Abstract

Se describen métodos para obtener no destructivamente el genotipo de una célula vegetal de una muestra vegetal. La célula vegetal se aísla de una muestra de células vegetales que utiliza un dispositivo microfluídico integrado. El dispositivo microfluídico integrado incluye una trampa celular individual situada corriente abajo de un canal microfluídico y, el dispositivo microfluídico se configura para atrapar la célula vegetal en la trampa celular individual. El ARNm se extrae de la célula vegetal al poner en contacto la célula vegetal con una sonda de microscopio de fuerza atómica (AFM) y al extraer ARNm de los sitios de interés al extremo de la sonda utilizando una fuerza DEP de dielectroforesis bajo el campo de corriente alterna (AC) aplicado a la sonda. El genotipo de la célula vegetal se determina de ADNc obtenido del ARNm extraído. Alternativamente, el ARNm se analiza para determinar los patrones de expresión génica de la célula vegetal.
