Abstract
Przedmiotem wynalazku jest sposób prowadzenia hodowli komórkowej chondrocytów in vitro do uzyskiwania materiału do leczenia ubytków chrząstki stawowej, polegający na tym, że pobraną tkankę chrzęstną szklistą tnie się na co najmniej dwa fragmenty tkanki, a następnie fragmenty te układa się na płytce hodowlanej obok siebie w medium hodowlanym zawierającym minimum 2 mM glutaminy, minimum 2% FBS, minimum 4.5 g/l glukozy. Kształt fragmentów tkanek dopasowuje się co najmniej na jednym odcinku krawędzi każdego fragmentu tak aby po ułożeniu fragmentów blisko siebie przestrzeń pomiędzy zbliżonymi co najmniej dwoma fragmentami tkanki była jak najmniejsza i tak aby wypełniana fragmentami tkanek przestrzeń płytki hodowlanej była jak najmniejsza. Tak dopasowuje się krawędzie aby w trakcie hodowli złączyć ze sobą fragmenty na co najmniej jednym odcinku stycznym. Do hodowli dodaje się zawiesiny chondrocytów i prowadzi się hodowlę komórkową fragmentów tkanki z chondrocytami. Medium hodowlane zmieniania się w ustalonym czasie, dodając do medium hodowlanego chondrocyty, przy czym hodowlę prowadzi się aż do uzyskania złączenia fragmentu tkanki na co najmniej części odcinka stycznego.
