Abstract
Foreliggende oppfinnelse gjelder et oligonukleotid med en ny struktur og en fremgangsmåte for syntese av en nukleinsyre ved anvendelse av oligonukleotidet som primer. I sin 5'-ende er oligonukleotidet utstyrt med en basesekvens som i det vesentlige er identisk med området som skal syntetiseres ved anvendelse av primeren som utgangspunkt for syntesen. Således kan en nukleinsyre syntetiseres i en iso-term reaksjon ved anvendelse av en enkel reagensblanding. Videre tilveie-bringes en fremgangsmåte for syntese av en nukleinsyre med høy spesifisitet, basert på den ovenfor beskrevne fremgangsmåte for syntese av en nukleinsyre .The present invention relates to an oligonucleotide having a novel structure and a method for synthesizing a nucleic acid using the oligonucleotide as a primer. At its 5 'end, the oligonucleotide is provided with a base sequence that is substantially identical to the region to be synthesized using the primer as a starting point for the synthesis. Thus, a nucleic acid can be synthesized in an isothermal reaction using a simple reagent mixture. Furthermore, a method for synthesizing a high specificity nucleic acid is provided, based on the above-described method for synthesizing a nucleic acid.
