Description
PROCESSO PARA A RECUPERAÇÃO DE ACTÍNIO-225 E BISMUTO-213
A presente invenção refere-se a um processo para a obtenção de actínio-225 e bismuto-213.
Os métodos radioterapêuticos para o tratamento local de perturbações provocadas por cancro (metástases) têm tido cada vez sempre mais importância com os avanços verificados na biologia molecular. Em princípio, inserem-se nuclídeos que originam radiação alfa, de curtos tempos de semitransformação, em anticorpos monoclonais que, quando colocados nos organismos dos pacientes, são preferivelmente absorvidos pelas células malignas e destroem estas últimas por acção de irradiação intensa com muito curtos tempos de actuação. Para o efeito, impõem-se requisitos especiais ao radionuclídeo, nomeadamente: deve poder ligar-se a um anticorpo apropriado para a conjugação, deve ter um período de semitransformação curto (da ordem de grandeza de algumas horas) e os seus produtos de desintegração radioactiva devem ser química e radiologicamente preponderantemente não prejudiciais.
De entre os candidatos possíveis como radionuclídeos deste tipo, têm um papel saliente o actínio-225 e o bismuto e, de entre os isótopos de bismuto salientam-se o isótopo Bi-212 (período de semitransformação 60,6 minutos) e o isótopo Bi-213 (período de semitransformação 47 minutos). A preparação de Bi-212 para fins medicinais foi descrito na revista Ing. J. Nucl. Med. Biol., 9 (1982), página 83. Este isótopo tem no entanto o inconveniente de originar, como produto da desintegração, um isótopo de tálio que origina radiações gama com a consequente exposição do paciente a radiação indesejável.
Isso não acontece realmente no caso do Bi-213 muito embora este isótopo não se encontre â disposição em quantidades suficientes e até agora não possa ser preparado em quantidades representativas a partir de U-233. Este isótopo do urânio U-233 forma, mediante várias fases de desintegração, actínio-225 e este, finalmente, Bi-213.
inconveniente decisivo desta cadeia de reacções de desintegração radioactiva reside na pequena quantidade de Th-229 que se obtém na desintegração de U-233. Como para se obter um abastecimento clínico significativo é necessária uma quantidade de Th-229 compreendida entre 10 e 100 gramas, seria preciso dispor de uma quantidade de U-233 da ordem de grandeza de uma tonelada durante um tempo de resistência longo compreendido entre 20 e 30 anos para se efectuar a separação da quantidade preten-3* dida. No entanto, em toda a terra nao existem quantidades de U-233 deste género.
objectivo da presente invenção é proporcionar um processo que, com um nível de gastos razoável, origine uma quantidade suficiente de actínio-225 e de bismuto-213 para a mencionada finalidade radioterapêutica.
processo de acordo com a presente invenção consiste em submeter rádio á irradiaçao com o fluxo de neutrões térmicos de um reactor nuclear, isolar do produto resultante da irradiação a parte que contém o tório mediante processos químicos e, a partir dela, separar novamente por processos químicos os radionuclídeos rádio-225 e actínio-225 formados permanentemente por desintegração de tório-229, os quais depois servem como matéria prima para os nuclídeos actínio-225 e bismuto-213.
Preferivelmente submete-se o rádio a um fluxo intenso IA 2 de neutrões térmicos igual a cerca de 5.101 /cmz segundo.
Seguidamente, esclarece-se mais pormenorizadamente a presente invenção por meio de um exemplo de uma forma de realização .
A Figura 1 representa a cadeia de desintegração natural que origina bismuto-213.
A Figura 2 representa a formação de tório-229 (em g de Th-229) em função do tempo (em anos) mediante irradiação de kg de rádio-226 com um fluxo de neutrões térmicos igual a . ι λ 9

A Figura 3 representa esquematicamente o processo completo de acordo com a presente invenção.
Na Figura 1 está representada esquematicamente a cadeia de desintegração atómica que, partindo do tório-229 permite chegar ao bismuto-213. Na extremidade superior desta cadeia de reaeções de desintegração atómica figura o isótopo U-233 com o número atómico 92 a partir do qual, por desintegração natural, se obtém o Th-229 pretendido, muito embora, como se mencionou antes, em quantidades insuficientes. A chave da preparação de Ac-225 e de Bi-213 em quantidades significativas reside na descoberta de que se pode obter o isótopo Th-229 de outra maneira diferente da desintegração do U-233.
Como mostra a Figura 3, como produto de partida do processo de acordo com a presente invenção, não se emprega o urânio-233 mas sim o rádio-226 com o número atómico 88. Para se obterem por exemplo 100 gramas, de Th-229, é preciso cerca de 1 kg de rádio-226. Irradia-se este produto de partida durante alguns anos, por exemplo, durante 3 anos, com um fluxo de neutrões térmicos o mais intensa possível por exemplo igual a

4,7 χ ΙΟ1 neutrões/eni segundo num reactor de elevado fluxo.
Isso é indicado por meio de uma seta (1) na Figura 3. Nessas condições, obtém-se entre outros isótopos o rádio-227 que, por emissão de radiação beta, se transforma em primeiro lugar em actínio-227. Por nova absorção de neutrões, obtém-se o actínio-228 que se desintegra com obtenção de Th-228 e com um tempo de semitransformação igual a 6,13 horas. Mediante nova absorção de iões forma-se finalmente o isótopo Th-229 pretendido. Por calculo, pode mostrar-se que, por irradiação da quantidade indicada de rádio com o referido fluxo de neutrões durante um intervalo de tempo de 3 anos, é possível esperar um rendimento em Th-229 da ordem de grandeza mencionada (veja-se a Figura 2).
produto final (2) da irradiação consiste por conseguinte numa mistura de rãdio-226 e dos citados produtos da série Ac-227, Th-228 e Th-229.
A partir desta mistura, isola-se em primeiro lugar quimicamente a fracção do tôrio (seta 3). Nesta, de acordo com o esquema de desintegração representado na Figura, por desintegração de Th-229, formam-se os radionuclídeos Ac-225 e Bi-213 pretendidos.
Seguidamente, a partir da fracção do tório, isola-se o Ac-225 e, por seu lado, separa-se o Bi-213 que aumentou a partir do Ac-225. No entanto, para se obter um rendimento máximo
em Ac-225 e Bi-213, separa-se nao só o Ac-225 mas também o nuclídeo de que deriva Ra-225 (em conjunto com Ra-224, que se obtém por desintegração de Th-228) (seta 4). Uma mistura deste tipo (5) ê também um fornecedor constante de Ac-225 e de Bi-213. Tanto o Ac-225 como também a mistura de isótopos de rádio podem ser expedidos sem problemas e também ser tratados no trabalho clínico como mistura de trabalho.
A partir desta mistura de neuclídeos separa-se o actínio-225 radioterapeuticamente activo o qual pode ser usado também para a obtenção do Bi-213 radioterapeuticamente importante, de acordo com a cadeia de desintegração representada na Figura 1.
Claims (2)
- REIVINDICAÇÕES1.- Processo para a recuperação de actínio-225 e de bismuto-213, caracterizado pelo facto de se irradiar rãdio-226 com um fluxo de neutrões térmicos de um reactor nuclear, de se isolar quimicamente a parte de tõrio do produto da irradiação, de se separar o rádio assim obtido e de se isolar do rádio o actínio-225 que penetrou, o qual eventualmente serve como produto de partida para o bismuto-213.
