Foreliggende oppfinnelse angår den i
og for seg kjente fremgangsmåte ved fremstilling av et legemiddel i form av perler
hvor legemidlet oppløses eller dispergeres
i en oppløsning av et svellende kolloid og
derpå i varm tilstand gjennom luft dråpevis
tilføres et forholdsvis høyt væskebad, i hvilket kolloidet stivner, hvorved legemidlet
fåes i form av runde perler. Det nye ved
oppfinnelsen består i å skaffe perler av
nøyaktig rund form og dette oppnås ifølge
oppfinnelsen ved at temperaturen oventil i
badet holdes på en verdi som i det vesentlige tilsvarer oppløsningens eller dispersjonens temperatur og holdes suksessivt
synkende i retning nedover, slik at væskedråpene under sin passasje gjennom den
varmere øvre del av badet antar kuleform
og deretter stivner i den kaldere nedre del
av badet. Avhengig av det anvendte lege-middels konsistens og oppløselighetsegen-skaper kan fremgangsmåten varieres etter
flere alternativer som i det følgende skal

Først tilberedes f. eks. som i og for seg
kjent en vannoppløsning av et svellende
kolloid, som gelatin, agar-agar, gummi,
acacia, pektin, stivelse, tragant, kasein
m. fl., disses nedbrytningsprodukter eller
blandinger av disse stoffer. Doseringen kan
derved gjøres slik at man får en kolloidal
oppløsning, som er flytende ved forhøyet
temperatur, men fast ved normal temperatur. Til denne kolloidale oppløsning, f. eks.
gelatinoppløsning, kan, hvis ønskes, settes
plastiseringsmiddel, såsom glykose, sorbitt,
etylenglykol, honning, sirup o.l. i en mengde
som er avveiet, slik at sluttproduktet får
ønsket hårdhet. Deretter tilsettes oppløs-
ningen en hensiktsmessig mengde av det eller de legemidler som kommer på tale.
Hvis legemidlet er vannoppløselig, opp-løses det enten direkte i kolloidaloppløsnin-gen eller oppløses i et med denne blandbart oppløsningsmiddel, f. eks. vann. Foreligger legemidlet i form av en vannuoppløseiig olje, dispergeres det i kolloidaloppløsningen under effektiv omrøring til en stabil emul-sjon eller kolloidal oppløsning, idet man kan anvende emulgatorer, så som lecitin, tween m. fl. Man kan også hvis ønskes blande legemidlet med en olje og siden dis-pergere blandingen. Ved anvendelse av et fast og vannuoppløseiig legemiddel kan det oppløses i en hensiktsmessig olje og opp-løsningen dispergeres i kolloidoppløsningen eller også dispergeres direkte i fast, til-strekkelig finfordelt form, hvilket sist-nevnte også skjer hvis det er tale om et fast, helt uoppløselig legemiddel. Oppløsningen kan videre tilsettes et eller flere aseptiske midler, f. eks. nipagin, med smakkorrigens, så som peppermynte-eller sitronolje, og farges på ønsket måte.
For å få klare perler, kan brytningsindeks
for begge fasene justeres til omtrent samme
verdi, hvilket f. eks. kan skje ved at man til olj efasen setter en hensiktsmessig mengde harpiks. Det har også vist seg fordelaktig
før inndryppingen i væskebadet å innstille den kolloidale oppløsnings resp. dispersjonens pH i hovedsaken til det svellende kol-loidets isoelektriske punkt.
Deretter dryppes kolloidoppløsningen med det uoppløste resp. dispergerte legemiddel gjennom luft i en væske som ikke
oppløser kolloidet, f. eks. parafinolje, en annen mineral-, vegetabilsk eller animalsk
olje, et organisk oppløsningsmiddel, som ikke er blandbart med oppløsningsmidlet som anvendes for den kolloidale oppløs-ning, eller blandinger av de nevnte stoffer. Legemidlet oppløses eller dispergeres i en vannoppløsning av et cellulosederivat, f. eks. karboksymetylcellulose, som deretter dryppes inn i en vannoppløsning av en syre eller et organisk oppløsningsmiddel. Inndryppingen kan foregå ved hjelp av hvilken som helst hensiktsmessig, i og for seg kjent an-ordning for oppnåelse av en regulerbar dråpemengde, f. eks. en pumpeanordning. Kolloidaloppløsningen resp. -dispersjonen holdes ved forhøyet temperatur og ifølge oppfinnelsen skal badet først holdes hovedsakelig ved samme temperatur, som siden suksessivt senkes, slik at dråpene stivner. Dette kan gjennomføres f. eks. slik at badet har form av et vertikalt rør med forholdsvis stor høyde, idet temperaturen oventil i røret holdes høyere og synker suksessivt nedover. Ved inndryppingen antar dråpene kuleform, idet de langsomt synker nedover i den varme væske og stivner siden under bi-behold av denne form når de kommer ned i de koldere regioner.
De stivnede perler oppsamles siden og befris fra badvæske ved vasking med et hensiktsmessig oppløsningsmiddel, f. eks. hvis det gjelder olje, med bensin, metylenklorid, bensen, trikloretylen osv., hvoretter de tørkes. Vannet kan også fjernes fra perlene ved ekstraksjon med et hensiktsmessig vannoppløselig oppløsningsmiddel, så som aceton, metyl-, etyl-, propylalkohol, dioksan osv. For å hindre at perlene ved tørkingen kleber ved hinannen eller deformeres ved for intens tørking, kan det være hensiktsmessig ved vaskingen å la det bli tilbake en tynn film av badvæsken på perlene og først etter tørkingen fullføre vaskingen.
De ferdige perler kan siden eventuelt drageres, idet det da, hvis ønskes kan in-korporeres ytterligere en virksom substans i dragéovertrekket. Eksempel : Fremstilling av multivitaminperler: 1140 g gelatin får svelle i koldt vann. Deretter tilsettes 1125 g stivelsessirup, 435 g glyserin, 21 g B:-vitamin, 9 g B2-vitamin, 21 g B(;-vitamin, 210 g nikotylamid, 21 g Capantotehat og 5,4 g nipagin oppløst i varmt vann. Den sammenlagte vannmengde er 2 kg. Heretter tilsettes 75 g A-vitamin palmitat, 1,5 g D.,-vitamin, 21 g E-vitamin og 16,6 g peppermynteolje oppløst i 270 g rapsolje. Blandingen omrøres godt til en homogen suspensjon er oppnådd, og holdes ved konstant temperatur under omrøring i en 50° C termostat. Gelatinoppløsningen
får deretter dryppe ned i parafinolje. Para-finoljens temperatur holdes maks. ved 50° C, men senkes suksessivt nedover så den ved bunnen av parafinoljekaret bare er 20° C. Gelatindråpene synker langsomt nedover og antar rund form, mens de passerer den varme olje og stivner siden under bi-behold av denne form når de kommer nedover i de koldere regioner. Gelatinperlene befris for den største del av vedhengende parafinolje ved vasking med bensin. Perlene tørkes til passende hardhet og vaskes helt olj efrie med bensin og overtrekkes i drage-ringskjel med et lag C-vitamin og sukker-sirupbelegg.
Claims (5)
1. Fremgangsmåte til fremstilling av et legemiddel som er dispergert i et bære-stoff i dosert tilstand hvor legemidlet opp-løses eller dispergeres i en oppløsning av et svellende kolloid og derpå i varm tilstand gjennom luft dråpevis tilføres et forholdsvis høyt væskebad i hvilket kolloidet stivner, hvorved legemidlet fåes i form av perler, karakterisert ved at temperaturen oventil i badet holdes ved en verdi som i det vesentlige tilsvarer oppløsningens eller dispersjonens temperatur og holdes suksessivt synkende i retning nedover, såle-des at væskedråpene under sin passasje gjennom den varmere øvre del av badet antar fullstendig kuleform.
2. Fremgangsmåte ifølge påstand 1, karakterisert ved at som badvæske anvendes parafinolje, en annen mineral-, vegetabilsk eller animalsk olje eller et i vann uoppløselig organisk oppløsningsmiddel.
3. Fremgangsmåte ifølge påstandene 1—2, karakterisert ved at legemidlet opp-løses eller dispergeres i en vannoppløsning av et cellulosederivat, f. eks. karboksymetylcellulose, som deretter dryppes i en vannoppløsning av en syre eller et organisk oppløsningsmiddel.
4. Fremgangsmåte ifølge påstand 1—3, karakterisert ved at de stivnede perler vaskes ufullstendig og at badvæsken fjernes fullstendig fra perlene etter tørking.
5. Fremgangsmåte ifølge påstand 1—4, karakterisert ved at man for oppnåelse av klare perler før inndryppingen i badet inn-stiller brytningsindeks for de forskjellige faser i dråpene hovedsakelig på samme verdi, f. eks. ved å tilsette en oljefase i dråpene en hensiktsmessig mengde harpiks.
