Abstract
La invención proporciona un método para el análisis de una muestra biológica para determinar múltiples tipos de información de la misma en un flujo de trabajo combinado, simple, donde toda la información se obtiene en un análisis basado en secuenciación. La información incluye la presencia y concentración de proteínas plasmáticas específicas en una muestra de sangre; el número, ubicación, y tipos de modificaciones de histona asociadas con el ADN libre de célula obtenido de la misma muestra; la secuencia de ARNcf y ADNcf en la muestra de ADN libre de célula; e información epigenética que se refiere al ADN libre de célula, tal como perfile de hidroximetilación y metilación, es decir, la distribución de los residuos de 5-hidroximetilcitosina (5hmC) y 5-metilcitosina (5mC), respectivamente. La invención se refiere adicionalmente a un método basado en secuenciación clásica para analizar una muestra biológica para determinar una o más características de secuencia no clásicas de la muestra. También se proporcionan composiciones, kits, y métodos relacionados, que incluyen una modalidad en la cual los adaptadores de secuenciación truncados se utilizan en conjunto con los cebadores de PCR marcados con códigos de barras.
