Abstract

La presente invención se refiere a métodos para producir entidades químicas codificadas. En particular, los oligonucleótidos y métodos pueden incluir entidades químicas codificadas teniendo eslabones tipo silvestre formados a través de técnicas de ligación química. Una estrategia que puede utilizarse que aprovecha simultáneamente la ligación química como medio para codificar la historia química, mientras también conserva la capacidad de las polimerasas a recuperar directamente secuencia de marbete e información de asociación, es la de llevar a cabo ligación química en una manera que genere eslabones fosfodiéster tipo silvestre. Tales métodos utilizan generalmente agentes de condensación tales como bromuro de cianógeno o similares junto con 5´-fosfato y 3´-hidroxil oligonucleótidos en una contexto de doble hélice o de plantilla. De manera similar, el bromuro de cianógeno también ha mostrado ligar químicamente pares de oligonucleótidos de sustrato que son 5´-hidroxil y 3´-fosfato. Sin embargo, estos métodos sufren de una pobre eficiencia que los hace no adecuados para uso en un proceso iterativo tal como marbetar bibliotecas codificadas en ADN.
