Abstract
본 발명은 특히 관절염 상태(arthritic condition) 뿐만아니라 치료 및/ 또는 대체적 외과술(replacement surgery)을 연구하는데에 사용하기 위한 골격 조직을 발생시키기 위한 연골세포 및 연골세포 세포계에 관한다. 잠재적으로 유익한 치료 제제인지 여부를 검토하기 위하여 칼슘을 무기화시키는 관절 연골세포 세포계의 용도에 관한여 기술하고 있다. 뿐만아니라, 이러한 세포계를 제조하는 방법은 또한 상기 세포계의 특징과 함께 기술되어 있다. 매트릭스 조직을 제조하기 위한 비대 생장성 연골세포 세포계의 용도에 관하여도 기술되어 있다. 뿐만아니라, 비대 생장성 세포계를 제조하는 방법은 또한 상기 세포계의 특징과 함께 기술되어 있다.

Claims (30)
- 칼슘을 무기화시키는 것을 특징으로 하는 관절 연골세포(articular chondrocytes).
- 제 1 항에 있어서, 복수개의 연골세포가 펠렛 형태(pellet form)로 배열된 관절 연골세포.
- 제 1 항 또는 제 2 항에 있어서, 상기 연골 세포가 성숙한 개체이거나 또는 성숙한 개체에서 얻어진 관절 연골 세포 및 바람직하게는 관절염을 앓고 있거나 또는 관절염의 소인이 있는 개체에서 얻은 관절 연골세포.
- 제 1 항 또는 제 2 항에 있어서, 상기 연골세포가 태아에서 얻어진 관절 연골세포.
- 제 1 항 또는 제 4 항에 있어서, 상기 연골세포가 세포계(cell-line) 로서 제공된 관절 연골세포.
- 제 5 항에 있어서, 상기 세포계가 인간에서 얻어진 관절 연골세포.
- 상기한 항들중 어느 한항에 있어서, 상기 연골세포가 종양 유전자를 포함하는 관전 연골세포.
- 제 7 항에 있어서, 상기 종양 유전자가 온도 민감성인 관절 연골 세포.
- 제 8 항에 있어서, 상기 종양 유전자가 SV 40 T 항원인 관절 연골 세포.
- a) 불멸화 제제를 사용하여 하나 이상의 성숙한 인간 관절 연골 세포 세포계를 불멸화시키는 단계; b) 인간 관절 연골세포 세포 군집을 제조하기 위하여 상기 불멸화된 세포를 배양시키는 단계를 포함하는, 칼슘을 무기화시키는 관절 연골세포 세포계(articular chondrocytes cell-line) 제조 방법.
- 제 10 항에 있어서, 상기 불멸화 제제가 종양 유전자인 방법.
- 제 11 항에 있어서, 상기 종양 세포가 온도 민감성인 방법.
- 제 12 항에 있어서, 상기 종양 유전자가 SV 40 T 항원인 방법.
- 제 12 항 또는 제 13 항에 있어서, 종양 유전자의 불허용 온도에서 상기 불멸화된 세포를 배양시키는 단계를 추가로 포함하는 방법.
- 관절염을 치료하기 위하여 고안된 치료 제제를 동정하기 위하여 칼슘을 무기화시키는 하나 이상의 관절 연골세포의 용도.
- a) 연골 세포, 또는 칼슘을 무기화시키는 관절 연골세포를 포함하는, 하나 이상의 연골세포 세포계를 하나 이상의 테스트 제제에 노출시키는 단계; 및 b) 상기 테스트 제제에 대한 반응성을 관찰하는 단계를 포함하는, 관절염을 치료하기 위하여 고안된 치료 제제 개발 방법.
- 제 16 항에 있어서, 상기 관찰 단계가 상기 연골세포 또는 연골 세포 세포계와 관련된 칼슘 무기화 정도를 측정하는 것을 포함하는 방법.
- 인간 비대 생장성 연골세포 세포계.
- 제 18 있어서, 상기 세포계가 태아를 공급원으로 하는 세포계(cell-line).
- 제 18 항 또는 제 19 항에 있어서, 상기 세포계가 종양 유전자를 포함하는 세포계.
- 제 20 항에 있어서, 상기 종양 세포가 온도 민감성인 세표계.
- 제 20 항 또는 제 21 항에 있어서, 상기 종양 유전자가 SV 40 T 항원인 세표계.
- a) 불멸화 제제(immortalizing agent)를 사용하여 하나 이상의 인간 태아 연골세포를 불멸화(immortalizing) 시키는 단계; b) 인간 태아-유래성 연골세포 군집을 생성시키기 위하여 상기 불멸화된 세포를 배양시키는 단계를 포함하는, 인간 비대 생장성 연골세포 세포계를 제조하는 방법.
- 제 23 항에 있어서, 상기 불멸화 제제가 종양 유전자인 방법.
- 제 24 항에 있어서, 상기 종양 유전자가 온도 민간성인 방법.
- 제 24 항 또는 제 25 항에 있어서, 상기 종양 유전자가 SV 40 T 항원인 방법.
- 제 25 항 또는 제 26 항에 있어서, 종양 유전자의 불허용 온도(non-permissive temperature)에서 상기 불멸화 세포를 배양시키는 단계를 추가로 포함하는 방법.
- 관절 매트릭스(matrix) 조직을 제공하는데에 사용하기 위한 비대 생장성 연골세포 또는 하나 이상의 비대 생장성 연골세포 세포계의 용도.
- a) 비대 생장성 연골세포 세포계를 제조하는 단계: b) 매트릭스(matuix) 생장을 촉진하는 조건하에서 상기 세포계를 배양시키는 단계 ; 및 c) 상기 세포 및 / 또는 상기 매트릭스 조직 및 / 또는 이들의 선택 부분을 수집하는 단계를 포함하는, 손상된 조직을 치료 및 / 또는 대체시키는데에 사용하기 위한 관절 매트릭스 조절 제조 방법.
- 관절염 치료를 위하여 고안된 치료 제제를 동정하는데에 사용하기 위한 비대 생장성 연골세포, 또는 하나 이상의 비대 생장성 연골세포 세포계.
