Была проведена оценка биосовместимости и цитотоксичности гидрогеля, полученного из заявляемой композиции. При этом использовали кремнийхитозансодержащей глицерогидрогель, полученный по примерам № 3, № 4, № 12. Гидрогель диспергировали в бидистиллированной воде в разведении 1:10 – 1:15, помещали в стерильную чашку Петри, заливали ростовой средой DMEM с добавлением 10% эмбриональной сыворотки крупного рогатого скота и вносили суспензию клеток дермальных фибробластов в концентрации не меньше 106 кл./мл. Культивирование осуществляли в СО2-инкубаторе в атмосфере 5% СО2 при Т = 37°С. Жизнеспособность клеток оценивали на флуоресцентном микроскопе «МикМед-2» (Россия), окрашивание проводили акридиновым оранжевым и этидиумом бромидом. Наблюдение за адгезией и пролиферацией клеток проводили на сканирующем электронном микроскопе Tescan MIRA LMU (Чехия).The biocompatibility and cytotoxicity of the hydrogel obtained from the claimed composition was evaluated.
