Согласно методических указаний по определению качества питательных сред для выращивания холерного вибриона (Инструкция по бактериологическому контролю диагностических питательных сред для холерного вибриона), готовят культуру авирулентного штамма НАГ-вибриона Р-9741. Для чего культуру, хранящуюся на полужидком агаре, высевают в 17% пептонную воду или бульон Мартена (рН 7,6-7,8) и инкубируют при 37°С в течение 3-5 ч, после чего высевают на чашку с агаром Мартена или Хоттингера (рН 7,6-7,8). Через 18-20 ч роста с агара отбирают гладкие колонии НАГ-вибриона Р-9741 пересевают их на агаровые пластинки (2-пассаж). Культуру выращивают 3-6 ч при 37°С, после чего используют для контроля. Из суточной культуры готовили взвесь м.т., равной 5 ед. по оптическому стандарту мутности по ОСО ГИСК им. Л.А.Тарасовича. Затем серийными десятикратными разведениями в физиологическом растворе в объеме 4,5 мл доводили до содержания в 1 мл 100 и 10 м. к. Из данных разведении взвеси культур высевали по 0,1 мл стерильной бактериологической пипеткой по 3 флакона опытной и контрольной среды. В качестве последней используется заранее проверенная высококачественная 1% пептонная вода, приготовленная на дистиллированной воде. Посевы инкубируют при 37°С 6 ч, после чего из каждого флакона с поверхности высевают петлей № 5 по Чаплевскому на чашку Петри с заранее проверенной высококачественной плотной средой. Выращивание производят при 37°С. Питательная среда жидкая считается пригодной в том случае, если при посеве 100 м.к. тест-штамма в 100 мл через 6 ч выращивания и последующего высева на агаровые пластинки вырастают соответственно не менее 10 и 1 колоний.
