Abstract

La presente invención se refiere a métodos para la biosíntesis de ergotioneína; de manera más particular, la presente descripción se relaciona con métodos para biosíntesis microbiana de ergotioneína; la presente descripción se relaciona de manera general con célula hospederas manipuladas y métodos para producir ergotioneína; más particularmente, la presente descripción se relaciona con una célula hospedera manipulada y métodos para biosíntesis microbiana de ergotioneína utilizando la célula hospedera manipulada. Una célula hospedera manipulada por la producción de ergotioneína, la célula hospedera caracterizada porque comprende una vía metabólica para la producción de ergotioneína, en donde la célula hospedera es Escherichia coli, Sacharomyces cerevisiae, o Pichia pastoris, y en donde la célula hospedera comprende: una oxidorreductasa (EgtB) heteróloga que comprende una secuencia de aminoácidos que tiene por lo menos 99% de identidad de secuencia con la SEQ ID NO. 2, una antidohidrolasa (EgtC) heteróloga que comprende una secuencia aminoácidos que tiene por lo menos 99% de identidad de secuencia con la SEQ ID NO. 4, una histidina metiltransferasa (EgtD) heteróloga que comprende una secuencia de aminoácidos que tiene por lo menos 99% de identidad de secuencia con la SEQ ID NO: 6, y una proteína que une 5-fosfato de piridoxal (EgtE) heteróloga que comprende una secuencia de aminoácidos que tiene por lo menos99% de identidad de secuencia con la SEQ ID NO 8, en donde EgtB, EgtD y EgtE y hasta dos marcadores seleccionables son las únicas enzimas heterólogas en la célula hospedera.
