Abstract

Invenţia se referă la medicină, în special la medicina preventivă şi poate fi utilizată la prognozarea riscului dezvoltării cancerului glandei tiroide.Conform invenţiei, procedeul de prognozare a riscului de dezvoltare a cancerului glandei tiroide constă în aceea că se prelevează un fragment de ţesut din tumoarea glandei tiroide, se extrage din acesta ADN-ul genomic prin digerare enzimatică cu proteinaza K, se modifică ADN-ul prin conversia citozinei în uracil prin metoda Qiagen EpiTect Bisulfite Kit, apoi ADN-ul modificat se amplifică cu ajutorul unor praimeri corespunzători ADN-ului genei p15 metilate şi celei nemetilate prin metoda Methylation Specific Polimerase Chain Reaction, produşii se introduc într-un gel de electroforeză din agaroză dizolvată în tris-borat-etilendiamintetraacetat şi se colorează cu bromură de etidiu, gelul se iradiază cu radiaţie ultravioletă cu lungimea de undă de 312 nm, şi în cazul, în care se vizualizează numai banda AND-ului metilat, se prognozează riscul de dezvoltare a cancerului glandei tiroide, iar în cazul în care se vizualizează banda AND-ului metilat şi banda AND-ului nemetilat - lipsa riscului de dezvoltare a cancerului glandei tiroide.
