Description
DESCRIPTION Novel Biopesticide Method for Efficiently Controlling Asparagus Stem Blight
TECHNICAL FIELD The invention relates to a novel biopesticide method for efficiently controlling asparagus stem blight, in particular to a biopesticide for efficiently controlling asparagus stem blight prepared by comprehensively utilizing chitosan oligosaccharide, Chinese herbal medicine extract and carbendazim, belonging to the technical field of agricultural biological control.
BACKGROUND Asparagus officinalis, also known as Asparagus, 1s a perennial herb of Asparagus (genus) in Asparagus, Asparagaceae. Its young plants can be used for vegetables, and it has the reputation of "the king of vegetables" in the international market. Asparagus has high nutritional value, can moisten lung, relieve cough, eliminate phlegm and inhibit tumor growth, which is very popular among people. In recent years, with the expansion of Asparagus cultivation area, the occurrence of diseases is increasing year by year, especially the occurrence and harm of stem blight, which seriously affects the yield and quality of Asparagus. The pathogen of asparagus stem blight is Phomopsis asparagi, which is a worldwide devastating disease. The disease spreads rapidly in rainy and windy conditions, and it can also be spread by rain splashing. Air infection is the main cause of large-scale disease. The direction of field spread and rapid disease are often affected by wind. In addition, low-lying, sticky soil, excessive nitrogen fertilizer, etc., are easy to aggravate the disease. The disease is serious in Asian Asparagus planting countries such as China, Japan, Thailand, Indonesia, etc., especially in China, which is common in all Asparagus producing provinces in China, and the south is more important than the north. Light results in poor growth and development of Asparagus, which reduces the yield and quality; the seriously ill plants die in advance, and the whole field is destroyed. At present, the control of asparagus stem blight is mainly chemical control. However, due to the extensive use of chemical agents for many years, the pathogen showed strong drug resistance in many areas, which greatly reduced the effect of pesticides. Lin Peili et al. and Kang Yebin tested the toxicity of different pesticides to asparagus stem blight. In the case of Peng Mingsheng and Xiao Feng, the control effects of 60% carbendazim hydrochloride wettable powder, 70% thiophanate-methyl wettable powder, 50% benomyl wettable powder, 50% Captafol wettable powder and 12.5% diniconazole wettable powder were determined. However, these fungicides are all fungicides with a long service life, and asparagus stem blight is easy to be resistant to these fungicides, so it is particularly important to find alternative fungicides to control stem blight.
Patent 2017105568113 discloses a new type of biological pesticide for preventing and treating asparagus stem blight and asparagus brown spot. Its main components are Poly(hexamethylenebiguanide)hydrochloride, zineb and thiophanate methyl. It can be seen that the patented technology is still dominated by chemical control; Patent 2013103015752 discloses a chemical fertilizer for preventing and treating asparagus stem blight and its fertilization method. The key technology lies in adjusting the ratio of nitrogen, phosphorus and potassium of chemical fertilizer to keep the soil acidic and destroy the disease condition of asparagus stem blight, which is a temporary solution. To sum up, in the prior art, the control of asparagus stem blight is mainly based on chemical agents, and the new biological agents are mainly used to prevent the occurrence of asparagus stem blight, but the control effect on the already-occurring asparagus stem blight is relatively small, thus easily causing excessive pesticide residues, affecting food safety and reducing Asparagus production benefits.
SUMMARY In order to make up for the defects of the prior art, the invention provides a novel biological pesticide for efficiently preventing and controlling asparagus stem blight, which can significantly prevent and control asparagus stem blight through the synergistic effect of chitosan oligosaccharide, active extracts of Chinese herbal medicines, carbendazim and other raw materials, and improve the yield and quality of Asparagus, thereby improving the planting benefit of Asparagus; the new bio-pesticide is safe, green and pollution-free, which is beneficial to the sustainable development of Asparagus planting industry.
To achieve the above purpose, the technical scheme adopted by the invention is as follows:
a novel biopesticide for efficiently controlling asparagus stem blight is prepared from the following raw materials in parts by weight: 10-20 parts of chitosan oligosaccharide, 20- 40 parts of active extracts of Chinese herbal medicines, 10-40 parts of carbendazim, 8-15 parts of surfactant and 100-200 parts of water.
Preferably, as mentioned above, a novel biopesticide for effectively preventing and controlling asparagus stem blight 1s prepared from the following raw materials in parts by weight: 12-16 parts of chitosan oligosaccharide, 25-32 parts of active extract of Chinese herbal medicine, 20-30 parts of carbendazim, 8-15 parts of surfactant and 100-200 parts of water.
More preferably, as mentioned above, a novel biological pesticide for effectively preventing and controlling asparagus stem blight is prepared from the following raw materials in parts by weight: 15 parts of chitosan oligosaccharide, 30 parts of active extract of Chinese herbal medicine, 25 parts of carbendazim, 10 parts of surfactant and 150 parts of water.
The molecular weight of chitosan oligosaccharide is 1,000-2,000 Da, among which chitosan oligosaccharide with molecular weight of 1,500 Da 1s the best.
The carbendazim 1s 50% wettable powder.
The active extracts of Chinese herbal medicines are water extracts of Peucedanum praeruptorum dunn, cordate houttuynia. and liquorice, wherein the compatibility ratio of Chinese herbal medicines 1s 25 parts of Peucedanum praeruptorum dunn, 18 parts of cordate houttuynia. and 7 parts of liquorice.
The active extract of Chinese herbal medicine is prepared by the following methods: crushing Chinese herbal medicine raw materials with impurities removed, sieving with a 100-mesh sieve, adding purified water 5 times of Chinese herbal medicine raw materials, adding 5% cellulase and 3% xylanase in the mass ratio of Chinese herbal medicine raw materials, and performing enzymolysis at 50-60°C for 12 h under the condition of ultrasonic assistance; after enzymolysis, filtering, and the filtrate is the active extract of Chinese herbal medicine.
The invention also provides a preparation method of a novel biological pesticide for efficiently preventing and controlling asparagus stem blight, which comprises the following steps:
1) weighing chitosan oligosaccharide, carbendazim, active extracts of Chinese herbal medicines, surfactants and water in a formula amount; 2) putting the chitosan oligosaccharide, carbendazim and surfactant weighed in step 1) into the water of the formula amount, and stirring until they are completely dissolved; 3) putting the active extracts of Chinese herbal medicines in the formula amount into the solution prepared in step 2), and stirring evenly to obtain a novel biological pesticide for effectively preventing and controlling asparagus stem blight.
In the present invention, chitosan oligosaccharide are easy to penetrate through the bacterial cell membrane and easily enter the cytoplasm and nucleus, so that the related enzymes with physiological activities inside the cells are leaked, and at the same time, they interact with negatively charged substances such as DNA to interfere with the replication and transcription of DNA, thus exerting their antimicrobial activity; chitosan oligosaccharide can also induce the disease resistance of plants, and can immunize and kill a variety of fungi, bacteria and viruses.
The active extract of Chinese herbal medicine plays an important role in promoting the invention, and the antibacterial, bactericidal and disease prevention effects of the product of the invention are significantly improved by functional active substances such as saponins in the active ingredients of Chinese herbal medicine; in addition, it is found that the combined use of chitosan oligosaccharide, carbendazim and active extracts of Chinese herbal medicines has a strong sustained- release function and prolongs the action time of the product.
Carbendazim is a broad-spectrum fungicide, which can prevent and control diseases caused by fungi (such as fungi imperfecti and polyspora fungi) in many crops, and can be used for foliar spraying, seed treatment and soil treatment, etc.
The existing research results show that carbendazim has poor control effect on asparagus stem blight, and excessive application can produce drug resistance.
By compounding chitosan oligosaccharide, active extract of Chinese herbal medicine and carbendazim, it is found that carbendazim can quickly inhibit the incidence of asparagus stem blight and reduce the harm of asparagus stem blight to Asparagus production.
The invention further provides an application method of the novel bio-pesticide for efficiently preventing and controlling asparagus stem blight in Asparagus planting, which is characterized in that the novel bio-pesticide for efficiently preventing and controlling asparagus stem blight is diluted into 200-300 times of water for spraying, and the usage amount is 500-1,000 g/hm?.
