Description
Fremgangsmåte for opprensing av Faktor VIII samt peptider for slik bruk
Foreliggende oppfinnelse vedrører peptider basert på aminosyresekvensen av Faktor VIII som kan brukes til rensing av Faktor VIII.
Blodet fra personer med hemofili er defekt, med det resultat at det ikke koagulerer riktig. Den vanligste type hemofili stammer fra en defekt i Faktor VIII, idet Faktor VIII er et av en serie proteiner involvert i blodkoaguleringsprosessen. Eksogen Faktor VIII kan tilføres blødere med defekt Faktor VIII for å rette opp deres koaguleringsdefekt.
Forskjellige metoder har blir utviklet for rensing av Faktor VIII. Imidlertid består det et behov for forbedrede Faktor Vlll-rensningsmetoder for å få høyere utbytte av Faktor VIII. Foreliggende oppfinnelse oppnår dette ved å bruke peptider basert på aminosyresekvensen til Faktor VIII for rensing av Faktor VIII. Derved renses Faktor VIII

Foreliggende oppfinnelse angår peptider basert på aminosyresekvensen av.Faktor VIII som kan bli brukt for rensing av Faktor VIII. Disse peptider kan brukes for å fremskaffe monoklonale eller andre antistoffer for Faktor VIII-opprensning. Disse peptider kan også brukes for å eluere Faktor VIII fra monoklonale eller andre antistoffer for å rense Faktor VIII.
Spesielt foretrukket er et peptid med følgende sekvens:
TDSEMDWRFDDDNS
hvis aminosyresekvens er den til aminosyreresidiene 351-365 av Faktor Vlll-sekvensen.
Ytterligere foretrukket er et peptid med følgende sekvens:
EEIDYDDTISVEMKK
hvis aminosyresekvens er den til aminosyreresidiene 1660-1674 av Faktor Vlll-sekvensen.
Oppfinnelsen angår ytterligere et peptid omfattende et sekvensielt subsett på minst tre aminosyreresidier av et peptid basert på aminosyresevensen av Faktor VIII, som kan brukes for rensing av Faktor VIII.
Videre angår foreliggende oppfinnelse forbedrede metoder for fremstilling av renset Faktor VIII ved å bruke peptider basert på aminosyresekvensen av Faktor VIII og ethvert sekvensielt subsett på minst tre aminosyrer av peptidene.
Oppfinnelsen angår ytterligere metoder for fremstilling
ved rekombinante DNA- eller syntetiske peptidmetoder peptider basert på aminosyresekvensen av Faktor VIII og ethvert sekvensielt subsett på minst tre aminosyrer av peptidene.
Som angitt, omfatter foreliggende oppfinnelse peptider basert på aminosyresekvensen av Faktor VIII som kan brukes for rensing av Faktor VIII. Disse peptider kan bli brukt for å fremskaffe monoklonale eller andre antistoffer til Faktor Vlll-opprensing. Selv om beskrivelsen gir eksempler på foreliggende oppfinnelse, må disse ikke betraktes som spesifikt begrensende for oppfinnlsen, og slike variasjoner som vil ligge innenfor fagmannens kompetanse, anses å falle innenfor bredden av denne oppfinnelse.
Forskjellige peptider basert på aminosyresekvensen av Faktor
VIII og et sekvensielt subsett på minst tre aminosyreresidier av peptidene, er egnet for bruk i foreliggende oppfinnelse. For eksempel kan peptidene TDSEMDWRFDDDNS og EEIDYDDTISVEMKK og et sekvensielt subsett på minst tre aminosyreresidier av hvert av disse peptider bli brukt.
Peptider basert på aminosyresekvensen av Faktor VIII og et sekvensiekt subsett på minst tre aminosyreresidier av peptidet syntetiseres som beskrevet av Houghton et al., Proe. Nati. Acad. Sei. 82:5135 (1985).
I den velkjente fremgangsmåte for fastfasesyntese av et peptid settes det ønskede peptid sammen ved å starte fra et uoppløselig støttemateriale, såsom benzhydrylamin eller klormetylert resin (avledet fra fornettet polystyren og tilgjengelig fra forhandlere av kjemikalier). Aminosyren ved den karboksylterminale ende av det ønskede peptid som bærer beskyttende grupper på a-amino-nitrogenet og ethvert annet reaktivt sete, festes til resinet fra oppløsning ved å bruke kjente peptidkoblingsteknikker. Den beskyttende gruppe på oc-aminogruppen fjernes (ved å etterlate eventuelle andre beskyttende grupper intakte) og neste aminosyre av den ønskede sekvens (som bærer egnede beskyttende grupper) festes, osv. Når det ønskede peptid er blitt bygget opp fullstendig, blir det spaltet fra resinstøtten, alle beskyttende grupper fjernes, og peptidet gjenvinnes. Eksempler på egnede beskyttende grupper er: a-tert-butyloksykarbonyl for cx-aminogruppen; benzyl, 4-metoksybenzyl eller 4-metyl-benzyl for tiolgruppen av cystein, P-karboksylsyregruppen av asparaginsyre, T-karboksylsyregruppen av glutaminsyre og hydroksygruppene av serin, treonin og tyrosin; benzyloksy-karbonyl eller et 2-klor eller 3,4-dimetoksy-derivat derav for ringnitrogenene av histidin og tryptofan og epsilon-aminogruppen av lysin; p-nitrofenyl for amidnitrogenene av asparagin og glutamin og nitro eller tosyl for guanidin-gruppen av arginin.

Én og tre bokstavers aminosyreforkortelser
Den fullstendige aminosyresekvens av Faktor VIII har blitt bestemt (se Vehanr et al., Structure of Human Factor VIII, Nature 312: 337-342 (1984)). Forskjellige monoklonale antistoffer er velegnet for bruk i foreliggende oppfinnelse. For eksempel kan monoklonale antistoffer med egneskapen å binde Faktor VII bli brukt. Også monoklonale antistoffer fremstilt mot peptider basert på aminosyresekvensen av Faktor VIII og ethvert sekvensielt subsett på minst tre aminosyrer av peptidene kan bli brukt.
Det monoklonale antistoff C5 (tidligere beskrevet i Fulcher et al., Human Factor VIII Procoagulant Protein; Monoclonal Antibodies Define Precursor-Product Relationships and Functional Epitops, J. Clin. Invest., Vol. 76, side 117-124
(1985)) er et høyaffinitets antistoff som binder til Faktor VIII og derved nøytraliserer Faktor Vlll-aktivitet.
For å danne hybridomcellelinjen som danner monoklonalt antistoff C5 ble Balb/c mus immunisert ved intraperitoneal injeksjon av 1 pm av enten renset Faktor VIII:C eller trombinspaltet renset Faktor VIII:C i fullstendig Freund<1>s hjelpemediura. 10 og 50 pg forsterkninger i ufullstendig Freund<*>s hjelpemedium ble gitt med én-ukers intervaller. Den endelige 100 pg forsterkningsinjeksjon ble gitt uten hjelpemedium 3 dager før fusjon. Ved 4 uke ble musemilt-celler fusert med P3X63 muse plasmacytomceller som beskrevet i Brown et al., J. Biol. Chem. 255:4980-4983 (1980). Celle-dyrkning og produksjon av monoklonale antisto ffer i musebukvæske foregikk i hovedsak som hos Kiu et al., J. Immunol. 124:2728-2736 (1980). Antistoffene ble valgt ut ved å bruke et fastfase assay i Linbro-Titertek plater (Flow Laboratories, Inglewood, CA) og et enzymtilknyttet immunosorbent påvisningssystem som beskrevet i Engvall og Perlmann, Immunochemistry 8:871-874 (1971) ved å bruke et peroksydase antistoff-konjugat (Zymed Laboratories, Burlingame, CA). Platene ble belagt med 100 ng av enten renset Faktor VIII:C eller renset trombinspaltet Faktor VIII:C per brønn. Kloner ble også skjermet mot plater belagt med 100 ng av humant fibrinogen, plater belagt med 100 ng av humant fibronektin og plater belagt med 100 ng human von Willebrand-Faktor per brønn, hvor hvert protein er en potensiell kontaminant av immunogenet.
Kloner som var positive kun på Faktor VIII:C-belagte plater ble også undersøkt med hensyn til sin egenskap å inhibere plasma Faktor VII:C aktivitet, ved å bruke et modifisert Kasper-assay (se Kasper et al., Thromb. Diath. Haemorrh. 34:869-872 (1975)). Kultursupernatanter ble inkubert med et likt volum normalt sammeslått humant plasma som hadde blitt fortynnet 1:10 eller 1:20 i assaybuffer som beskrevet i Fulcher et al., Proe. Nati. Acad. Sei. 79:1648-1652 (1982), til hvilket det hadde blitt tilsatt 2% (vol/vol) av 0,5 M natriumcitrat, pH 6,5. Inkubering foregikk i 2 timer ved 37°C, hvorpå prøver ble undersøkt for Faktor VTII:C prokoagulantaktivitet. Kultursupernatanter fra klon C5 inhi-berte 75% av den gjenværende plasma Faktor VTII:C aktivitet, sammenlignet med kontroll- (ikke anti Faktor VIII:C super-natant) inkuberinger.
Kultursupernatanter ble undersøkt for immunoglobulinklasse og subklasse ved dobbel diffusjon i agarosegeler ved å bruke kommersielt tilgjengelige antisera (Miles Laboratories,Inc., Elkhart, IN). Protein A-sefarose-kromatografi (Pharmacia Fine Chemicals, Piscataway, NJ) av musebukvæske var som beskrevet av Ey et al., Immunochemistry 15:479-436 (1978). Rensede antistoffer ble konsentrert i en omrørt trykk-ultrafiltreringscelle (Amicon Corp., Danvers, MA) ved å bruke en YM-10 membran, posjonert og lagret nedfryst ved -70°C. Protein A-sefarose rensende monoklonalt antistoff ble undersøkt for sin egenskap å inhibere plasma Faktor VIII:C prokoagulantaktivitet ved å bruke det modifisert Kasperassay (se Kasper et al., Thromb.Diath. Haemorrh. 34:869-872 (1975)).
Peptidet EEIDYDDTISVEMKK ble også brukt for å fremskaffe monoklonale antistoffer i mus.
Immunogener for fremstilling av monoklonale antiostoffer mot peptidet EEIDYDDTISVEMKK ble fremskaffet ved å koble polypeptidfragmentet til et bæreprotein, kai-igle hemocyanin (KLH), ved på bruke glutaraldehyd. Like vektdeler av polypeptidet og bæreprotein ble oppløst i fosfatbufret saltvann (PBS; 1,6 g monobasisk natriumfosfat, 8,4 g dibasisk natriumfosfat, 61,4 g natriumklorid, destillert
vann til 7 liter, pH 7,2) til en endelig konsentrasjon på
2 mg bæreprotein pr. ml oppløsning. Fersk glutaraldehyd arbeidsoppløsning ble fremstilt ved å tilsette 200 ml stamløsning på 25% glutaraldehyd til 13 ml PBS, 124 pl fersk glutaraldehyd arbeidsoppløsning ble så tilsatt pr. 1,0 ml bærepeptidoppløsning. Den resulterende oppløsning ble omrørt over natten ved romtemperatur, dialysert i minst 6 timer mot destillert vann og frysetørket.
Balb/c mus ble immunisert ved intraperitoneal injeksjon med 100 pg immunogen i fullstendig Freund's hjelpemedium.
100 pg forsterkningsoppløsninger i ufullstendig Freund's hjelpemedium ble gitt med én ukes intervaller i 3 uker. En endelig 200 pg forsterkningsinjeksjon ble gitt uten hjelpemedium 3 dager før fusjonen. Ved ca. 8 uker ble milten fjernet og musemiltcellene ble fusert med P3X653-AG8.653 (musemyelom cellelinje).
P3X653-AG8.653 ble holdt (før fusjon) ved logaritmisk vekst-fase i et medium på 90% Dulbecco's modifisert Eagle's medium (høy glukose) og 10% føtalt bovint serum (FBS). De følgende anbefalte tilsetninger ble tilsatt til 475 ml av ovenfor nevnte medium: glutamin (100x) 5 ml, natriumpyruvat (100x) 5 ml, ikke-essensielle aminosyrer (100x) 5 ml, Pen-strep-fungizon (100x) 5 ml og 8-azaguanin 6,6 x 10~<3>M (50x) 10 ml. Milt- og myelomceller ble vasket grundig uten FBS i Dulbecco's modifisert Eagle's medium før fusjon. Cellene ble fusert med 1 ml 40% PEG 1500 i 1 minutt. Derpå ble cellene fortynnet 1:2 med vekstmedium i 1 minutt. Cellene ble fortynnet ytterligere 1:5 med vekstmedium i 2 minutter. Derpå ble cellene sentrifugert ved 900 rpm i 10 minutter. Supernatanten ble fjernet. Cellene ble valgt ut ved suspensjon i HAT medium og plassert i 9 6 brønners plater. (50x) 10 ml. 14 uker etter utvelgelse ble cellene holdt i vekstmedium-HT (seleksjonsmedium minus aminopterin). Sub-kloning ble utført ved begrensende fortynning. Brønner med vekst ble undersøkt med ELISA assay. Testplatene ble belagt med 100 ng pr. brønn av immunogen. 50 pl kultursupernatanter ble undersøkt. De brønner som inneholdt celler hvis supernatanter var positive for immunogen, ble dyrket ved 370C i 10% C02•
For produksjon av bukvæske ble musene på forhånd sensitivi-sert med 0,5 ml pristin minst 4 dager før celleinjeksjonen. Cellene ble injisert intraperitonealt (5xl0<6>pr. mus) i 0,5 ml medium uten FBS. Bukvæsken ble innsamlet når musene este. Det monoklonale antistoff J31B3 inneholdt i musebukvæske er av IgG-1 type.
Den følgende protein A sefarose rensning av monoklonalt antistoff J31B3 fra musebukvæske er en modifisert fremgangsmåte av den beskrevet i Ey et al., Immunochemistry 15; 429-436 (1978). De anvendte mengder var for en kolonne på 1 cm x 15 cm, som bandt ca. 15-30 mg IgG-1, men som tillot separasjon av ca. 50 mg IgG-1 fra ikke-IgG-proteiner. Kolonnen kan også binde 50 mg IgG2a. IgG2b binder også til kolonnen, men IgM, IgA og IgE binder ikke. 4-6 ml bukvæske ble sentrifugert ved 30.000 omdr/min i 45 minutter. Lipidene ble fjernet fra toppen. Tilsetning av 2 0% sukrose vekt/volum til bukvæsken hjalp til med fjerningen av lipidene. Bukvæsken ble fortynnet til 25-30 ml med 140 mM Na3P04buffer, pH=8, inneholdende 0,02% NaN3. Bukvæsken ble fortynnet for å forhindre innvirkningen av kloridioner på bindingen av IgG. Ca. 2 g protein A sefarose (Sigma) ble svellet i 10 mM fosfatbufret saltvann med 0,02% NaN3og pakket i en kolonne med diameter 1 cm. Kolonnen ble ekvilibrert i 140 mM Na3P04-buffer med 0,02% NaN3. Kolonnen ble påsatt fortynnet bukvæske ved 0,06-0,08 ml/min eller mindre. Kolonnen fikk sette seg ved 4°C over natten etter pålasting for å øke bindingen av IgG. Kolonnen ble vasket med buffere ved 0,6 -0,8 ml/min i følgende rekkefølge: 1) 140 mM Na3P04, pH 8,0, 2) 0,IM Na sitrat - sitronsyre, pH 6,0 (IgG-1 eluert), 3) 0,1 M Na sitrat-sitronsyre, pH 5,0 - IgG2a eluert og en liten prosentdel av det gjenværende IgG-1, 4) 0,1 M Na-sitrat - sitronsyre, pH 4,0 (liten prosentdel av gjenværende IgG2a eluert) og 5) 0,IM Na-sitrat-sitronsyre, pH 3,0 (IgG2b eluert). Så snart kolonnen var vasket med pH 3,0-buffer, ble den vasket med 140 mM Na3P04-buffer, pH 8,0 + 0,02% NaN3, inntil pH av utførselen var 8,0. Kolonnen ble lagret ved 4°C. Under vaskingen av kolonnen ble ca. 5 ml fraksjoner samlet opp. Til enhver fraksjon med pH 5,0 ble 1 ml IM tris HC1, pH 9,0, tilsatt.
Egenskapen av peptider basert på aminosyresekvensen av Faktor VIII og et sekvensielt subsett på minst tre aminosyrer av peptidene til å innvirke på bindingen av Faktor VIII til monoklonale antistoffer som har egenskapen å binde Faktor VIII, kan bli brukt ved rensing av Faktor VIII. Således ble egenskapen av peptid TDSEMDWRFDDDNS til å innvirke på bindingen av monoklonalt antistoff C5 til Faktor VIII vist i et assay hvor TDSEMSWRFDDDNS blokkerte C5 fra å nøytralisere Faktor Vlll-aktivitet.
En oppløsning av TDSEMDWRFDDDNS (22,5 pl) oppløst i barbital saltbuffer (pH 7,56) og 22,5 pl barbital saltvann ble blandet med 5 pl (totalt 10 pg) av C5 i barbital saltvann og inkubert over natten ved romtemperatur. Som kontroller ble barbital saltvann eller andre peptider substituert for TDSEMDWRFDDDNS, andre peptider ble substituert for de 22,5 pl av barbital saltvann, eller 5 pl barbital saltvann ble substituert for C5. Etter inkubering over natten ble 50 pl normalt plasma inneholdende Faktor VIII tilsatt TDSEMDWRFDDDNS-C5 blandingen og inkubert i 2 timer ved 37°C. Gjenværende Faktor Vlll-aktivitet i det normale plasma ble så målt i et aktivert partielt trom- boplastin tidsassay ved å bruke en MLA electra 700 automa-tisk koaguleringstids-måler. I dette assay gjelder det at jo kortere koaguleringstid, desto mer Faktor VIII er til stede i forsøksblandingen.
I et typisk forsøk resulterte inkubering av barbital saltvann (50 pl) med plasma (50 pl) i en koaguleringstid på 66,6 og 65,6 sekunder i duplikate assay. Dette representerer mengden av Faktor Vlll-aktivitet som er tilbake dersom intet Faktor VIII inaktiveres av C5. Dersom C5 og barbital saltvann ble inkubert med plasma, ble de duplikate koagule-ringstider 83,4 og 84,0 sekunder. Dette representerer inaktivering av Faktor VIII av C5. Imidlertid, dersom C5 først ble inkubert med en blanding inneholdende 22,5 pl av ImM TDSEMDWRFDDDNS, ble koaguleringstiden redusert til 71,1 sekunder, noe som indikerte delvis blokkering av C5.
Dersom konsentrasjonen av TDSEMDWRFDDDNS ble øket til 6 mM, ble koaguleringstiden minket til 67,1 sekunder, noe som indikerer nesten fullstendig nøytarlisering av C5.
De følgende eksempler er ytterligere illustratoriske for-foreliggende oppfinnelse. Disse eksempler er ikke tenkt å begrense omfanget av foreliggende oppfinnnelse, men gir bedre forståelse av oppfinnelsen.
Eksempel 1
Peptidet TDSEMDWRFDDDNS kan brukes ved rensing av Faktor VIII. Monoklonalt antistoff C5 eller ethvert antistoff som binder til Faktor VIII og hvor bindigen til Faktor VIII kan blokkeres med peptidet TDSEMDWRFDDDNS eller et subsett av dette, forbindes til et støttemateriale. Typisk er dette et fastfase støttemateriale såsom agarose, men andre støttema-terialer kan bli brukt effektivt. En kolonne forberedes fra antistoff-støttemateriale og en kilde for Faktor VIII passeres gjennom denne. Kilden kan være plasma, et plasmakonsentrat eller Faktor VIII, dannet ved rekombinante DNA-teknikker. Antistoff-kolonnen vaskes derpå og etterlater Faktor VIII bundet til antistoffkolonnen. Peptidet TDSEMDWRFDDNS blir så påført kolonnen. Dette vil erstatte den bundne Faktor VIII og således eluere Faktor VIII i høyt renset tilstand.
Eksempel 2
Monoklonalt antistoff J31B3 fremskaffet mot peptidet EEIDYDDTISVEMKK ble koblet til Sepharose 4B kuler ved cyanogen bromidteknikken.
Cyanogenbromid-aktivert Sepharose 4B (Sigma) ble svellet i 1 mM HC1 i 15 minutter og derpå vasket med 300 ml 1 mM HC1. Etter en kort vask med koblingsbuffer (0,2M natriumbikar-bonat, 0,5 M natriumklorid, pH 9,0), ble ca. 1 ml av resinet blandet med 5,4 mg renset J31B3 monoklonalt antistoff i koblingsbuffer. Resinet og antistoffet ble blandet sammen over natten ved 4°C på en rotator. Kobling ble bestemt ved sammenligning mellom absorbsjonen av pre og post koblingssupernatanter ved 2 80 nm. Det koblede resin ble så blandet på lignende måte i 2 timer i koblingsbuffer inneholdende 0,2 M glysin for å blokkere alle gjenværende aktive grupper. Det koblede resin ble så vasket 5 ganger vekselvis med koblingsbuffer og acetatbuffer (0,1 M natriumacetat, 0,5 M natriumklorid, pH 4) for å fjerne alle ikke kovalent bundne antistoff. Det koblede resin ble så pre-eluert med 1 ml 3M natriumtiocyanat (i 0,05 M natrium- acetatbuffer, pH 5,5, inneholdende 0,1 M lysin). Denne kaotropiske oppløsning sikret ytterligere fjerning av ethvert ikke kovalent bundet antistoff. Det koblede resin ble så vasket og lagret ved 4°C i imidazolbufret saltvann (0,02 M imidazol, 0,15 M natriumklorid, 0,02% natriumazid, pH 7,0).
En 1,1 milliliters kolonne med J31B3-Sepharose ble så dannet og vasket med imidazolbufret saltvann. 2,5 ml av en opp-løsning inneholdende 0,9 enheter/ml Faktor VIII, renset i henhold til teknikken beskrevet i US patent 32011, ble så tilført. Ingen Faktor Vlll-aktivitet ble funnet i bufferen som passerer gjennom kolonnen, noe som indikerer at all Faktor VIII hadde blitt bundet til kolonnen. Kolonnen ble så vasket med imidazolbufret saltvann og imidazolbufret saltvann inneholdende 1% bovin serum albumin. Faktor VIII ble så eluert med en oppløsning på 0,25 molar kalsiumklorid i imidazolbufret saltvann inneholdende 1% bovin serum albumin. Faktor VIII ble eluert i to 500 pl fraksjoner, henholdsvis med en konsentrasjon på 2,24 enheter/ml og 0,57 enheter/ml. Således ble Faktor VIII bundet av kolonnen og eluert i en mer konsentrert tilstand enn når den ble påsatt. Gjenvinningen var 62% av Faktor VIII satt på kolonnen.
Eksemplet ovenfor viser at et monoklonalt antistoff fremskaffet mot peptided EEIDYDDTISVEMKK kunne bindes til
en fast matrise og i denne tilstand binde Faktor VIII fra en oppløsning. Videre kunne Faktor VIII elueres fra antistoffet med 0,25 molar kalsiumklorid, et middel som er tilstrek-kelig mildt tillot 62% av aktiviteten ble gjenvunnet.
Eksempel 3
Peptidet EEIDYDDTISVEMKK kan brukes ved rensing av Faktor VIII. Monoklonalt antistoff J31B3 eller ethvert antistoff fremskaffet mot peptidet med aminosyresekvensen EEIDYDDTIISVEMKK og som binder Faktor VIII, og som ved binding av Faktor VIII kan blokkeres med peptidet EEIDCYDDTISVEMKK kobles til et støttemateriale. Typisk er dette et fastfase støttemateriale, såsom agarose, men andre støttematerialer kan bli brukt effektivt. En kolonne fremstilles fra antistoff-støttematerialet og en kilde for Faktor VIII passeres gjennom denne. Denne kilde kan være plasma, et plasmakonsentrat eller Faktor VIII dannet ved rekombinante DNA-teknikker. Antistoffkolonnen blir så vasket og etterlater Faktor VIII bundet til antistoffkolonnen. Peptidet EEIDYDDTISVEMKK blir så påsatt kolonnen. Dette vil erstatte den bundne Faktor VIII og således eluere denne i høyt renset tilstand.
Claims (9)
1. Peptid basert på aminosyresekvensen av Faktor VIII hvilket peptid kan benyttes for opprensking av Faktor VIII, eventuelt ved å fremskaffe monoklonale eller polyklonale antistoff mot Faktor VIII, og som kan benyttes ved eluering av Faktor VIII fra slike antistoff for opprensking av Faktor VIII, karakterisert ved at peptidet omfatter følgende aminosyresekvens
TDSEMDWRFDDDNS
tilsvarende aminosyreresidiene 351-365 av Faktor VIII-sekvensen, eller
EEIDYDDTISVEMKK
tilsvarende aminosyreresidiene 1660-1674 av Faktor VIII-sekvensen.
2. Peptid ifølge krav 1, karakterisert ved at det omfatter et sekvensielt subset på minst tre aminosyreresidier av Faktor Vlll-sekvensen.
3. Peptid ifølge krav 1 eller 2, karakterisert ved at det består av et sekvensielt subset på tre aminosyreresidier av Faktor Vlll-sekvensen.
4. Fremgangsmåte ved fremstilling av Faktor VIII, karakterisert ved at den omfatter:
(a) å absorbere Faktor VIII fra et rekombinant dannet plasma eller kommersielt tilgjengelig konsentrat på partikler bundet til et monoklonalt antistoff rettet mot et peptid basert på aminosyresekvensen av Faktor VIII eller ethvert sekvensielt subset på minst tre aminosyrer av et slikt peptid;
(b) å eluere det absorberte Faktor VIII; og
(c) gjenvinne det rensede Faktor VIII.
5. Fremgangsmåte ifølge krav 4, karakterisert ved at det monoklonale antistoffet er rettet mot et peptid med aminosyresekvens
TDSEMDWRFDDDNS
tilsvarende aminosyresekvensen av aminosyreresidiene 351 - 365 av Faktor VIII eller ethvert sekvensielt subset på minst tre aminosyrer derav, eller mot et peptid med aminosyresekvens
EEIDYDDTISVEMKK
tilsvarende aminosyresekvensen av aminosyreresidiene 1660 - 1674 av Faktor VIII eller ethvert sekvensielt subset på minst tre aminosyrer derav.
6. Fremgangsmåte ifølge krav 4 eller 5, karakterisert ved at trinn (b) omfatter å eluere absorbert Faktor VIII med et peptid basert på aminosyresekvensen av Faktor VIII eller ethvert sekvensielt subset derav på minst tre aminosyreresidier.
7. Fremgangsmåte ifølge krav 4-6, karakterisert ved at trinn (b) omfatter eluering med en saltoppløsning, fortrinnsvis en oppløsning av kalsiumklorid .
8. Frem <g> angsmåte ifølge krav 7, karakterisert ved at kalsiukloridopplø sningen ligger innenfor konsentrasjonsområdet 0,25M til 0,5M.
9. Fremgangsmåte ifølge krav 4-8, karakterisert ved at de avsorberende partikler i trinn (a) er av agarose.
