Description
Analogifremgangsmåte til fremstilling av en ny, substituert antranilsyre med terapeutisk virkning.
Nærværende oppfinnelse vedrorer en fremgangsmåte til fremstilling av N-(a,a,a,a',a',a'-heksafluor-3,5-xylyl)-antranilsyre med formel I og dens salter med uorganiske og organiske baser. Det er nå blitt funnet at forbindelsen med formel I og dens salter innehar verdifulle farmakologiske egenskaper, spesielt antiflogistisk (antiinflammatorisk), analgetisk og antipyretisk virkning med gunstig terapeutisk indeks. Den antiflogistiske virkning av forbindelsen med formel I lar seg påvise ved forsok med dyr f.eks. ved oral administrasjon ved UV-erythem hos marsvin og ved Bolus alba-odem hos rotte (pote); Videre i vattgranulom og sår-granulom.
Den antiinflammatoriske virkning for
1. N-(a,a,a,a<1>,a',a'-heksafluoro-3,5-xylyl)-antranilsyre,
som fremstilles ifolge nærværende oppfinnelse, ble sammenlignet med den for 2. N-(a,a,a-trifluoro-m-tolyl)-antranilsyre (flufenaminsyre; fremstilt ifolge det amerikanske patent nr. 3 144 387).
Den nedenfor angitte prøvemetode ble anvendt og de etterfølgende resultater ble oppnådd.
Den antiinflammatoriske virkning ble bestemt ved å måle innvirk-ningen av provesubstansene på odema på rottepoter som på sin side ble indusert ved å injisere en 10%'s suspensjon av bolus alba i rottepotene (G. Wilhelmi: Japanese J. Pharmacol. 15, 187-198 (1965). Provedyrene i grupper på 20 hvite hanrotter som veide mellom 110 og 130 g ble brukt ved hver dose. Forbindelsene som skal undersokes ble administrert oralt i form av en tragant
suspensjon i en mengde som angitt i den nedenstående tabell I.
En time senere ble 0,1 cm 3 av en 10%'s suspensjon av bolus alba injisert subkutant i en bakpote i hvert av provedyrene.
Dyrene ble drept fem timer senere og bakpotene ble amputert. Svellingen ble bestemt ved å måle vektdifferansen mellom den venstre normale pote og den hoyre svellede pote. Gjennomsnitts-vekten for svellingen ble sammenlignet med gjennomsnittsvekten for svellingen hos en kontrollgruppe som ikke hadde mottatt proveforbindelsene men ble behandlet på samme måte som tidligere beskrevet med bolus alba. Reduksjonen i svellingen er et mål for den anti-inflammatoriske virkning og uttrykkes i prosent av kontrollen.
Proveforbindelsene administreres i doser på 100, 50, 25, 10, 5, 2,5 og 1 mg/kg legemsvekt inntil virkningen opphorte f or å bestemme den minste vesentlig aktive dose.
Den etterfolgende tabell viser denne minimale effektive dose (DE<min*>) og den gjennomsnittlige reduksjonen i svelling uttrykt som en prosentandel.
Resultatene som er angitt i tabell I viser den farmakologiske virkning av forbindelsen som fremstilles etter nærværende oppfinnelse, og som det vil ses er denne overlegen den for den kjente forbindelse.
Spesielt fordelaktig overfor den kjente flufenaminsyre er
den ytterst lave toksisitet forden ifolge oppfinnelsen fremstill-bare forbindelse.
Den frie syre og dens salter med farmakologisk aksepterbare uorganiske og organiske baser kan anvendes oralt, rektalt,
eller lokalt, og vandige oppløsninger av salter også parenteralt, spesielt intramuskulært, såvel som intravenost eller subcutant til behandling av revmatiske, artritiske og andre betennelses-sykdommer hos varmblodige dyr.
N-(a,a,a,a',a',a'-heksafluor-3,5-xylyl)-antranilsyre med formel
I og dens salter fremstilles ifolge oppfinnelsen ved at man kondenserer en o-halogenbenzosyre eller et salt av denne med a,a,a,a<1>,a',a<1->heksafluor-3,5-xylidin og, hvis onsket, frisetter syren fra et forst erholdt salt eller overforer et forst erholdt salt til et annet salt, eller overforer den frie syre til et salt med en uorganisk eller organisk base. Fortrinnsvis gjennomføres kondensasjonen i varme og i nærvær av en kobberholdig katalysator såvel som et syrebindende middel i et under reaksjonsbetingelsene inert organisk opplosningsmiddel. Som o-halogenbenzosyre anvendes spesielt o-klor- eller o-brom-benzosyre. Som kobberholdige katalysatorer kan f.eks. metallisk kobber med stor overflate, som kobberpulver eller svampkobber, eller kobberforbindelser, som kobber-(I)-klorid eller -bromid, kobber-<II)-klorid, -bromid,
-sulfat, -acetat, -karbonat eller oksyd, anvendes, som syrebindende midler egner seg f.eks. alkalimetallkarbonater, som kaliumkarbonat, videre kobber-(I)-karbonat eller kobber-(II)-karbonat eller tertiære organiske aminer. Anvendes som utgangsstoff en fri o-halogenbenzosyre, så anvender man fortrinnsvis to ekvivalenter syrebindende middel og avdestiUerer eventuelt det ved den inntrådte saltdannelse frisatte vann alene eller sammen med en del av opplosningsmidlet for hovedreaksjonen. Derimot vil
det være tilstrekkelig med i det vesentlige en ekvivalent mengde syrebindende middel når man anvender et på forhånd dannet salt av en o-halogenbenzosyre, f.eks. et alkalimetallsalt , som kaliumsaltet, som utgangsstoff, da i det tilfelle bare det ved kondensasjonen frigjorte hydrogenhalogenid skal nøytraliseres. Reaksjonstemperaturene ligger fortrinnsvis mellom 100 og 200°. Gjennomfores kondensasjonen ved normaltrykk, anvender man derfor som reaksjonsmedium spesielt slike organiske opplosningsmidler som koker innenfor eller over dette område. Som eksempel skal nevnes n-pentanol, isopentanol, n-butanol, dimetylformamid, di-metylsulfoksyd eller dietylenglykoldimetyleter. Hovedreaksjonen kan også gjennomfores i lukket, kar, og i dette tilfelle kan også et lavere kokende opplosningsmiddel, f.eks. en lavere alkanol, som etanol, isopropanol eller metanol finne anvendelse som reaksjonsmedium.
N-(a,a,a,a',a1,a'-heksafluor-3,5-xylyl)-antranilsyre med formel
1 såvel som dens farmakologisk anvendbare salter administreres, som foran nevnt, peroralt, rektalt eller parenteralt. Daglige doser for voksne pasienter er mellom 2 og 15 mg/kg, fortrinnsvis mellom 5 og 10 mg/kg kroppsvekt.
Det etterfølgende eksempel redegjbr nærmere for fremstillingen
av N-(a,a,a,a',a',a<1->heksafluor-3,5-xylyl)-antranilsyre. Tempera-turene er angitt i Celsiusgrader.
EKSEMPEL
N-(a,a,a,a',a<1>,a<1->heksafluor-3,5-xylyl)-antranilsyre.
En blanding av 7lO g o-klorbenzosyre og 298 g 85%'ig kalium-hydroksyd i 2500 ml n-pentanol oppvarmes under roring til 160°
(badtemperatur). I lopet av 30 minutter avdestilleres ca. lOOO ml n-pentanol. Deretter tilsetter man 1560 g a/a/a/a,/a,/a'-heksafluor-3,5-xylidin og 17 g kobberpulver og koker blandingen i 15 timer under tilbakelop. så avkjoler man, heller blandingen i en opplosning av 244 g natriumkarbonat i 2000 ml vann og des-tillerer den erholdte opplosning med vanndamp. Etter at det overskytende a,a,a,a' , a' ,<x'-heksafluor-3, 5-xylidin er avdestil-lert, tilsetter man aktivkull til den vandige opplosning og
frafiltrerer over Hyflo. Filtratet surgjores med konsentrert saltsyre. De utskilte krystaller filtreres fra og oppslemmes i 10 liter varmt vann. Man frafiltrerer og krystalliserer fra etanol. N-(a,a,a,a',a<1>,a'-heksafluor-3,5-xylyl)-antranilsyre smelter ved 195-197°.
Man opploser 3,5 g N-(a,a,a,a',a1,a'-heksafluor-3,5-xylyl)-antranilsyre i 10 ml l-n NaOH og konsentrerer opplosningen under 11 Torr til torrhet. Resten krystalliserer man fra eter/petrol-eter. Man oppnår natriumsaltet av N-(a,a,a,a 1,a 1,a1-heksafluor-3,5-xylyl)-antranilsyre som fargelose krystaller med smeltepunkt 289-293°.
