Abstract
Esta invencion proporciona polinucleotidos aislados que codifican ADN polimerasas de tres miembros de una sub-familia de los virus del herpes Gama. Dos fueron obtenidos de monos macacos afectados con fibromatosis retroperitoneal, el otro de pacientes humanos de SIDA afectados con sarcoma de kaposi. Un fragmento de 454 pares de bases codificando una region cercana al sitio activo de la ADN polimerasa es de 69-83% idéntico en los tres virus, pero solamente 54-68% idéntico a otras secuencias de herpes gama conocidas y menos de 55% idéntico a secuencias de herpes alfa y beta. También se proporcionan polinucleotidos que codifican ADN polimerasa de virus relacionados en la sub-familia RFHV/KSHV. Los nucleotidos preparados de acuerdo con los datos de secuencia pueden ser usados como reactivos para detectar y caracterizar secuencias relacionadas. Tales reactivos pueden ser usados para detectar miembros de la sub-familia RFHV/KSHV, incluyendo pero sin limitarse a RFHV, RFHV2 y KSHV. Polipéptidos y fragmentos de péptidos correspondientes pueden ser obtenidos expresando el polinucleotido o mediante síntesis química. Pueden usarse para detectar anticuerpo específico presente en el suero de sujetos infectados. También pueden usarse para diseñar o analizar compuestos farmacéuticos que limitan la replicacion viral inhibiendo la actividad de la ADN polimerasa.
