Description
Fremgangsmåte ved fremstilling av terapeutisk aktive [ h-(2-inethylen-alkanoyl)-I'enoxyJ -eddiksyrer.
Foreliggende oppfinnelse angår en ny fremgangsmåte ved fremstilling av [ ^--(2-methylenalkanoyl)-fenoxyJ -eddiksyrer som er virksomme diuretiskc og/eller saluretiske midler og som således er nyttige ved behandling av lidelser som skyldes en for hoy kon-sentrasjon av væske i legemet, f.eks. ved behandling av vutter-sottiidelser som skyldes hjertesvikt.
Generelt omfatter foreliggende oppfinnelse oxyderJng av
en l.l+-(l-hydroxy-2-methy.l.enalkyl)-fenoxy j-eddi kny re med o-kloranil som er egnet for å overfore 1-hydroxy-gruppen til den tiLsvnrcnde oxygruppe.
Ved foreliggende fremgangsmåte fremstilles terapeutisk
aktive forbindelser av formel 1:

Foreliggende oppfinnelse angår også syreaddjsjonssaltene av I.'+-(2-methylenalkanoyl)-fenoxyJ-eddjksyre fremstilt ved foreliggende fremgangsmåte. Generelt er saltene ikke-giftige farmasbytisk godtag-bare syreaddisjonssalter som bekvemt fremstilles ved omsetning av I<1>+-(2-methylenalkanoyl)-fenoxyj-eddiksyrene med en base, idet egnede baser innbefatter f.eks. alkali- og jordalkalimetallhydroxydene,
-car bonatene, etc, ammoniakk og aminer, såsom mono-, di- og trialkyl-aminer, piperidin, etc. 1 alminnelighet kan den anvendte base va:re et hvilket som helst reagens som er istand til å forene seg med en
monocarboxylsyre, forutsatt at kationen ikke er giftig og kan inntas i det menneskelige legeme uten å gi uheldige farmakologiske bivirkninger og uten å oppvise egenskaper som er uforenlig med den diuretiske virk-somhet av fenoxy-eddiksyrene. Det vil lett irmsees at det er et stort antall baser som kan reagere med fenoxy-eddiksyrene ifolge oppfinnelsen og danne de tilsvarende ikke-giftige syreaddisjonssalter, og i den utstrekning at disse salter ikke demper den briskede diuretiske virkning som tilskrives syrene, betraktes de som de funksjonelle ekvi-valenter av sistnevnte og kommer innenfor rammen av oppfinnelsen.
O-kloranil er særlig egnet som oxydasjonsmiddel for å overfore 1-hydroxy-carbinolgruppen tii det tilsvarende oxoderivat. Anvend-elsen av temperaturer over eller under den omgivende, er ikke nbdvend-ig for å få et vellykket resultat, og oxydasjonen forlbper mest for-delaktig ved værelsetemperatur. bit hvilket som helst inert fortyn-ningsmiddel som ikke vil bevirke en omordning av hydroxygruppen i carbinolreaktanten eller vil bli oxydert under reaksjons betingelsene, kan anvendes som opplbsningsmiddel ved foreliggende fremgangsmåte. ning av et vinylmagnesiumhalogenid med en (^-forrnylfenoxy)-eddiksyre i henhold til den nedenfor angitte ligning:
hvor R, R"<*>", X og m er som ovenfor angitt. Reaksjonen kan utfores i nærvær av et inert opplosningsmiddel, f.eks. tetrahydrofuran, og ved temperaturer i området fra ca. 0 - 10°C. (>+-f orrnylf enoxy)-eddiksyrene
(B) som anvendes som utgangsmaterialer ved den ovenfor beskrevne syn-tese, fremstilles bekvemt med omsetning av fenol eller en passende
kjernesubstituert fenol, med hydrogencyanid og hydrogenklorid (gass)
i nærvær av vannfritt aluminiumklorid. Reaksjonen utfores fortrinnsvis i et inert opplosningsmiddel, som f.eks. i en benzenopplosning. Den således dannede *+-forrnylfenol omsettes så med 2-halogeneddiksyre
i nærvær av en base, og fortrinnsvis tilsettes basen og 2-halogen-eddiksyrereaktanten gradvis til h- Torrnylfenolen eller den kjernesubstituert ^-forrnylfenol, slik at reaksjonsblandingen blir hverken sur eller basisk, men holdes praktisk talt noytral. Passende baser innbefatter, f.eks. en fortynnet opplosning av natriumhydroxyd, etc. Vinyl-magnesiumhalogenidreaktantene (A) fremstilles ved omsetning av magnes-iummetall med vinylhalogenid eller et passende substituert vinylhalogenid.
Eksempel 1
Fremstilling av utgangsmateriale
6 g (0,25 mol) magnesium ble anbragt i en kolbe forsynt med
en kjoler avkjolt med tbrris-aceton, en dråpetrakt, et nitrogeninn-lbpsror og rorer. Etter spyling av apparaturen med nitrogen og under
opprettholdelse av en sakte strom av nitrogen ble reaksjonen utfort som folger: 19,0 g (0,25 mol) 2-klorpropen ble opplost i 100 ml tetrahydrofuran og 5 ml av denne opplosning sammen med 0,5 ml ethylbromid ble tilsatt til magnesiumet i kolben. Etter at reaksjonen har begynt, tilsettes resten av 2-klorpropenopplosningen med en slik hastighet at temperaturen holdes ved ca. 50°C. Etter at tilsetningen var avsluttet, ble blandingen oppvarmet ved 50°C i 3/*+ time og avkjolt til 0 - ]0°C for å få en opplosning av 2-propenyImagnesiumklorid.
Ved å anvende de fblgende alkanylhalogenider istedenfor det angitte 2-klorpropen i ovenstående prosess, fikk man de tilsvarende alkenyl-magnesiumhalogenider av nevnte alkaner: 2-klor-l-buten, 3-klor-2-penten, 2-klor-<1>+,It-,<i>+-trifluor-l-buten, 2-klor-2-penten, 1-fenyl-1- bromethylen, 1-cyclohexyl-l-klor-ethylen, 3-fenyl-2-klor-]-propen og 2- methyl-3-klor-2-hexen.
Til den erholdte 2-propenylmagnesiumklorid-opplbsning ble
18,0 g (0,1 mol) (^--f orrnylf enoxy)-eddiksyre i 50 ml tetrahydrofuran tilsatt under omrbring med en slik hastighet at temperaturen ble holdt ved 0 - 10°C. Blandingen ble omrbrt i ytterligere en time og derpå helt i 200 ml mettet ammoniumkloridopplbsning under omrbring og avkjbling. Tetrahydrofuranskiktet ble fjernet, tbrret over natriumsulfat og tetrahydrofuranet fordampet. Residuet ble destillert under nedsatt trykk og produktet identifisert som ( (l-hydroxy-2-methylenpropyl)-fenoxy]-eddiksyre.
[ h-(l-hydroxy-2-methylenpropyl)-fenoxyJ-eddiksyren kan fremstilles ved en alternativ metode ved å anvende 2-propenyllithlum istedenfor 2-propeny]magnesiurnklorid anvendt ovenfor.
Til en opplosning av 0,25 mol 2-propenyllithium i ether i et apparat som anvendt ved fremstillingen under anvendelse av 2-propenylmagnesiumklorid, ble 0,1 mol (^--f orrnylf enoxy)-eddiksyre i ether tilsatt på den der beskrevne måte ved 0 - 5°C. Etter omrbring i ytterligere en time ble blandingen helt i 200 ml mettet ammoniumkloridopplbsning under omrbring og avkjbling. Tetrahydrofuranskiktet ble fjernet, tbrret over natriumsulfat og tetrahydrofuranet fordampet. Residuet ble destillert under nedsatt trykk og identifisert som [ h-(1-hydroxy-
2-methylenpropyl)-fenoxyJ-eddiksyre.
Fremstilling av sluttprodukt.
2,<>>H> g (0,01 mol) o-kloranil ble opplost i 182 ml kloroform og 2,2 g (0,01 mol) I'f-(1-hydroxy-P-mothylenpropyl)-fenoxyJ-eddiksyre ble tilsatt. Blandingen ble holdt i morke i lb timer. Keaksjonsbland-ingen ble så fort gjennorn en kort kolonne av aluminiumoxyd og eluert med kloroform. Ilydrokinon-biproduk tet forble i kolonnen og produktet ble isolert ved inndampnlng av opplesningen. Det faste residuum ble krystallisert fra benzen, hvorved man fikk |'i-(2-methylenpropionyl)-f enoxy J-eddiksyre med smeltepunkt 12<!>r,5 - 126,5°C.
Ved å anvende den passende k j ernesubsti tuerte (lf-f orrnylf en-oxy) -eddiksyre- og alkenyJmagnesiumhalogenid-reaktant istedenfor (<>>i-forrnylferioxy) -eddiksyren og propenyimagnesiumhalogenidet angitt ovenfor og ved praktisk talt samme fremgangsmåte som beskrevet der, ble de tilsvarende kjernesubsti tuerte | W-(l-hydroxy-2-methylenpropyl)-fenoxyj-eddiksyreforbindelser fremstilt og disse hydroxyderivater ble så omdannet til de tilsvarende | 'i—(2-rnethy lenalkanoyl) -fenoxy j-eddiksyrer med i'rerngangsmåtenDeskrevet ovenJ'or.
