Abstract
Description
The invention relates to the medical and food industry, in particular to the production of pectin preparations used in the manufacture of food products, as well as prophylactic and therapeutic agents.
A known method of producing a food additive, including cellulose fiber and pectin, from waste plant materials / 1 /. In the known method, the deesterification of pectin is carried out using a chemical reagent saturated with a solution of lime, which makes it necessary to repeatedly wash the intermediate with distilled water. This makes the method time-consuming and inappropriate from the point of view of the ecology of production.
There is also a method of producing food additives / 2 /, which consists in the fact that the raw materials used are treated with amylolytic and proteolytic enzymes. This method allows to obtain a food supplement containing mainly cellulose fibers and highly esterified pectins with low radioprotective ability. This does not allow their use in the medical industry. The low water solubility of such an additive limits its use in the food industry.
The closest way to obtain pectin preparations to the proposed method is a method for producing pectin from industrial wastes, for example apple pomace, which involves the enzymatic treatment of plant materials with microbial enzymes, followed by filtration, concentration of the extract and precipitation of the product / 3 / - prototype. In this case, enzymatic preparations of cellulolytic action are used, which provide a slight increase in the yield of the final product due to hydrolysis of the plant cell wall and the isolation of the soluble form of pectin. However, such pectin is a highly esterified form of pectin and, therefore, has a weak radioprotective ability. This makes it impossible to use it in the medical industry.
The action of cellulolytic enzymes does not ensure the transfer of water-insoluble pectin to the extract and does not allow to obtain the maximum yield of the product. The cellulolytic preparations used also include endopoligalacturonase, an enzyme that catalyzes the hydrolysis of pectin to low molecular weight products, in particular galacturonic acid. It is impossible to preserve the natural gelling properties of the secreted polysaccharide, which limits its use in the food industry.
The aim of the present invention is to increase the yield of the product and improve its radioprotective properties.
This goal is achieved in that the method for the production of pectin preparations involves the enzymatic treatment of plant materials with microbial enzymes, followed by filtration, concentration of the extract, precipitation and drying of the product, and enzymatic processing of the raw materials using complex preparations, which include pectin lyase with activity from 10,000 to 40,000 units / g, pectin esterase with an activity of 5 to 120 u / g, hemicellulase with an activity of 10 to 100 u / g and amylase with an activity of 20 to 100 u / g.
In this case, before the introduction of the enzyme preparation, a preliminary swelling of the raw material in water is carried out at a temperature of 20-100 o C for 1-8 hours.
The dose of the complex enzyme preparation is 0.1-0.5% of the total weight of the raw material.
The enzymatic processing of plant materials is carried out at a temperature of 37 ± 3 o C, the natural pH of the raw materials and the weight ratio of raw materials and water in the range from 1: 5 to 1:20, depending on the type of raw material.
The duration of the enzymatic treatment of plant materials is 1-4 hours.
The products of the enzymatic treatment in the form of an extract are separated from the raw materials periodically after 0.5-2 hours.
Purification of raw materials is carried out in an alkaline environment during the enzymatic treatment.
1:1,5-1:5. Precipitation of pectin substances is carried out with ethanol in a volume ratio of pectin extract to ethanol as 1: 2-1: 4 or a calcium-containing product in a weight ratio of pectin to calcium-containing product as 1: 1.5-1: 5.
Drying of pectin concentrate is carried out by spraying at an initial process temperature of 100-130 o C and a final temperature of 60-65 o C or freeze drying at a temperature of 20-40 o C.
