Description
Foreliggende oppfinnelse angår en gruppe av 2(1H)-kinolinoner med en tetrazolsubstituent ved 3- eller 4-stillingen. Nærmere bestemt angår oppfinnelsen forbindelser av følgende generelle formel:
hvori A er -CH= eller -N=; R er H eller alkyl med 1-4 C-atomer; n er 0, 1 eller 2; X er H, alkyl med 1-4 C-atomer;alkoxy med 1-4 C-atomer, halogen, methylmercapto, methylsulfonyl, eller hvor to X'er kan være kombinert som methylendioxy; forutsatt at når X er methylmercapto eller methylsulfonyl, må n være 1; og farmasøytisk akseptable salter derav.
Substituenten med den frie valens som inntrer i ringen mellom stillingene merket som 3 og 4, kan bare være bundet til én av disse to stillinger. X-substituenten kan bare være bundet til tilgjengelige 6-, 7- og/eller 8-stillinger på venstre side av ringen i den ovenfor viste struktur. Halogen er fluor, klor eller brom. Eksempler på alkylgrupper er methyl, ethyl, propyl eller isopropyl; og eksempler på alkoxygrupper er methoxy,' ethoxy, propoxy, isopropoxy eller butoxy.
Foretrukne forbindelser er de hvori tetrazolsubsti-tuenten er bundet til 3-stillingen av kinolinonet.
Ekvivalent med hensyn til formålet med oppfinnelsen er de farmasøytisk akseptable salter og også hydratene av forbindelsene og deres salter. Uttrykket "farmasøytisk aksep-tabelt salt" som anvendt her, er beregnet på å innbefatte ikke-toksiske kationsalter slik som alkalimetallsaltene, f.eks. natrium og kalium; jordalkalimetallsalter slik som calcium, magnesium eller barium; salter med ammoniakk, og salter med organiske baser, f.eks. aminer slik som triethyl-amin, n-propylamin og tri-n-butylamin.
Forbindelsene fremstilles ved omsetning av natriumazid og ammoniumklorid med et cyanokinolinon av formel:
hvori n, A, R og X er som ovenfor definert. Reaksjonen ut-føres under oppvarming i et inert løsningsmiddel slik som dimethylformamid. Natriumazid er det foretrukne azid ved reaksjonen selv om andre alkalimetallazider kan anvendes.
Det nødvendige utgangsmateriale erholdes fra et egnet substituert acetanilid etter to forskjellige metoder. Ved den første metode oppvarmes acetanilidet med dimethylformamid og fosforoxyklorid under dannelse av et 2-klor-3—kinolin-carboxaldehyd. Denne reaksjon og variasjoner derav er diskutert i detalj av Meth-Cohn et al., J. Chem. Soc., Perkin Trans. 1, 1981, 1520. Carboxaldehydet behandles deretter med hydroxylamin-hydroklorid, maursyre og natriumformiat under dannelse av det tilsvarende 3-cyano-2(1H)-kinolinon. Dette er en ny og hensiktsmessig metode for den generelle syntese ay slike 3—cyano—2(1H)-kinolinoner og lignende forbindelser. Den nye fremgangsmåte er særlig fordelaktig fordi (1) den gir det ønskede produkt i høyt utbytte, (2) flere reaksjoner kombineres hensiktsmessig i en enkelt prosess, og (3) de nød-vendige utgangsmaterialer kan erholdes lett. Derimot har de tidligere beskrevne prosedyrer for fremstilling av 3-cyano-2(1H)-kinolinoner innbefattet flertrinns-synteser eller anvendelse av reaktanter slik som 2-aminobenzaldehyd som er ustabilt.
Alternativt kan acetanilidet oppvarmes med dimethylformamid, fosforylklorid og hydroxylamin-hydroklorid under dannelse av et 2-klor-3-cyanokinolin. Denne fremgangsmåte er en særlig hensiktsmessig metode for dannelse av klorcyano- kinolinet direkte fra et acetanilid. Den angitte klor-cyanoforbindelse kan deretter hydrolyseres enten under sure eller basiske betingelser under dannelse av det ønskede 3-cyano-2(1H)-kinolinon. Hvis 2-klor-3-cyanokinolin kokes i eddiksyre i 1 time eller kokes under tilbakeløpskjøling i 4N saltsyre i 10-15 minutter, erholdes det tilsvarende 3-cyano-2(1H)-kinolinon.
Omsetning av 3-cyano-2(1H)-kinolinoner med en sterk base slik som kalium-t-butoxyd og et alkylhalogenid av formel R-halogen hvori R er som ovenfor definert og halogen er klor, brom eller jod, i et egnet løsningsmiddel slik som dimethylsulfoxyd, gir den tilsvarende forbindelse med en R-substituent på kinolinonnitrogenet. Slike forbindelser tjener selvsagt som utgangsmaterialer for forbindelsene ifølge oppfinnelsen hvori R er alkyl.
Tetrazolene ifølge oppfinnelsen omdannes til de tilsvarende farmasøytisk akseptable salter ved omsetning av disse med en hovedsakelig ekvimolar mengde av den egnede base i en vandig løsning eller i et egnet organisk løsningsmiddel slik som methanol eller ethanol. Saltene gjenvinnes etter standardmetoder slik som filtrering hvis disse er uløselige i det opprinnelige medium, eller hvis de er løselige i dette medium, utfelles saltet ved fordampning av løsningsmidlet eller ved tilsetning av et ikke-løsningsmiddel for saltet.
Forbindelsene ifølge oppfinnelsen utviser antiallergisk aktivitet. Således er de anvendbare ved behandling av tilstander hvori antigen-antistoffreaksjoner er ansvarlige for sykdom, og i særdeleshet for behandling av allergiske sykdommer slik som (men ikke begrenset til) ytre astma, høyfeber, urticaria, eksem eller atopisk dermatitis og øvre respirasjonstilstander slik som allergisk rhinitis.
Forbindelsene ifølge oppfinnelsen kan administreres enten som individuelle terapeutiske midler eller som blandinger med andre terapeutiske midler. De kan administreres alene, men administreres generelt i form av farmasøytiske preparater, dvs. blandinger av de aktive midler med egnede farmasøytiske bærere eller fortynningsmidler. Eksempler på slike preparater innbefatter tabletter, piller, kapsler, pulvere, aerosolspray, vandige eller oljeaktige suspensjoner, siruper, eliksirer og vandige løsninger for injeksjon. Forbindelsene administreres fortrinnsvis i oral doseringsform.
Arten av det farmasøytiske preparat og den farmasøyt-iske bærer eller fortynningsmiddel vil selvsagt avhenge av den ønskede administreringsmåte, dvs. oralt, parenteralt eller ved inhalering. Orale preparater kan være i form av tabletter eller kapsler og kan inneholde konvensjonelle eksi-pienter slik som bindemidler (f.eks. sirup, acacia, gelatin, sorbitol, tragant eller polyvinylpyrrolidon), fyllstoffer (f.eks. lactose, sukker, maisstivelse, calciumfosfat, sorbitol eller glycin), smøremidler (f.eks. magnesiumstearat, talkum, polyethylenglycol eller silica), oppløsende midler (f.eks. stivelse) eller fuktemidler (f.eks. natriumlauryl-sulfat). Oralt væskeformige preparater kan være i form av vandige eller oljeaktige suspensjoner, løsninger, emulsjoner, siruper, eliksirer, etc, eller kan presenteres som et tørt produkt for rekonstituering med vann eller annen egnet bærer før bruk. Slike væskepreparater kan inneholde konvensjonelle additiver slik som suspenderingsmidler, smaksgivende midler, fortynningsmidler eller emulgeringsmidler. For parenteral administrering eller inhalering kan løsninger eller suspensjoner av en forbindelse ifølge oppfinnelsen med en hensiktsmessig farmasøytisk bærer anvendes, f.eks. som en aerosolspray for inhalering, eller som en vandig løsning for intra-venøs injeksjon eller som en oljeaktig suspensjon for intra-muskulær injeksjon. Forbindelsene kan også administreres ved hjelp av inhalatoranordninger eller andre anordninger som muliggjør at de aktive forbindelser i form av tørre pulvere kommer i direkte kontakt med lungene. Prosedyrer for fremstilling av preparater som ovenfor diskutert, er beskrevet i standardbøker slik som Remington's Pharmaceutical Sciences, Mack Publishing Company, Easton, Pennsylvania.
Forbindelsene ifølge oppfinnelsen eller farmasøytiske preparater derav kan administreres til humane astmatiske pasienter i enkle orale doser på ca. 1-1000 mg aktiv bestanddel og multiple orale doser på totalt opptil 4000 mg/dag av
aktiv bestanddel. Når de administreres ved inhalering,
gis generelt lavere doser, dvs. i en størrelsesorden på ca. 0. 1 av den normale dose for den bestemte forbindelse. Disse verdier er bare illustrative, og legen vil selvsagt bestemme den mest egnede dose for en bestemt pasient på basis av faktorer slik som alder, vekt, diagnose, alvorlighet av symptomene og det bestemte middel som skal administreres.
Den antiallergiske aktivitet av forbindelsene demon-streres ved den IgE-formidlede, passive kutane anafylakse (PCA)-test på rotter. Denne test er generelt akseptert som en av de beste dyremodeller for kvalitativ bestemmelse av antiallergisk aktivitet. Dinatrium-kromglycat er aktivt i denne test når det administreres intraperitonealt, men ikke oralt. Metoden kan kort beskrives som følger:
PCA- testmetode
1. Antisera - Forskjellige standardmetoder beskrevet i litteraturen, ble anvendt for fremstilling av reaginisk antisera overfor ovalbumin i enten Hooded Lister eller Brown Norway voksne rotter. 2. Dyr - Voksne Sprague-Dawley hannrotter eller Wistar Kyoto hunnrotter ble anvendt som antisera-resipienter i testen. Dyrene fikk akklimatisere seg i 5-14 dager med mat og vann ad lib. 3. Sensibilisering - Resipientrottene ble passivt sensibilisert ved intradermal injeksjon av 100 mikroliter av to fortynninger av antiserum (en injeksjon på hver side av ryggen). Sensibilisering fant sted 48-72 timer før antigenutfordring. 4. Administrering av testforbindelse - 4 til 6 dyr ble anvendt for hver testforbindelse/fortynning. Forbindelsene ble homogenisert i en egnet bærerløsning og administrert intraperitonealt til 60 mg/kg 5 minutter før utfordring eller peroralt til 100 mg/kg 5 til 240 minutter før utfordring. 5. Antigenutfordring og reaksjonsvurdering - Ovalbumin (0,1-1,0 mg i eri 0,5% løsning av Evan<1>s blå farvestoff)
i saltvann ble gitt til hver rotte ved intravenøs administrering. 30 minutter senere ble de resulterende PCA-reaksjoner målt med hensyn til midlere diameter og farveintensitet fra den reflekterte overflate på huden. Aktiviteten av testforbindelsen uttrykkes som prosent inhibering basert på kontrollreaksjoner.
Da forbindelsene ifølge oppfinnelsen ible testet ved den ovenfor angitte prosedyre, ble de funnet å være aktive både intraperitonealt og oralt.
I tillegg til aktiviteten ved den ovenfor beskrevne PCA-test, inhiberte forbindelsene ifølge oppfinnelsen også fri-givelsen av histamin ved den passive peritoneale anafylakse (PPA)-test på rotter. Denne metode kan kort beskrives som følger:
PPA- testmetode
1. Antisera - Reaginisk antistoff overfor ovalbumin for
denne test ble fremstilt i voksne B.D„F. hannmus. 6 2 1 2. Dyr - Voksne Sprague Dawley hannrotter eller Wistar Kyoto hunnrotter ble anvendt som antistoff-
resipienter. Dyrene fikk akklimatisere seg i 5-14 dager med mat og vann ad lib. 3. Sensibilisering - Resipientrotter ble sensibilisert i.p. med 2 ml av en egnet saltvannfortynning av rause-anti-ovalbumin-antiserumet bestemt fra foregående for-søk. Sensibilisering fant sted 2 timer før antigenutfordring . 4. Administrering av testforbindelse - 5 til 10 dyr ble anvendt for hver testforbindelse/fortynning. Forbindelsene ble homogenisert i saltvann med en ekvivalent natriumbicarbonat for å bevirke oppløseliggjøring om
nødvendig, og ble administrert intraperitonealt til 60yUg 30 sekunder før antigenutfordring eller p.o.
5 til 60 minutter før antigenutfordring.
5. Antigenutfordring og vurdering — 2 mg av ovalbumin i
5 ml modifisert Tyrodes løsning ble administrert ved i.p. injeksjon, og dyrene ble avlivet 5 minutter senere. Peritoneale sjokkvæsker ble oppsamlet og klassifisert ved sentrifugering. Protein ble fjernet fra prøvene ved perklorsyreutfelling og etterfølgende sentrifugering. Prøvene ble deretter analysert med hensyn til histamininnhold ved automatisert fluorometrisk bestemmelse. Histaminnivåer i peritoneal-sjokkvæsker fra behandlede dyr ble deretter sammenlignet med de i sjokkvæskene fra kontrolldyrene. Legemiddeleffekten ble uttrykt som prosent inhibering av histaminfrigivelse.
De etterfølgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Til en blanding av 11.880 ml fosforylklorid og 2.500 g acetanilid ble tilsatt under avkjøling og omrøring på et isbad, 3.380 g dimethylformamid i en slik hastighet at temperaturen ikke overskred 60°C. Tilsetningen tok ca. 45 minutter på hvilket tidspunkt kjølebadet ble fjernet og blandingen ble oppvarmet til 75°C i 22 timer. Blandingen ble deretter avkjølt, og overskudd av fosforylklorid ble fjernet ved rotasjonsfordampning. Den mørkebrune restolje ble deretter helt over i 32 1 vann under omrøring. Is ble tilsatt til den vandige blanding for å holde temperaturen under 50°C. Det mørkegule, faste materiale som ble dannet, ble fraskilt ved filtrering og tørket i en luftovn ved 70°C under dannelse av 2-klor-3-kinolincarboxaldehyd som smeltet ved 145-147°C.
Eksempel 2
Til blandingen erholdt ved tilsetning av 210 g 4-(methylthio)-acetanilid til 1. 246 g fosforylklorid, ble til satt 254 g dimethylformamid i løpet av en 30 minutters periode og under omrøring. Reaksjonen var eksoterm, og tilsetnings-hastigheten ble regulert slik at temperaturen ikke overskred 75°C. Etter at tilsetningen var fullført, ble reaksjonsbland-ingen oppvarmet til 75°C i 2,5 timer. Blandingen ble deretter slukket i vann, og det gule bunnfall som ble dannet, ble fraskilt ved filtrering og tørket under dannelse av 2-klor-6-methylthio-3-kinolincarboxaldehyd.
Når den ovenfor angitte prosedyre ble gjentatt under anvendelse av det egnede substituerte acetanilid, ble følgende produkter erholdt: 2-klor-6,7-dimethoxy-3-kinolincarboxaldehyd 2,6-diklor-3-kinolincarboxaldehyd
2—klor-7-methylthiokinolincarboxaldehyd
E ksempel 3
En blanding ble fremstilt fra 6 1 97% maursyre, 300 g hydroxylamin-hydroklorid, 500 g natriumformiat og 700 g 2-klor-3-kinolincarboxaldehyd, og denne blanding ble oppvarmet til tilbakeløpskokning (110°C). Den resulterende løsning ble deretter holdt ved 110°C i 18 timer. Løsningen ble deretter avkjølt, og det faste materiale som krystalliserte, ble fraskilt ved filtrering og deretter suksessivt vasket to ganger med vann, en gang med ethanol og en gang med methylenklorid under dannelse av 3-cyano-2(1H)-kinolinon.
Eksempel 4
En blanding ble fremstilt fra 15 g 2-klor-6,7-dimethyl-3-kinolincarboxaldehyd, 5,4 g hydroxylamin-hydroklorid, 8,5 g natriumformiat og 155 ml 9 7% maursyre, og denne blanding ble oppvarmet til tilbakeløpskokning i 3 timer. Først ble blandingen en tung, gul pasta, men en homogen, brun løsning ble deretter dannet. Ved slutten av 3-timers tilbakeløpskoknings-perioden ble blandingen igjen heterogen, og den ble avkjølt og helt over i 300 ml vann. Det faste materiale som ble dannet, ble fraskilt ved filtrering og tørket under anvendelse av 3-cyano-6,7-dimethy1-2(1H)-kinolinon som smelter ved ca.
300°C. Det angitte produkt inneholdt 1/4 molekyl hydratvann.
Når den ovenfor angitte prosedyre ble gjentatt under anvendelse av egnede utgangsmaterialer, ble følgende forbindelser erholdt: 3-cyano-6,7-dimethoxy-2(1H)-kinolinon (1/4 H20) som smelter over 300°C.
3-cyano-6-methylthio-2(1H)-kinolinon (1/6 H20) som smelter ved 287-288°C.
6-klor-3-cyano-2(1H)-kinolinon.
3-cyano-7-methylthio-2(1H)-kinolinon.
Eksempel 5
Til en løsning av 10 ml 30%-ig hydrogenperoxyd og 100 ml eddiksyre ble tilsatt 4,0 g 3-cyano-6-methylthio-2(1H)-kinolin, og blandingen ble oppvarmet til tilbakeløpskokning i 1,5 timer. En homogen løsning ble først dannet, men under reaksjonsfor-løpet fremkom det et lyst gult bunnfall. Blandingen ble av-kjølt, og det faste materiale ble fraskilt ved filtrering under dannelse av 3-cyano-6-methylsulfonyl-2(1H)-kinolinon som smelter over 310°C.
Når den ovenfor beskrevne prosedyre ble gjentatt under anvendelse av 3-cyano-7-methylthio-2(1H)-kinolinon, var det erholdte produkt 3-cyano-7-methylsulfonyl-2(1H)-kinolinon.
Eksempel 6
Til en blanding av 118 ml fosforylklorid og 25 g acetanilid ble tilsatt under omrøring og avkjøling på et isbad,
41 g dimethylformamid i en slik hastighet at temperaturen ikke overskred 75°C. Etter at tilsetningen var fullført, ble en varmelampe tilført, og temperaturen ble holdt ved 75°C i 20 timer. Oppvarmingen ble deretter stoppet, og blandingen fikk avkjøle i noen få minutter, og temperaturen falt til 62°C. 14 g hydroxylamin-hydroklorid ble tilsatt alt på en gang til den omrørte blanding. Etter 2-3 minutter startet en langsom eksoterm reaksjon, og blandingen begynte å koke med betydelig gassutvikling. Temperaturen steg langsomt fra 6 2°C til 77°C i løpet av 15 minutter. På dette tidspunkt hadde gassutviklingen praktisk talt stoppet. Blandingen fikk deretter avkjøle til romtemperatur, og et tungt, gult, fast materiale utfeltes. Blandingen ble deretter stanset for-siktig ved tilsetning av 1000 ml vann under kraftig omrøring. Det faste materiale ble deretter fraskilt ved filtrering og ble oppløst i methylenklorid, og methylenkloridløsningen ble behandlet med benkull, filtrert, konsentrert og avkjølt. Filtrering ga deretter lysegule krystaller av 2-klor-3-cyano-kinolin.
Eksempel 7
En blanding ble fremstilt ved tilsetning av 5,0 g 3-cyano-2(1H)-kinolinon, 10 g kalium-t-butoxyd og 4,6 g methyljodid til 50 ml dimethylsulfoxyd, og blandingen ble oppvarmet til 80°C i 3 timer. En mørk, homogen løsning ble erholdt. Denne ble av-kjølt og helt over i vann, og den vandige blanding ble ekstra-hert med methylenklorid. Det organiske ekstrakt ble deretter behandlet med silicagel og filtrert, og filtratet ble konsentrert inntil krystallisering fant sted. Dette ga 3-cyano-l-methyl-2(1H)-kinolinon som smelter ved 202-205°C.
Eksempel 8
Til en løsning av 250 g 3-cyano-2(1H)-kinolinon i 3000 ml dimethylformamid ble tilsatt 86,5 g ammoniumklorid og 105 g natriumazid. Den omrørte heterogene løsning ble oppvarmet til 110°C i 17 timer. Blandingen ble deretter avkjølt og helt over i 8 1 vann og ble surgjort til pH 2 med konsentrert saltsyre under kraftig omrøring. Bunnfallet som ble dannet, ble fraskilt ved filtrering, vasket med vann og tørket i vakuum. Det urene produkt ble omkrystallisert fra varmt dimethylformamid hvori vann ble tilsatt til den varme løsning inntil utfellingen startet, og blandingen ble deretter avkjølt til romtemperatur. Det erholdte faste materiale var 3-(lH)-tetrazol-5-yl)-2(1H)-kinolinon.

Eksempel 9
En suspensjon av 690 g av produktet erholdt i foregående eksempel i 8 1 50%-ig vandig ethanol, ble oppvarmet til 70°C. En løsning av 136 g natriumhydroxyd i 500 ml vann ble tilsatt til den omrørte løsning i løpet av 10 minutter. Det opprinnelige faste materiale ble praktisk talt fullstendig oppløst, og etter flere minutter ble det dannet et massivt bunnfall. Blandingen fikk deretter avkjøle og det faste materiale ble fraskilt ved filtrering under dannelse av natriumsaltet av 3-(lH-tetrazol-5-yl)-2(lH)-kinolin.
Eksempel 10
En blanding av 5,0 g 3-cyano-2(1H)-kinolinon, 1,9 g ammoniumklorid og 2,4 g natriumazid i 50 ml dimethylformamid ble oppvarmet til 120°C i 16 timer. Blandingen ble deretter helt over i 250 ml vann, og den resulterende blanding ble surgjort til pH 2 med konsentrert saltsyre. Det faste materiale som ble dannet, ble fraskilt ved filtrering og tørket. Det ble deretter oppløst i vandig natriumhydroxyd, og den vandige løsning ble vasket med methylenklorid og filtrert, og filtratet ble surgjort med saltsyre. Det lysebrune, faste materiale som ble dannet, ble fraskilt ved filtrering og tørket under dannelse av 3-(lH-tetrazol-5-yl)-2(1H)-kinolinon 1/9 hydrat som smelter ved 301-303°C under spaltning.
E ksempel 11
Når prosedyren beskrevet i det foregående eksempel, ble gjentatt under anvendelse av det egnede substituerte 3-cyano- 2(1H)-kinolinon, ble følgende forbindelser erholdt: 6,7-dimethyl-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon-hemihydrat som smelter ved 316-318°C under spaltning.
6-klor-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon, solvat med dimethylsulfoxyd, og som smelter over 300°C.
6- methylsulfonyl-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon-tetrahydrat med smeltepunkt over 300°C.
l-methyl-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon med smeltepunkt på 302-303°C under spaltning.
7- methylthio-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon som smelter over 300°C.
7-methylsulfonyl-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon som smelter over 300°C.
6,7-dimethoxy-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon ble også erholdt fra den egnede cyanoforbindelse.
Eksempel 12
En løsning ble fremstilt ved oppvarming av 6,0 deler 3-cyano-2(1H)-nafthyridinon i 150 ml dimethylformamid. Deretter ble det tilsatt 1,9 g ammoniumklorid og 2,3 g natriumazod, og den resulterende løsning ble oppvarmet til 120°C i 84 timer. Blandingen ble deretter helt over i 500 ml vann og ble surgjort til pH 2 med konsentrert saltsyre. Det lysegule bunnfall som ble dannet, ble fraskilt ved filtrering, vasket med vann og tørket under dannelse av 3-(lH-tetrazol-5-yl)-2(1H)-nafthyridinon (1/3 hydroklorid) som smelter over 300°C.
Claims (8)
1. Fremgangsmåte for fremstilling av en forbindelse av formel:
hvori A er -CH= eller -N=; R er H eller alkyl med 1-4 C-atomer; n er 0, 1 eller 2; X er H, alkyl med 1-4 C-atomer, alkoxy med 1-4 C-atomer, halogen, methylmercapto, methylsulfonyl, eller hvor to X'er kan kombineres som methylendioxy; forutsatt at når X er methylmercapto eller methylsulfonyl, må n være lik 1} og farmasøytisk akseptable salter derav, karakterisert ved at et cyano-2(1H)-kinolinon av formel:
hvori n, A, R og X er som ovenfor definert, omsettes med natriumazid og ammoniumklorid i et inert løsningsmiddel, under dannelse av den ønskede tetrazol, eventuelt etterfulgt av behandling med en løsning av en egnet base under dannelse av det tilsvarende farmasøytisk akseptable salt.
2. Fremgangsmåte ifølge krav 1 for fremstilling av en forbindelse av formel:
hvori A er -CH= eller -N=; R er H eller alkyl med 1-4 C-atomer; n er 0, 1 eller 2; X er H, alkyl med 1-4 C-atomer, alkoxy med 1-4 C-atomer, halogen, methylmercapto, methylsulfonyl, eller hvor to X'er kan være kombinert som methylendioxy; forutsatt at når X er methylmercapto eller methylsulfonyl, må n være lik 1; og farmasøytisk akseptable salter derav; karakterisert ved at et cyano-2(1H)-kinolinon av formel:

hvori n og X er som ovenfor definert, omsettes med natriumazid og ammoniumklorid i et inert løsningsmiddel under dannelse av den ønskede tetrazol, eventuelt etterfulgt av behandling med en løsning av en egnet base under dannelse av de tilsvarende farmasøytisk akseptable salter.
4. Fremgangsmåte ifølge krav 1 for fremstilling av 3-(lH-tetrazol-5—yl)-2(1H)-kinolinon og farmasøytisk akseptable salter derav, karakterisert ved at 3-cyano-2(1H)-kinolinon omsettes med natriumazid og ammoniumklorid i et inert løsningsmiddel under dannelse av 3-(lH-tetrazol-5-yl)-2(1H)-kinolinon, eventuelt etterfulgt av behandling med en løsning av en egnet base under dannelse av det tilsvarende farmasøytisk akseptable salt.
5. Fremgangsmåte ifølge krav 1 for fremstilling av 3-(lH-tetrazol-5-yl)-2(1H)-kinolinon,
karakterisert ved at 3-cyano-2(1H)-kino-linon omsettes med natriumazid og ammoniumklorid i et inert løsningsmiddel.
6. Fremgangsmåte ifølge krav 1 for fremstilling av natriumsaltet av 3-(lH-tetrazol-5-yl)-2(1H)-kinolinon, karakterisert ved at 3-cyano-2(1H)-kinolinon omsettes med natriumazid og ammoniumklorid i et inert løs-ningsmiddel under dannelse av 3-(lH-tetrazol-5-yl)-2(1H)-kinolinon, som deretter omsettes med en vandig løsning av natriumhydroxyd.
7. Fremgangsmåte ifølge krav 1 for fremstilling av 1-methyl-3-(lH-tetrazol-5-yl)-2(1H)-kinolinon, karakterisert ved at 3-cyano-l-methyl-2(1H)-kinolinon omsettes med natriumazid og ammoniumklorid i et inert løsningsmiddel.
8. Fremgangsmåte ifølge krav 1 for fremstilling av 3~ (1H-tetrazol-5-yl)-2(1H)-nafthyridinon, karakterisert ved at 3-cyano-2(1H)-nafthyri-dinon omsettes med natriumazid og ammoniumklorid i et inert løsningsmiddel.
