Abstract
Przedmiotem zgłoszenia jest zmodyfikowane, wzmocnione białko zielonej fluorescencji TST-eGFP-MBL-Ni2+, które ma sekwencję aminokwasową białka eGFP, w której w miejsce sekwencji DGFP obejmującej reszty aminokwasowe od 190 do 193 wstawiono co najmniej jedną, 7 aminokwasową sekwencję SEQ ID NO.1, która wiąże jony niklu. Zgłoszenie ujawnia też konstrukt genetyczny o sekwencji SEQ ID NO.3, którego ekspresja daje produkt w postaci zmodyfikowanego białka TST-eGFP-MBL-Ni2+ o SEQ ID NO.2. Zgłoszenie obejmuje też sposób wytwarzania zmodyfikowanego białka TST-eGFP-MBL-Ni2+ i jego zastosowanie do oznaczania niklu w próbkach ciekłych.
