Abstract

Inventia se refera la medicina, in special la medicina preventiva si poate fi utilizata la prognozarea riscului dezvoltarii cancerului glandei tiroide. Conform inventiei, procedeul de prognozare a riscului de dezvoltare a cancerului glandei tiroide consta in aceea ca se preleveaza un fragment de tesut din tumoarea glandei tiroide, se extrage din acesta ADN-ul genomic prin digerare enzimatica cu proteinaza K, se modifica ADN-ul prin conversia citozinei in uracil prin metoda Qiagen EpiTect Bisulfite Kit, apoi ADN-ul modificat se amplifica cu ajutorul unor praimeri corespunzatori ADN-ului genei p15 metilate si celei nemetilate prin metoda Methylation Specific Polimerase Chain Reaction, produsii se introduc intr-un gel de electroforeza din agaroza dizolvata in tris-borat-etilendiamintetraacetat si se coloreaza cu bromura de etidiu, gelul se iradiaza cu radiatie ultravioleta cu lungimea de unda de 312 nm, si in cazul, in care se vizualizeaza numai banda AND-ului metilat, se prognozeaza riscul de dezvoltare a cancerului glandei tiroide, iar in cazul in care se vizualizeaza banda AND-ului metilat si banda AND-ului nemetilat - lipsa riscului de dezvoltare a cancerului glandei tiroide.The invention relates to medicine, in particular to preventive medicine and can be used to predict the risk of developing thyroid gland cancer. According to the invention, the process for predicting the risk of developing thyroid gland cancer consists in the fact that a tissue fragment is taken from the tumor of the thyroid gland, the genomic DNA is extracted from it by enzymatic digestion with proteinase K, the DNA is modified by the conversion of cytosine to uracil by the Qiagen EpiTect Bisulfite Kit method, then the modified DNA is amplified by DNA-matching progenitors of the p15 methylated and unmethylated DNA by the Methylation Specific Polymerase Chain Reaction method, the products are introduced into an electrophoresis gel of agarose dissolved in tris-borate-ethylenediaminetetraacetate and stained with ethidium bromide, the gel is irradiated with ultraviolet radiation with a wavelength of 312 nm, and if only the methylated AND band is visualized, the risk of development is predicted of thyroid gland cancer, and if the methylated AND and non-methylated AND band are visualized - lack of risk thyroid cancer development.
