Abstract

Se proporciona un método, que incluye la hibridación de cebadores de prueba a cebadores inmovilizados, en donde los cebadores inmovilizados incluyen una secuencia de nucleótidos predeterminada y se unen a un sustrato a través de sus extremos 5-prima, los cebadores de prueba individuales son complementarios a una porción de cada uno de al menos algunos de los cebadores inmovilizados, y no más de una molécula de cebador de prueba se híbrida con una molécula de cebador inmovilizado, extendiendo, mediante solo un nucleótido, al menos algunos de los cebadores de prueba con una polimerasa de acuerdo con plantillas, en donde dichas plantillas comprenden cebadores inmovilizados hibridados con dichos cebadores de prueba y los nucleótidos incorporados en los cebadores de prueba extendidos comprenden una etiqueta fluorescente, y detectar una cantidad de cebadores de prueba fluorescentes.
