Abstract

본 발명은 루신 탈수소효소 (LeuDH) 활성을 갖는 효소, 이를 코딩하는 바실루스 세레우스(Bacillus cereus) 뉴클레오타이드 서열, 이 서열을 함유하는 플라스미드를 사용한 이. 콜라이(E. coli) 미생물의 형질전환 및 효소의 제조방법에 관한 것이다.

루신 탈수소효소 활성을 갖는 효소, 이를 코딩하는 뉴클레오타이드 서열 및 효소의 제조 방법

제1도는 pUC18에 포함된 바실루스 세레우스(Bacillus cereus)로 부터의 루신 탈수소효소(LeuDH)에 대한 유전자를 나타낸다.
제2도는 비. 세레우스로부터의 LeuDH에 대한 유전자의 서열분석을 나타낸다(Sma Ⅰ/XbaⅠ 단편을 포함하는 pleu2).

Claims (9)

  1. 루신 탈수소효소(LeuDH) 활성을 갖는 효소를 코딩하고; (a) 서열 목록에 도시된 뉴클레오타이드 서열, (b) 유전 코드의 축퇴범위에 속하는 (a)의 서열에 상응하는 뉴클레타이드 서열, 또는 (c) 서열(a), 서열(b) 또는 서열(a)와 (b)모두의 서열과 하이브리드화하는 뉴클레오타이드 서열을 포함함을 특징으로 하는 바실루스 세레우스(Bacilus cereus) DNA 서열.
  2. 제1항에 따른 뉴클레타이트 서열이 삽입되어 있는, 에스케리키아 콜라이(Escherichia coli)에서 자가 복제할 수 있는 백터.
  3. 제2항에 있어서, 플라스미드 pUC18의 DNA 서열과 제1항의 DNA 서열을 함유하고, 에스케리키아 콜라이 XL1 blue 중의 pLeu 2로서 기탁번호 DSM 10441로 기탁되어 있으며, 제2도의 제한 지도로 특정지워짐을 특징으로 하는 벡터.
  4. 제2항 또는 제3항에 따른 벡터를 함유하는 LeuDH에 대해 고 생성능을 갖는 에스케리키아 콜라이.
  5. 제4항에 있어서, 에스케리아 콜라이 XL1 blue로서 기탁번호 10441로 기탁되어 있는 에스케리키아 콜라이.
  6. (a) 제1항에 따른 바실루드 세레우스로부터 유도된 뉴클레오타이드 서열 및 (b) 에스케리키아 콜라이내에서 자가 복제되는 플라스미드 DNA를 함유하는 재조합체 DNA를 벡터의 형태로 에스케리키아 콜라이에 삽입시킨 후, 생성된 형질전환체를 적합한 배지에서 발효시키고, Leu-DH를 미생물로부터 분리시킴을 특징으로 하는 , 루신 탈수소효소(LeuDH)의 제조 방법.
  7. 제6항에 있어서, 제4항 또는 제5항에 따른 미생물이 사용되는 방법.
  8. 상응하는 α-케토산으로부터 비-단백질원성 L-아미노산을 제조하기 위한, 제6항 또는 제7항에 따라 수득된 LeuDH 활성을 갖는 효소의 용도.
  9. 사용되는 α-케토산이 2-옥소-4-메틸펜타노산, 2-옥소-3-메틸펜타노산, 2-옥소-3-메틸부타노산, 2-옥소펜타노산, 2-옥소-4-메틸헥사노산, 2-옥소부타노산, 2-옥소-3, 3-디메틸부타노산, 2-옥소헥사노산, 2-옥소-4, 4-디멜틸펜타노산, 2-옥소-3,3-디메틸펜타노산, 2-옥소-4-에틸헥사노산, 2-옥소-5, 5-디멜틸헥사노산, 2-옥소-3-사이클로헥실프로파노산 및 2-옥소-4, 4-디멜틸헥사노산임을 특징으로 하는 제6항 또는 제8항에 따른 효소의 용도.
