Abstract

The invention relates to biotechnology, in particular to a process for the production of cabbage somaclones.The process includes the mechanical infection of plants in the phase of two cotyledonous leaves with the cauliflower mosaic virus, selection of mesophilic tissue explants from the chlorotic areas adjacent sectors, their sterilization, inoculation of explants in the Murashige-Skoog nutrient medium, containing 7 g/l of agar, 0.01 mg/l of kinetin and additionally 0.5 mg/l of 6-benzylaminopurine, 0.5 mg/l of naphthaleneacetic acid, followed by their incubation in the dark at the temperature of 25°C to initiate the formation of morphogenic callus, and then with a photoperiod of 16 hours at the light of 2000 lx and the temperature of 27°C, transfer of morphogenic calluses to fresh nutrient media to obtain somaclones and their subsequent establishment in the Murashige-Skoog nutrient medium, containing additionally 0.5 mg/l of naphthaleneacetic acid, followed by adaptation to ex-vitro conditions and planting in soil of somaclones.The result of the invention is to increase the somaclonal variation of cabbage and the possibility of selecting somaclones with quantitative traits, non-specific for the original variety.
