본 발명은 탐 - 홀스폴 글리코프로테인 (THG)및 우로모둘린의 정제법, 이방법은 단순한 조작으로 THG 및 우로모둘린이 고 효율로 정제되도록 한다. 및 이들 양자의 식별법을 제공한다.

탐-홀스폴 글리코프로테인 또는 우로모둘린의 정제방법, 정제물 및 양자의 식별법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 인뇨(人尿)를 동결 - 융해 법에 의하여 정제하여 실시예 1 에서 제조된 시료를 겔 여과시킬 때 수득된 용리 패턴이다.

Claims (12)

  1. 인뇨를 저온 방치 또는 동결시킨후 융해시켜 생성된 침전 분획을 수집하는 것을 특징으로 하는 탐 - 홀스폴 글리코프로테인 (Tamm - Horsfall glycoprotein)또는 우로모둘린 (uromodulin)의 정제법.
  2. 제1항에 있어서, 침전 분획이 겔 여과에 의하여 더 정제되는 정제법.
  3. 제1항 또는 2항에 있어서, 인뇨로서 정상뇨가 사용되고 탐 - 홀스폴 글리코프로테인이 정제되는 정제법.
  4. 제1항 또는 2항에 있어서, 인뇨로서 임신부의 뇨가 사용되며 우로모둘린이 정제되는 정제법.
  5. 인간 면역글로블린 클래스인 IgG 의 F(ab´)2 단편, IgA (단량체)와 IgM, 및 IgG 서브클래스인 IgG1, IgG2, IgG3 및 IgG4 모두에 결합할 수 있는 정제 탐-홀스폴 글리코프로테인.
  6. 인간 면역글로블린 클래스인 IgG 와 IgA (단량체), 및 IgG 서브클래스인 IgG1, IgG2, 및 IgG4 에 결합할 수 있으나, 인간 면역글로블린 클래스인 IgG의 F(ab‘)2 단편 과 IgM, 및 IgG 스브클래스인 IgG3와는 쉽게 결합할 수 없는 IgG 및 IgA(단량체) 에 결합할 수 있는 정제 우로모둘린.
  7. 탐 - 홀스폴 글리코프로테인 또는 우로모둘린으로 아직 식별되지 않은 시료에 인간 면역글로블린 클래스인 IgM 또는 IgG 서브클래스인 IgG3 을 작용시키고 이어서 효소 표식 프로테인 C 를 사용하여 이러한 면역글로블린의 작용량을 측정하는 것을 특징으로 하는 탐 - 홀스폴 글리코프로테인 및 우로모둘린의 식별 방법.
  8. 인뇨에 벤조산 나트륨 또는 벤조산 암모늄을 용해시키고 벤조산이 석출되기에 충분하도록 용액을 산성으로 만들어 생성된 침전분획을 수집하는 것을 특징으로 하는 탐 - 홀스폴 글리코프로테인 또는 우로모둘린의 정제법.
  9. 제8항에 있어서, 침전 분획이 겔 여과에 의하여 더 정제되는 정제법.
  10. 제8항 또는 9항에 있어서, 인뇨로서 정상뇨가 사용되고 탐 - 홀스폴 글리코프로테인 또는 우로모둘린의 정제법.
  11. 제8항 또는 9항에 있어서, 인뇨로서 임신부의 뇨가 사용되며 우로모둘린이 정제되는 정제법.
  12. 효소 표식된 IgG의 F(ab‘)2단편을 직접 탐 - 홀스폴 글리코프로테인 또는 우로모둘린으로 아직 식별되지 않은 시료에 작용시키고 이어서 이러한 IgG의 F(ab‘)2 단편의 작용량을 측정하는 것을 특징으로 하는 탐 - 홀스폴 글리코프로테인 및 우로모둘린의 식별 방법.
