерилизации колбы с содержащейся в них средой охлаждали до комнатной температуры. Стерильность и рН контролировали путем стерильного отбора пробы среды. Колбы инокулировали внесением 1 мл посевного материала, приготовленного следующим образом: в стандартную пробирку с 5 мл стерильного физиологического раствора, помещали бактериальную культуру, собранную петлей с агаризованной среды «А». Оценку инсектицидной активности для личинок колорадского жука (Leptinotarsa decemlineata) первого-второго возраста проводили в лабораторных условиях. Для этого в чашки Петри разливали 1,5% голодный агар, на который помещали по 2 отчлененных листа картофеля (площадью по 4-5 см2). На каждый лист наносили по 40 мкл споро-кристаллической суспензии штамма Bacillus thuringiensis ssp.morissoni ВКПМ В-14326 с титром 1,0 х107 спор/мл, споро-кристаллической суспензии штамма Bacillus thuringiensis ssp. thuringiensis с титром 5,0×108 спор/мл, в качестве контроля использовали физиологический раствор и препарат Битоксибациллин.The assessment of insecticidal activity for larvae of the Colorado potato beetle ( Leptinotarsa decemlineata ) of the first and second instars was carried out in laboratory conditions. Лабораторные эксперименты по оценке акарицидного действия споро-кристаллической суспензии заявляемого штамма ВКПМ B-14326 проводили чашечным методом, включающим нанесение рабочей суспензии на кормовой субстрат с дальнейшей подсадкой тестируемых клещей Tetranychus urticae. На кормовое растение наносили по 100 мкл споро-кристаллической суспензии штамма Bacillus thuringiensis ssp.morissoni ВКПМ В-14326 с титром 1,0×108 спор/мл. Для опыта отбирали одновозрастных клещей, варианты лабораторных экспериментов закладывали в 3-5-ти кратной повторности. Количество тестируемых паутинных клещей - от 25 до 95 особей на одну повторность. Учеты количества выживших клещей проводили на 3-й и 5-й день после начала опыта.
