Description
Oppfinnelsen vedrører en fremgangsmåte for å bevare virkningen av bakteriepreparater, hvor bakteriene har sin opprinnelse fra tarmen til friske fjærkre, og hvor bakteriene, når de administreres som vandige blandinger, er nyttig for behandlingen av den normale tarmbakteriefloraeii hos fjærkre samt for sykdomsforebyggende behandling av forstyrrelser i bakteriefloraen. Virkningen opprett-holdes ved å tilsette preparatet cystein, som reduserer den giftige virkningen av oksygenet som er oppløst i vannet. Det henvises til det medfølgende patentkrav.
Friske varmblodige dyr har en tarmbakterie-likevekt som
er typisk for hver art. Individuelle forskjeller innenfor de samme artene skyldes særlig dietten og alderen til dyret. Vanligvis er likevekten stabil og har en vesentlig buffervirkning mot ytre forstyrrelser. Forstyrrelser i likevekten oppstår der-som en av bakteriene som vanligvis er til stede i likevekts-blandingen eller en fullstendig fremmed bakterie tar overhånd over de øvrige. Dette kan forårsakes av en sykdom eller ved virkningen av antibiotika som brukes i behandlingen av sykdom-
men.. Det kan også skyldes matforgiftning eller en smittende tarminfeksjon. Nyfødte, eller svært unge individer av de fleste arter er særlig utsatt for å bli smittet av tarminfeksjoner, ettersom tarmbakteriefloraen ikke har nådd en likevektstil-
stand med buffervirkning.
I prinsippet er det to typer preparater for behandlingen
av den normale bakteriefloraen og for den sykdomsforebyggende behandling av forstyrrelser i denne. Én type representeres typisk av næringsstoffer, f.eks. bestemte sukkere, som selektivt fremmer veksten av visse bakteriearter som tilhører den norma-
le floraen eller understøtter denne floraen. Slike preparater er spesielt nyttige ved gjenopprettingen og opprettholdelsen av den tidligere likevekt.
Det andre typiske preparatet omfatter administrering av levende, anaerobe bakterier enten i form av hele normalfloraen eller som et representativt utvalg derav. Bruken av slike preparater øker i vesentlig grad etableringen av en stabil like-
vekt i den normale floraen hos svært unge individer, samt etter alvorlige forstyrrelser i floraen. Visse preparater basert på dette prinsippet inneholder bare én bakterieart, og er derfor bare egnet for støttebehandlingen av lette forstyrrelser.
Flertallet av de normalt tilstedeværende tarmmikrobene er obligate anaerober. I motsetning til fakultativt anaerobe eller oksygen-tolerante anaerobe bakterier er de svært følsomme for den giftige virkningen av oksygen. Det er derfor svært viktig å innrette dyrkingsbetingelsene slik at denne virkningen minimaliseres. Den giftige virkningen av oksygen står i forbindelse med cellenes metabolisme og derfor er f.eks. lyofiliserte celler, som har en svært langsomt virkende metabolisme, mye mindre følsomme for den giftige virkningen av oksygen.
Når vanninnholdet i cellene øker ved en egnet temperatur, star-ter metabolismen opp igjen og følsomheten for virkningen av oksygen kommer tilbake.
Ved bruken av et slikt preparat som inneholder lyofilisex-te celler og som skal blandes opp med vann, er det viktig å beskytte cellene mot virkningen av oppløst oksygen, særlig der-som preparatet blandes med vann lenge før det skal brukes. Oppfinnelsen vedrører tilsetningen til preparatet av en slik forbindelse som reagerer med det oksygenet som er oppløst i vannet, hvorved oksygenet gjøres ufarlig for cellene. Et eksempel på en slik forbindelse er cystein. Ettersom virkningen av cystein er avhengig av surheten i oppløsningen, må bufferen i oppløsningen også være justert til en tilnærmet nøytral pH-verdi når cysteinet brukes.
Den viktigste tarminfeksjonen i verden, salmonellosis, spres vanligvis via oppdrettshus for fjærkre, særlig via moder-ne oppdrettshus for broilere.
Den industrielle broilerproduksjon avskjærer den naturli-ge, koprofage overføring av den anaerobe bakteriefloraen fra voksne fugler til nyklekkede kyllinger. I løpet av de to førs-te leveukene er disse kyllingene særlig følsomme for Salmonella-infeksjoner. For å avskjære denne smitteveien har det vært gjort anstrengelser for å utvikle preparater som inneholder anaerobe bakterier, idet preparatene utblandet med det første drikkevannet til den nyklekkede kyllingen ville fremme oppret-telsen av en Salmonella-resistent tarmflora. Finsk patentskrift nr. 59925 beskriver slike bakteriepreparater som er virkningsfulle ved den sykdomsforebyggende behandling av Salmonella-infeksjoner. Et annet bakteriepreparat er beskrevet i europeisk patentpublikasjon nr. 66 95. Den hurtige opprettel-sen av en likevekt i den normale floraen øker veksthastigheten til kyllingene og forbedrer økonomien ved fåringen. Fremgangsmåten for å beskytte obligate anaerobe bakterier ifølge foreliggende oppfinnelse er særlig egnet for bruk med dette spesielle preparatet ettersom det kan gå flere timer fra det. øyeblikket preparatet blandes med vann og til kyllingen drik-ker det første vannet.
Eksempel 1
Bakteriecellemassen som er blitt fremstilt Ifølge fremgangsmåten (eksempel 1) beskrevet i finsk patentskrift nr-59925, separert fra dyrkingsmediet og dypfryst til -70°C, ti-nes opp. Cellemassen fortynnes til en celletetthet på 1,5 x 10 g/ml eller trinnvise, ti-gangers fortynninger fra denne ved å bruke enten ubehandlet vann fra vannkranen eller vann fra vannkranen som er 0,05 mol dm ^ med hensyn til en fosfatbuffer ved pH 7,0 og som inneholder 0,25 % (vekt/volum) L-cystein.
De 1 dag gamle broilerkyllingene oppdeles i grupper på seks kyllinger pr- gruppe og 1 ml av hver oppløsning pr. gruppe administreres direkte i kyllingenes hals. En gruppe holdes som kontroll, idet den ikke får noen behandling med oppløsning. Etter 1 dag infiseres kyllingene med Salmonella infentis ved å administrere omtrent 1000 celler direkte i halsen. Deretter oppales kyllingene i 5 dager idet de får f6r som ikke inneholder noen coccidiostatiske forbindelser og friskt vann etter be-hag. På den 6. dagen avlives kyllingene ved hjelp av karbon-monoksydgass, og fra innholdene i blindtarmene bestemmes meng-den salmonella-organismer ved inkubering av 10 ^, 10 ^ og 10 BTB-LS og SS-0n5z-fortynninger av innholdene i blindtarmene (S. Schneitz et al., Acta Path. Microbiol. Scand. Sect. B 89: 109-116, 1981; E. OnSz & K. Hoffman, Zbl. Bakt. Hyg., I Abt., Orig. A 240: 16-21, 1978) på dyrkingsmedier som er selektive for salmonella. Salmonella-organismer anrikes videre fra innholdene i blindtarmene i en selenitt-væske (E- Leifson, Am. J. Hyg. 24: 423-432, 1936), hvorfra dyrkingen gjøres på BTB-LS og SS-OnSz-dyrkingsmedier. Resultatene fra forsøket er samlet i tabell 1.
Eksempel 2
Bakteriecellemassen fremstilt ifølge fremgangsmåten beskrevet i finsk patentskrift nr. 59925 og separert fra dyrkingsmediet lyofiliseres. Denne lyofiliserte cellemasse brukes enten som sådan eller oppblandet med mono- og dikaliumhydro-genfosfater og L-cystein i slike mengder at fortynningen av cellemassen for undersøkelsen av den profylaktiske virkningen på salmonella-kolonisering gir en oppløsning som inneholder

0,05 mol dm fosfatbuffer ved pH 7 og 0,25 % (vekt/volum)
L-cystein. Disse cellemassene fortynnes med vann fra vannkranen til blandinger som inneholder 10 mg ml 1 eller 25 mg ml * cellemasse. Én dag gamle broilerkyllinger fordeles i grupper med seks kyllinger pr. gruppe og 1 ml av hver oppløsning pr. gruppe inkuberes direkte i halsen til kyllingene. Kontroll-
gruppen behandles ikke med noen oppløsning. Prøven følger el-lers prinsippene fra forsøket i eksempel 1. Prøveresultatene som ble oppnådd med en cellemasse er fremlagt i tabell 2.
Claims (1)
- Fremgangsmåte for å bevare virkningen av bakteriepreparater, hvor bakteriene har sin opprinnelse fra tarmen til friske fjærkre og hvor bakteriene, når de administreres som vandige blandinger, er nyttige ved behandlingen av den normale tarmbakteriefloraen hos fjærkre samt ved den sykdomsforebyggende behandling av forstyrrelser i bakteriefloraen, karakterisert ved at det ved siden av selve bakteriepreparatet er tilsatt cystein som reduserer den giftige virkningen av oksygenet som er opplost i vannet, idet cystein brukes som virkningsbevarende middel, i en slik mengde at innholdet av cystein er minst 0,05 vekt%, og oppløsningen buf res til en tilnærmet nøytral pH-verdi,. fortrinnsvis med fosfatsalter.
