Inventia se refera la un procedeu pentru prepararea unui vehicul de clonare pentru transformarea unei gazde bacteriene pentru a deveni generator de polipeptida heterologa, procedeu ce include tratarea unei plasmide cu o enzima restrictiva, obtinerea zisei plasmide în prezenta de ADN dublu caternar aditional, în modul acesta fiind înmcorporata secventa aditionala în plasmida realizata, aceasta plasmida fiind capabila, dupa transfoemare în organismul gazda, sa fie amplificata prin multiplicare. Asezarea în plasmida a unui regulator omolog fata de bacteria gazda, în starea sa netransformata, plasînd în faza de citire cu regulatorul o inserttie de ADN, care codifica secventa de aminoacizi ai polipeptidei heterogene.The invention relates to a process for the preparation of a cloning vehicle for transforming a bacterial host to become a heterologous polypeptide generator, which process comprises treating a plasmid with a restriction enzyme, obtaining the plasmid in the presence of additional double-stranded DNA in this fashion adding the additional sequence to the resulting plasmid, this plasmid being able, after transfusion in the host organism, to be amplified by multiplication. Placing plasmid of a homologous regulator relative to the host bacterium in its unmodified state by placing in the read phase with the regulator a DNA insert which encodes the amino acid sequence of the heterogeneous polypeptide.
