Abstract
Description
The invention relates to veterinary and medical microbiology. The predominant area of use is the creation of listeriosis diagnostics.
An analogue of the invention is the publication of I.A. Bakulova, where it is stated: “As a result of numerous studies, the absence of any connection between the object from which the causative agent of listeriosis was isolated and its serological characteristics was revealed. In this regard, the separation of serological types of listeria into“ gnawing, “ruminant” and “equine ". Observations have also not been confirmed that listeria in the body of an animal can change its serological affiliation."
С. 1981). The prototype of the invention are the strains currently used for the production of listeriotic diagnosticums (FS Shulyak "Listeriosis". Sat. "Veterinary preparations", M. 1981).
In particular, to create diagnostic drugs that contribute to the intravital diagnosis of listeriosis using the CSC method, the antigen obtained from four strains from the VGNKI collection of veterinary preparations is used: Buryn, Ternopol N 17, N 1. These listeriose strains were isolated from pathological material from sheep, cow, rabbit, pig, studied and proposed to create diagnosticums in the late 50s.
The main disadvantage of the prototype is that the selection of these strains for listeriosis antigen was carried out primarily in accordance with the false theory existing at that time about the division of all listeriosis strains into animal types. Serological characteristics of the strains were taken into account secondarily. Therefore, currently used these 4 listeriosis strains are serologically typified only within serogroups I and II. Orientation to “animal” types did not require the use of strain selection for more specific parameters for listeriosis antigen, taking into account the combination of antigenic determinants characteristic of each serovar. This, in the first place, is due to the presence of false-specific results when using the DSC method for the diagnosis of listeriosis.
The second significant disadvantage of the listeriosis strains of the described prototype is that they are virulent strains. This greatly complicates the work on the production of listeriosis antigen, since the bacterial culture of these strains poses a threat to human health, which requires additional measures to comply with safety rules in working with them.
The aim of the invention is a strain of L. monocytogenes USHI-6 N 20 related to the serovariant 3C, contributing to an improved diagnosis of the disease caused by listeria.
The essence of the invention lies in the fact that the obtained strain of L. monocytogenes USHI-6 N 20 has the following morphological and biological characteristics.
Cultural properties. Bacterial culture of said strain grows well in a meat-peptone broth (BCH) at 30 ° C on the first day BCH slightly turbid, uniform turbidity, while shaking the tubes are observed so-called moire wave. After 2-4 days, the broth in the test tube brightens, and a mucous precipitate forms at the bottom of it, which, when shaken, rises with a pigtail. On meat-peptone gelatin (MJ) when sowing with an injection of Listeria culture, after two days they grow in the form of a bayonet only along the injection, gelatin liquefaction is not observed. In Kitt-Tarozzi broth, the growth of listeria is characterized by a very slight turbidity of the medium. On meat-peptone agar (MPA) in the first day of growth, small (0.2-0.5 mm in diameter), transparent in transmitted light, slightly convex colonies are found. After 2-3 days, the colonies increase in size (up to 1-3 mm in diameter), become flatter and acquire a grayish tint under incident light.
Morphology. Gram-positive bacilli, located singly and in pairs, were observed in smears of cultures of the listeriosis strain prepared with MPA and MPB stained by Gram.
Mobility. Daily broth cultures, cultured at 22 ° C Mobile.
Biochemical properties. The bacterial culture of this strain ferments to form an acid without gas: glucose, maltose, ramnose, salicin, fructose. Does not ferment: dulcite, inulin, mannitol, discolor litmus milk, indole does not form. On blood agar, after 3 days it causes hemolysis.
According to the results of staging a growth sample, strain USHI-6 N 20 is assigned to serogroup I.
1. Virulent properties. The culture strain USHI-6 N 20 does not cause death of rabbits when administered intraperitoneally at doses of 5 and 10 billion microbial cells, respectively. 2 out of 5 infected guinea pigs, 1 out of 5 infected white mice.
The proposed listeriosis strain has the following antigenic structure (see table).
The use of this strain in the production of listeriosis diagnosticums (in combination with other strains of listeria) allows you to cover the maximum possible range of antigenic determinants characteristic of this type of microorganism, which, in turn, increases the specificity of diagnostic drugs.
