Данный способ основан на культивировании исследуемого материала на мясопептонном агаре в течение 24 ч при температуре 37°С, а затем выросшую культуру бактерий выдерживают при температуре +4°С в течение 48 ч.м. Недостатком прототипа является то, что культивирование на мясопептонном агаре некультивируемых форм стафилококков не представляется возможным. Затем способ осуществляют в два этапа. Первый этап – предварительный. На данном этапе в стандартную стерильную пластиковую пробирку объёмом 10,0 мл наливают в количестве 2,0 мл 1-4% глюкозный бульон, приготовленный на основе питательной среды ГРМ-бульона. ГРМ-бульон предназначен для культивирования различных микроорганизмов, неприхотливых по своим питательным потребностям, при проведении исследований в санитарной и клинической микробиологии. «ГРМ-бульон» представляет собой мелкодисперсный гигроскопичный порошок светло-желтого цвета. Производят «ГРМ-бульон» в ФБУН «ГНЦПМБ», Россия, Московская обл., Серпуховской р-он, п. Оболенск, в двух вариантах (из расчета, г/л): 1) панкреатический гидролизат рыбной муки 18,0 и натрия хлорид 2,0; 2) панкреатический гидролизат рыбной муки 8,0, пептон сухой ферментативный 8,0 и натрия хлорид 4,0. Затем окунают зонд-тампон с биоматериалом и оставляют в термостате при температуре 37˚С на 18-24 часа для инкубации.
