Abstract

Se presenta un metodo para preparar materiales biologicos para criopreservacion. El metodo reduce la cantidad de calor liberado por un crioprotector durante una fase de calor latente cuando se congela el protector, se descongela el protector y se tratan los materiales con actividad biologica con las sustancias protectoras descongeladas. En primer lugar, se congela la sustancia protectora para inducir un cambio de fases irreversible junto con una liberacion de energia irreversible. Despues de que ha ocurrido este cambio de fases, se descongela la sustancia protectora y se utiliza para tratar las celulas viables u otro material con actividad biologica que vaya a ser sometido a congelacion. La sustancia protectora descongelada dentro de las celulas con actividad biologica tiene una reaccion endotermica reducida tras el congelamiento ulterior. En una modalidad, el congelamiento del material biologico consiste en sumergir el material biologico en un liquido refrigerante y hacer circular el liquido refrigerante sobre el material biologico a una velocidad predeterminada, considerablemente constante y una temperatura tal que se vitrifique el material biologico, reduciendo asi al minimo la formacion de fracturas por esfuerzos en las membranas celulares. La sustancia protectora puede ser propilen glicol, glicerol, DMSO u otras sustancias protectoras convenientes.
