Claims (30)

1. Тау-пептидная иммуногенная конструкция может быть представлена формулами:
(Th)m-(A)n-(фрагмент Тау)-X
или
(фрагмент Тау)-(A)n-(Th)m-X,
где Th представляет собой гетерологичный T-хелперный эпитоп;
А представляет собой гетерологичный спейсер;
(фрагмент Тау) представляет собой В-клеточный эпитоп, имеющий от около 15 до около 40 аминокислотных остатков из полноразмерного Тау-белка SEQ ID NO: 100;
X представляет собой α-COOH или α-CONH2 аминокислоты;
m составляет от 1 до около 4 и
n составляет от 0 до около 10.
2. Тау-пептидная иммуногенная конструкция по п. 1, где Тау-фрагмент выбран из группы, состоящей из SEQ ID NO: 1-21 и 101-124.
3. Тау-пептидная иммуногенная конструкция по любому из пп. 1 или 2, где Th-эпитоп выбран из группы, состоящей из SEQ ID NO: 22-50.
4. Тау-пептидная иммуногенная конструкция по п. 1, где пептидная иммуногенная конструкция выбрана из группы, состоящей из SEQ ID NO: 51-71 и 125-155.
5. Тау-пептидная иммуногенная конструкция, содержащая: В-клеточный эпитоп, содержащий от около 15 до около 40 аминокислотных остатков из полноразмерной последовательности Тау-белка из SEQ ID NO: 100; Т-хелперный эпитоп, содержащий аминокислотную последовательность, выбранную из группы, состоящей из SEQ ID NO: 22-50; и необязательный гетерологичный спейсер, выбранный из группы, состоящей из аминокислоты, Lys-, Gly-, Lys-Lys-Lys-, (α, ε-N)Lys и ε-N-Lys-Lys-Lys-Lys (SEQ ID NO: 102), где В-клеточный эпитоп ковалентно связан с Т-хелперным эпитопом напрямую или через необязательный гетерологичный спейсер.
6. Тау-пептидная иммуногенная конструкция по п. 5, где В-клеточный эпитоп выбран из группы, состоящей из SEQ ID NO: 1-21 и 101-124.
7. Тау-пептидная иммуногенная конструкция по п. 5, где Т-хелперный эпитоп выбран из группы, состоящей из SEQ ID NO: 22-50.
8. Тау-пептидная иммуногенная конструкция по п. 5, где необязательный гетерологичный спейсер представляет собой (α, ε-N)Lys или ε-N-Lys-Lys-Lys-Lys (SEQ ID NO: 102).
9. Тау-пептидная иммуногенная конструкция по п. 5, где Т-хелперный эпитоп ковалентно связан с N-концом В-клеточного эпитопа.
10. Тау-пептидная иммуногенная конструкция по п. 5, где Т-хелперный эпитоп ковалентно связан с N-концом В-клеточного эпитопа через необязательный гетерологичный спейсер.
11. Композиция, содержащая пептидную иммуногенную конструкцию по любому из пп. 1-4.
12. Фармацевтическая композиция, содержащая:
а. пептидную иммуногенную конструкцию по любому из пп. 1-4 и
b. фармацевтически приемлемый носитель для доставки и/или адъювант.
13. Фармацевтическая композиция по п. 12, где
а. Тау-пептидная иммуногенная конструкция выбрана из группы, состоящей из SEQ ID NO: 51-71 и 125-155; и
b. Тау-пептидная иммуногенная конструкция смешана с CpG-олигодезоксинуклеотидом (ODN) с образованием стабилизированного иммуностимулирующего комплекса.
14. Выделенное антитело или его эпитоп-связывающий фрагмент, которые специфически связываются с В-клеточным эпитопом Тау-пептидной иммуногенной конструкции по любому из пп. 1-10.
15. Выделенное антитело или его эпитоп-связывающий фрагмент по п. 14, связанные с Тау-пептидной иммуногенной конструкцией.
16. Выделенное антитело или его эпитоп-связывающий фрагмент, которые специфически связываются с В-клеточным эпитопом Тау-пептидной иммуногенной конструкции по любому из пп. 1-10.
17. Композиция, содержащая выделенное антитело или его эпитоп-связывающий фрагмент по любому из пп. 14-16.
