Abstract
Description
The invention relates to medicine and to the pharmaceutical industry and relates to a method for producing funds with hepatoprotective activity.
A known method of obtaining funds with hepatoprotective activity (US Pat. No. 2259208, publ. 2005.08.27). The essence of the method consists in the extraction of rhizomes and roots of meadowsweet with six-petal water when heated at a ratio of raw materials: extractant 1: 10-15 for 30 minutes, filtration, evaporation to 1 / 3-1 / 5 of the volume and purification by cooling the evaporated concentrate, precipitation from it the target product, adding a triple volume of 96% ethanol, sedimenting, filtering the precipitate, washing it successively with 96% ethanol and ether. However, the thermal effect (evaporation of extraction) is the cause of additional energy costs, and may also have an undesirable effect on the safety of biologically active substances (BAS). The use of ether as a solvent is toxic to personnel, flammable and non-environmental.
There is also known a method of hepatoprotective and hypocholesterolemic drugs (US Pat. No. 2270686, publ. 2006.02.27). A method where ragweed ragweed collected in the developmental phase before flowering is used as a feedstock is characterized in that the feedstock is crushed and extracted three times with ethanol under certain conditions, the extracts are combined, filtered and evaporated in vacuo. The water-alcohol bottoms are mixed with hot water and heated until ethanol is completely removed, then they are purified from lipophilic compounds. The obtained purified aqueous extraction of the sum of polyphenolic compounds is divided into two equal portions, one is evaporated to dryness and the sum of polyphenolic compounds is obtained -
Closest to the claimed method is a method of obtaining funds with hepatoprotective activity "Lohein", by extraction of the dried aerial parts of the hodgepodge, crushed to 3-10 mm, 25% ethyl alcohol at room temperature by percolation in a battery of 6 percolators with a ratio of raw materials: finished product 1: 2.6. This method is adopted as a prototype (US Pat. No. 2250777, publ. 2004.11.10). However, the extraction efficiency does not allow to fully transfer to the preparation some groups of biologically active substances with high hepatoprotective activity.
The objective of the invention is to increase the efficiency of the method.
The technical result - increasing the yield of various groups of biologically active substances - is achieved by sequentially treating the dried aboveground part of the hill hodgepodge, crushed to 1-3 mm, first with 70% ethyl alcohol and then purified water at room temperature by the method of three-step countercurrent extraction with the ratio of raw material: finished product 1 : 2.6 at each stage for 6 hours, followed by a combination of extracts. The use of two extractants makes it possible to more fully extract all biologically active substances of the aerial part of the hillwort (water-soluble complex of biologically active substances, flavonoids, etc.)
Example 1 (prototype). The dried aerial part of the hillock hodgepodge is ground to 3-10 mm and extracted with 25% ethyl alcohol by the method of multistage countercurrent extraction on a battery of 6 percolators. Infusion time - 6 hours. The first discharge of the finished product is carried out after 36 hours in an amount corresponding to the amount 2.6 times the mass of raw materials loaded in 1 percolator (1: 2.6). After settling at 7-8 ° C for 2 days, the extract is filtered. Of 390 g (65 g for each degree of extraction) of dry grass, 1014 ml of a liquid extract is obtained with an extractive content of 3.69% (total yield 37.42 g), which corresponds to 233 therapeutic doses.
Example 2 (the proposed method). The dried aerial part of the hillock hodgepodge is ground to 1-3 mm and subsequently extracted three times in each percolator with 70% ethyl alcohol, then purified water. Infusion time - 6 hours. Primary alcohol and secondary aqueous extracts are collected at each stage in an amount 2.6 times the mass of raw materials loaded into 1 percolator (1: 2.6), and only from the “head” for each extractor percolators.
According to the scheme (see drawing), the first discharge of the finished product (GP) (primary alcohol extraction) is carried out after 18 hours in an amount corresponding to the amount 2.6 times the mass of raw materials loaded in 1 percolator (1: 2.6). At the same stage, the 4th reserve percolator is brought into operation in the ethanol battery, a “water” battery is formed in parallel: purified water is supplied to the raw materials in the 1st percolator, freed from ethanol extraction.
After 6 hours (the 5th stage), the second alcohol discharge of the finished product is carried out from the 4th percolator, the 5th reserve is put into operation, the raw material in which is processed by extraction from the 4th, and the 2nd is transferred to the “water” battery.
At the 6th stage, the third portion of the ethanol finished product is drained from the 5th percolator, the 6th reserve is put into operation, where the raw materials are processed by extraction from the 5th, and the 3rd is included in the “water” battery.
(1:2,6).At the 7th stage, the finished product is obtained from the 6th percolator (the ethanol battery is transferred to the stop period). At the same stage, from the 3rd percolator, the first discharge of the finished product from the “water” battery (secondary water extraction) is carried out in an amount corresponding to the amount 2.6 times the mass of raw materials loaded in 1 percolator (1: 2.6) .
After this, the 4th percolator is transferred from the ethanol battery to the “water” one, the 1st percolator is removed from the battery and sent for reboot. Further according to the scheme.
The primary alcohol and secondary aqueous extracts are combined, defended for 3 days at a temperature of 7-8 ° C and subjected to filtration. From 390 g of dry raw materials, 2028 ml of a liquid extract is obtained with an extractive content of 1.86% (total yield 37.72 g), which corresponds to 235 therapeutic doses.
Метод-прототип и предлагаемый способ экстракции статистически равнозначно обеспечивают извлечение глицинбетаина (ГБ): не менее 80% от их количества в сырье (табл.1).A comprehensive analysis of the quality of the finished product obtained by the prototype method and the proposed method with the same consumption of raw materials shows that both methods equally deplete raw materials for extractive substances (EV) (at least 90%). The prototype method and the proposed extraction method statistically equivalently ensure the extraction of glycine betaine (GB): at least 80% of their amount in raw materials (table 1).
The content of flavonoids in the extract obtained by the proposed method is 2.3 times higher than the corresponding index of the extract produced by the prototype method, providing a yield of flavonoids of no more than 30%. The quality of the extract according to flavonoids, indicating the specific content of the latter in the total mass of extractive substances, is twice as high for the product obtained by the proposed method. The method of percolation using 25% ethyl alcohol, providing a yield similar to the proposed method, the total mass of the marketable product (the sum of extractive substances), does not allow to obtain an extract with the same high content of flavonoids and the corresponding level of good quality for this group of biologically active substances.
У экстракта, полученного предлагаемым методом экстрагирования, выявлено наличие желчегонной активности, отсутствующее у продукта, получаемого с использованием 25% спирта этилового (табл.2).The therapeutic adequacy of the product obtained by the new technology is confirmed experimentally. In acute liver damage with carbon tetrachloride, the extract obtained by the proposed method, more efficiently than the product made by the prototype method, reduced the total toxicity of carbon tetrachloride, slowed down the formation of lipid peroxidation products, and helped restore the basic functions of the liver. The extract obtained by the proposed extraction method revealed the presence of choleretic activity, which is absent in the product obtained using 25% ethyl alcohol (Table 2).
Thus, the proposed method for extracting the extract from the aerial part of the hillock hodgepodge is more effective, since it allows enriching the extraction with phenolic compounds with high hepatoprotective and choleretic activity while preserving the water-soluble complex of biologically active substances and, as a result, enhance the therapeutic effect and increase the pharmacotherapeutic activity of the drug in pathology the liver.
* - ЭВ - экстрактивные вещества; ГБ - глицинбетаин.Note to table 1 - The differences are significant (p <0.05) 1 - compared with the prototype method.
* - EV - extractive substances; GB is glycine betaine.
