Abstract
본 발명은 (a) 생체 기관의 온도를 저하시킴과 동시에 예정된 일자 농도로부터 점차적으로 증가하는 농도의 동결방지제 용액으로 기관을 환류시키고; (b) 기관의 적절한 삼투 평형에 이루어지기에 충분한 시간동안 동결 방지제의 농도를 유지하고, (c) 좀 더 높이 예정된 이차 농도로 용액의 동결방지제 농도를 증가시키고 이 이차 농도에서 기관의 적절한 삼투 평형이 이루어지기에 충분한 시간동안 동결 방지제의 농도를 유지함을 특징으로 하는 신장, 간, 심장 등과 같은 사람을 포함한 동물의 생체 기관을 냉동보존하는 방법 및 이를 위한 컴퓨터-제어 장치를 제공한다.

제9도는 -20 내지-30℃ 부근의 온도에서 표준 환류가 외측에 있는 용액을 신장에 환류시키는 장치를 도시한 것이다.
Claims (35)
- (a) 점차적으로 증가하는 농도의 동결방지제 용액으로 기관을 환류시키고; (b)기관의 적절한 삼투 평형에 이루어지기에 충분한 시간동안 동결 방지제의 농도를 유지하고, (c) 좀더 높이 예정된 이차 농도로 용액의 동결방지제 농도를 증가시키고 이 이차 농도에서기관의 적절한 삼투 평형이 이루어지기에 충분한 시간동안 동결 방지제의 농도를 유지하는 단계를 포함함을 특징으로 하는, 생체 기관의 냉동보존 방법.
- 제1항 있어서, 생체기관을 점차적으로 증가하는 농도의동결방지제로 환류시키기전에 기관을 동결방지제 없이 환류시키는 단계를 추가로 수행하는 방법.
- 제2항 있어서,생체기관을 이로프로스트 또는 변형된 성장 인자β1으로 환류시키는 방법.
- 제1항 있어서, 예정된 이차 농도가 유리화를 허용하지 않으며 단계 (c)는 유리화가능한 농도의 동결방지제를 생체 기관에 유입하기전에 그 기관을 냉각시키는 단계를 추가로 수행하는 방법.
- 제1항 내지 4항 중 어느 한 항에 있어서, 생체 기관이 신장 또는 간인 방법.
- 제1항 내지 4항 중 어느 한 항에 있어서, 냉동 보존이 유리화에 의한 것이며 최종 동결방지제 농도가 유리화를 허용하는 방법.
- (a)기관의 손상을 최소화하고 기관의 제환류를 가능케하는 온도로 기관을 가온하고,(b) 기관의 적절한 삼투 평형이 이루어지기에 충분한 시간동안 동결방지제의 비-유리화 농도로 환류시키고, (c) 기관이 이식되기에 적합하도록 하기 위하여 기관의 온도를 높임과 동시에 기관으로부터 거의 모든 동결방지제를 환류유출시켜 기관이 시기되기에 적합하게 하는 단계를 포함함을 특징으로 하는, 냉동 보존후 기관을 이식하기 위해 예비 처리하는 방법.
- 제7항 있어서, 냉동보존이 유리화에 의해 이루어지는 방법.
- 제7항 있어서, 냉동보존이 동결에 의해 이루어지는 방법.
- 제7항 있어서, 단계(b)에서 기관을 비-유리화 농도의 동결방지제와 삼투 완충제와의 배합물로 환류 시키는 방법.
- 제10항 있어서, 삼투 완충제가 저분자량의 삼투 완충제인 방법.
- 제12항 있어서, 저분자량의 삼투 완충제가 말토스, 칼륨 및 나트륨 프락토즈 1,6-디포스페이트, 칼륨 및 나트륨 락토바이오네이트, 칼륨 및 나트륨 글리세로포스페이트, 라피노즈, 말토펜토스, 스타키오스, 스크로즈 및 만니톨로 이루어진 그룹중에서 선택되는 방법.
- 제11항 있어서, 저분자량 삼투 완충제가 수크로즈인 방법.
- 제11항 있어서, 저분자량 삼투 완충제가 만니톨인 방법.
- 제10항 있어서, 삼투 완충제가 고분자량 물질인 방법.
- 제15항 있어서, 고분자량 삼투 완충제가 하이드록실 전분(450,000달톤이하), 폴리비닐피롤리돈, 칼륨라피노즈 운데카아세테이트 및 피콜(1,000 내지 100,000달톤)으로 이루어진 그룹중에서 선택되는 방법.
- 제15항 있어서, 고분자량의 완충제가 하이드록시에틸 전분인 방법.
- 제17항 있어서, 하이드록시에틸 전분의 분자량이 약 450,000인 방법.
- 제10항 있어서, 기관을 비-유리화 농도의 동결방지제와 하나 이상의 저분자량 및 하나 이상의 고분자량 삼투 완충제와의 배합물로 환류시키는 방법.
- 제10항 있어서, 기관을 비-유리화 농도의 동결방지제와 하나의 저분자량 및 하나의 고분자량 삼투 완충제와의 배합물로 환류시키는 방법.
- 제11항 있어서, 저분자량 삼추 완충제의 농도가 점차적으로 0에 가깝게 감소됨과 동시에 동결방지제의 농도도 또한 점차적으로 200 밀리몰이하로 감소되는 방법.
- 제21항 있어서, 저분자량 삼투 완충제의 농도가 150mM 내지 1,000mM의 범위로 감소되는 방법.
- 제21항 또는 22항에 있어서, 동결방지제의 농도가 0으로 감소하는 방법.
- 제21항 있어서, 동결방지제의 농도가 200밀리몰이하로 감소된 후 저분자량 삼투 완충제의 농도가 점차적으로 감소되는 방법.
- 제19항 있어서, 저분자량 삼투 완충제가 만니톨 및 스크로즈로 이루어진 그룹중에서 선택되고 고분자량 삼투 완충제가 하이드록실 전분인 방법.
- 제25항 있어서, 모든 동결방지제가 기관으로부터 제거된 후 하이드록실에틸 전분 농도가 0에 가깝게 점차적으로 감소하고 수크로즈 농도가 점차적으로 0으로 감소함과 동시에 만니톨이 기관내로 환류되는 방법.
- 제15, 16, 18, 19 및 20항 중 어느 한 항에 있어서, 기관이 간인 방법.
- 제10, 19, 20, 25 및 26항 중 어느 한 항에 있어서, 기관이 신장인 방법.
- 제7 또는 8항에 있어서, 기관이 간인 방법.
- 제7항에 있어서,기관이 신장 또는 간일 때 단계(a)에서 온도가 -3℃ 내지 0℃인 방법.
- 제7, 10, 19 및 20항 중 어느 한 항에 있어서, 동결방지제의 비유리화 농도가 20 내지 40중량/용적%인 방법.
- NaH2PO4H2O, 칼륨글루코네이트, 마그네슘 글루코네이트, 글루코스, 글루타티온, 아데노신, HEPES, 아데닌, 리보스 및 염화 칼슘을 함유함을 특징으로 하는 기관의 생명을 유지해주면서 기관을 환류하기 위한 조성물.
- 제2항 있어서, 하이드록실에틸 전분을 추가로 함유하는 조성물.
- 성분의 농도가 3.45g/1의 NaH2PO4H2O, 23.42g/1의 칼륨 글루코네이트, 0.21g/1의 마그네슘 글루코네이트, 0.92g/1의 글루타티온, 1.34g/1 아데노신 하이드로클로라이드, 2.38g/1의HEPES, 0.17g/1의 아데닌, 0.15g/1의 리보스 및 0.0056g/1의 염화 칼슘인 조성물.
- 제33항 있어서, 하이드록실에틸 전분의 농도가 50g/1이고 글루코스의 농도가 0.90g/1인 조성물.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
