Заявляемое соединение 1 синтезируют обработкой 5-метил-2-(2'-аминоэтил)фурана N-Boc-валином при кипячении в абсолютном дихлорэтане в присутствии 1-этил-3-[3-(диметиламино)пропил]карбодиимида и N-метилморфолина. Реакционную смесь кипятят с обратным холодильником в течение 12 часов. Затем растворитель упаривают досуха при пониженном давлении. К раствору неочищенного амида в ледяной 22.5 мл АсОН добавляют 2.5 мл конц. HCl. Реакционную смесь перемешивают при комнатной температуре в течение 24 часов. Затем к реакционной смеси порционно присыпают 4.4 г Na2CO3 и кипятят с обратным холодильником в течение 30 минут. Далее растворитель упаривают досуха при пониженном давлении. Остаток очищают методом колоночной хроматографии на силикагеле, используя в качестве элюента смесь петролейный эфир/этилацетат (9:1), очищенный продукт 1 перекристаллизовывают из смеси петролейный эфир/этилацетат. Выход 57%. Тпл=130-131°С. Найдено, %: С, 69.69; Н, 8.69; N, 13.54. C12H18N2O. Вычислено, %: С, 69.87; Н, 8.80; N, 13.58. ИК спектр, (KBr), vmax, см-1: 3296, 3213, 1666, 1296, 1140. Спектр ЯМР 1Н, (400 МГц, ДМСО-d6), d, м.д.: 7.52 (уш. с, 1Н), 5.73 (д, J=3.0 Гц, 1Н), 5.66 (д, J=3.0 Гц, 1Н), 4.08 (д, J=10.8 Гц, 1Н), 3.39-3.37 (м, 1Н), 3.14-3.06 (м, 2Н), 2.92-2.83 (м, 1Н), 2.46-2.37 (м, 1Н), 2.12 (с, 3Н), 1.05 (д, J=6.4 Гц, 3Н), 0.55 (д, J=6.4 Гц, 3Н). Спектр ЯМР 13С (100 МГц, ДМСО-d6), d, м.д.: 169.9, 129.8, 128.7, 105.8, 105.6, 67.7, 42.9, 32.2, 25.7, 20.1, 19.0, 12.5; m/z (%) (ЭУ, 70 эВ) 206 (50) [М+], 191 (20), 163 (45), 134 (100), 120 (55), 107 (48), 94 (78), 43 (88). Для характеристики антибактериальной активности использовали стандартные параметры: минимальная подавляющая концентрация (МПК), которую определяли модифицированным методом двукратных серийных разведений (МУК 4.2.1890-04 Определение чувствительности микроорганизмов к антибактериальным препаратам) и минимальная бактерицидная концентрация (МБК) [Медицинские лабораторные технологии: Руководство по клинической лабораторной диагностике, п/р Каприщенко, 2013, Т. 2, стр. 407]. Тесты проводили с использованием культур модельных микроорганизмов Escherichia coli МС4100 и Staphylococcus epidermidis АТСС29887 на питательной среде Луриа-Бертани в 96-луночных полистироловых планшетах. Конечная концентрация микроорганизмов в лунках составляла 5*105 КОЕ/мл. Культивирование проводили при 37°С без перемешивания. Определение МПК и высевы для определения МБК производили через 24 ч. На первом этапе исследуемое соединение растворяли в диметилсульфоксиде (ДМСО), дальнейшие разведения производили в метаноле. В качестве начальных в тестах использовали концентрации, которые при внесении в питательную среду Луриа-Бертани не образовывали осадка (от 0.2 до 4 мг/мл).Example 2. Соединение 1 вводили внутрибрюшинно белым мышам массой 16-18 г в виде взвеси в 2% крахмальной слизи и наблюдали за поведением и гибелью животных в течение 10 суток.
