Abstract

Foreliggende oppfinnelse gjelder genetisk manipulering av genet som koder for et overflateprotein fra streptokokker i gruppe B (GBS) betegnet som beta-antigenet. Beta-antigenet vites å bindes til Fc-området i IgA- immunoglobuliner på en ikke-immun måte. Den del av beta-antigengenet som koder for den IgA-bindende funksjon, er iden- tifisert og fjernet ved bruk av genut- formingsteknikker. Det nye polypeptid som kodes av det endrede beta-antigen- gen, bindes ikke til IgA, men vil im- munoreagere med monospesifikke anti- beta-antigen-antisera fremkalt mot beta-antigenproteinet av villtype. Denne ikke-IgA-bindende form av beta- antigenet kan benyttes som bestanddel i en human vaksine for beskyttelse mot GBS-infeksj oner.
