Abstract
Se describen las matrices que soportan la adhesion y el desarrollo celular, que distribuyen los peptidos o proteinas del factor de crecimiento de baja afinidad de enlace a la heparina, de una manera controlada. Estas matrices comprenden la heparina o polimeros similares a la .heparina covalente o no covalentemente enlazados, los cuales sirven para secuestrar los peptidos de la proteina del factor de crecimiento de baja afinidad a la heparina, dentro de la matriz. La liberacion controlada de algun factor de crecimiento de baja afinidad a la heparina o peptidos del mismo, ocurre mediante degradacion de algun componente de matriz o la disociacion de los peptidos de la proteina del factor de crecimiento de baja afinidad a la heparina, provenientes de la heparina enlazada. Esto difiere de muchos dispositivos de distribucion controlada en que la liberacion no es controlada solamente por difusion, y la velocidad de liberacion puede por lo tanto ser regulada mediante la alteracion de la velocidad de degradacion del componente de matriz o la cantidad de heparina enlazada dentro de la matriz. La liberacion controlada de tales proteinas del factor de crecimiento de baja afinidad de enlace a la heparina tales como NGF??, NT-3, y BDNF, es demostrada. La invencion tambien identifica los dominios basicos que pueden ser utilizados para identificar otros peptidos de la proteina del factor de crecimiento de baja afinidad de enlace a la heparina, titiles en la distribucion como parte de las matrices descritas en la presente.
Claims (23)
1. Un sistema de distribución para un factor de crecimiento proteico o un fragmento peptídico del mismo, que comprende: un substrato,- un péptido que está covalentemente enlazado al substrato, y el cual tiene un dominio de enlace que se enlaza a la heparina o a un polisacárido similar a la heparina o a un polímero similar a la heparina, con alta afinidad; la heparina o un polisacárido similar a la heparina o un polímero similar a la heparina enlazado a ese péptido; un factor de crecimiento proteico o un fragmento peptídico del mismo, que tiene un dominio que se enlaza a la heparina o al polisacárido similar a la heparina o al polímero similar a la heparina, con baja afinidad, en donde la baja afinidad es definida como la provisión de la elución a partir de una columna de afinidad de heparina a una concentración de NaCl menor de aproximadamente 140 mM.
2. El sistema de distribución de conformidad con la reivindicación 1, en donde el factor de crecimiento proteico o el fragmento peptídico del mismo es definido como que eluye a partir de una columna de afinidad de heparina a una concentración de NaCl aproximadamente 25 mM hasta aproximadamente 140 mM.
3. El sistema de distribución de conformidad con la reivindicación 1 ó 2, en donde el dominio del factor de crecimiento proteico o el fragmento peptídico del mismo es además definido como poseedor de una longitud de aproximadamente 8 a 30 residuos de aminoácidos, que comprende al menos dos residuos de aminoácidos básicos, una proporción de aminoácidos básicos a ácidos de al menos 2, y una proporción de residuos de aminoácidos hidrofóbicos a residuos de aminoácidos básicos de al menos 0.67.
4. El sistema de distribución de conformidad con la reivindicación 3, en donde los residuos de aminoácidos básicos son K o
5. El sistema de distribución de conformi con la reivindicación 3 ó 4 , en donde los residuos aminoácidos ácidos son además definidos como D o E.
6. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 3 a 5, en donde los residuos de aminoácidos hidrofóbicos son además definidos como A, V, F, P, M, I o L o C, cuando C esté involucrado en un enlace disulfuro.
7. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 6, en donde el factor de crecimiento proteico o el fragmento peptídico del mismo es neuroturina, persefina, IGF-1A, IGF-?ß, EGF, GF , NT-2 , BDNF, NT-4, TGF-P2 , TGF-P3, o TGF- 4.
8. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 7, en donde el substrato comprende al menos uno de: fibrina, colágeno, ácido hialurónico, derivados de ácido hialurónico e hidrogeles poliméricos sintéticos.
9. El sistema de distribución de conformidad con la reivindicación 8, en donde el substrato es fibrina .
10. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 9, en donde la heparina o el polisacárido similar a la heparina o el polímero similar a la heparina está no covalentemente enlazado al péptido que se enlaza a la heparina o a un polisacárido similar a la heparina, o al polímero similar a la heparina, con alta afinidad .
11. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 10, en donde el péptido que se enlaza a la heparina o a un polisacárido similar a la heparina o a un polímero similar a la heparina con alta afinidad, es además definido como que eluye a partir de una columna de afinidad a la heparina a no menos de 140 mM de NaCl .

El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 11, en donde el péptido que se enlaza a la heparina o a un polisacárido similar a la heparina o a un polímero similar a la heparina con alta afinidad, es además definido como que comprende las SEQ ID No. 1, SEQ ID No. 2, SEQ ID No. 3, SEQ ID No. 4, o SEQ ID No. 5.
13. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 12 , en donde la heparina o el polisacárido similar a la heparina o el polímero similar a la heparina tienen un peso molecular entre aproximadamente 3000 y 10,000,000 Daltones .
14. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 13, en donde el polisacárido similar a la heparina tiene un peso molecular entre aproximadamente 3000 y 10,000,000 Daltones, y tiene al menos una carga negativa por 2 anillos sacáridos y no más de una carga positiva por 10 anillos sacáridos.
15. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 14, en donde el polímero similar a la heparina es sulfato de dextrano, sulfato de condroitina, sulfato de heparan, fucano, alginato o un derivado de los mismos.
16. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 15, en donde la proporción molar de la heparina o del polisacárido similar a la heparina o del polímero similar a la heparina, al factor de crecimiento proteico o al fragmento peptídico del mismo, es al menos 1.
17. El sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 16, en donde la proporción molar del péptido covalentemente enlazado que se enlaza a la heparina o a un polisacárido similar a la heparina, o a un polímero similar a la heparina, con alta afinidad a la heparina o al polisacárido similar a la heparina o al polímero similar a la heparina, es al menos 1.
18. Un artículo de fabricación que comprende un sistema de distribución para un factor de crecimiento proteico o un fragmento peptídico del mismo, de conformidad con cualquiera de las reivindicaciones 1 a 16, en donde el substrato es capaz de soportar la adhesión celular.
19. El artículo de fabricación de conformidad con la reivindicación 18, el cual es un injerto vascular .
20. El artículo de fabricación de conformidad con la reivindicación 18, el cual es un artículo para el tratamiento de heridas dérmicas.
21. El artículo de fabricación de conformidad con la reivindicación 18, el cual es una composición implantable, esterilizada.
22. Un método para proporcionar liberación controlada de un factor de crecimiento proteico o un fragmento peptídico del mismo, que comprende: la preparación de un sistema de distribución de conformidad con cualquiera de las reivindicaciones 1 a 16, y la colocación del sistema de distribución sobre una herida.
23. El método de conformidad con la reivindicación 22, en donde el factor de crecimiento proteico o el factor peptídico del mismo es liberado mediante disociación de factores proteicos de crecimiento o del factor peptídico del mismo, a partir de la heparina o del polisacárido similar a la heparina o del polímero similar a la heparina RESUMEN DE LA INVENCIÓN Se describen las matrices que soportan la adhesión y el desarrollo celular, que distribuyen los péptidos o proteínas del factor de crecimiento de baja afinidad de enlace a la heparina, de una manera controlada. Estas matrices comprenden la heparina o polímeros similares a la heparina covalente o no covalentemente enlazados, los ' cuales sirven para secuestrar los péptidos de la proteína del factor de crecimiento de baja afinidad a la heparina, dentro de la matriz. La liberación controlada de algún factor de crecimiento de baja afinidad a la heparina o péptidos del mismo, ocurre mediante degradación de algún componente de matriz o la disociación de los péptidos de la proteína del factor de crecimiento de baja afinidad a la heparina, provenientes de la heparina enlazada. Esto difiere de muchos dispositivos de distribución controlada en que la liberación no es controlada solamente por difusión, y la velocidad de liberación puede por lo tanto ser regulada mediante la alteración de la velocidad de degradación del componente de matriz o la cantidad de heparina enlazada dentro de la matriz. La liberación controlada de tales proteínas del factor de crecimiento de baja afinidad de enlace a la heparina tales como NGFP , NT-3, y BDNF, es demostrada. La invención también identifica los dominios básicos que pueden ser utilizados para identificar otros péptidos de la proteína del factor de crecimiento de baja afinidad de enlace a la heparina, útiles en la distribución como parte de las matrices descritas en la presente.
