Abstract
본 발명은 반코마이신을 함유하는 미생물 발효배양액을 pH2.0 내지 4.0으로 조절하여 강산성 양이온 교환수지, 강염기성 음이온 교환수지 및 다공성(porous)의 강산성 양이온 교환수지를 연속 통과시키고 활성 알루미나(Alumina activated)수지를 통과시킨 다음, C-8 역상 칼럼을 사용하는 분취용 고합 액체 크로마토그래피(Prep-HPLC)에 적용함으로써 반코마이신을 고수율로 신속하게 분리, 회수함을 특징으로하는 반코마이신의 정제방법에 관한 것이다.

제 2도는 C-8역상 칼럼을 사용한 Prep-HPLC 로부터 분취한 반코마이신을 HPLC로 분석한 결과도이다.(USP 분석법).
Claims (5)
- 반코마이신을 함유하는 미생물 발효배양액을 pH 2.0내지 4.0으로 조절하여 강산성 양이온 교환수지, 강염기성 음이온 교환수지 및 다공성의 강산성 양이온 교환수지를 연속 통과시키고 활성 알루미나 수지를 통과시킨 다음, C-8 역상 칼럼을 사용하는 분취용 고압 액체 크로마토그래피에 적용함으로써 반코마이신을 분리, 정제하는 방법.
- 제 1항에 있어서, 분취용 고압 액체 크로마토그래피를 수행함에 있어 이동상으로 10 내지 20% 메탄올을 함유한 10내지 100mM 인산암모늄 완충용액 또는 염화암모늄 완충용액을 사용하는 방법.
- 제 2항에 있어서, 완충용액의 pH가 2.0 내지 3.0 인 방법.
- 제 1항에 있어서, C-8 역상 칼럼이 Kromasil 100-5-C8 인 방법.
- 제 1항에 있어서, 미생물이 아미콜라톱시스 오리엔탈리스(Amycolatopsis orientalis)인 방법.
